3 reasons why in English you should read the passage as you go 2 Types of Questions: Usage questions General passage questions Usage Underline in a sentence Grammatically correct? Punctuated correctly? 1st choice is no change. General passage questions do not have an underline part in a sentence. The first choice is usually something other than No Change. These are questions more about the passage rather than just an underlined part in a sentence. The first several ACT tests I took in high school, I didn’t read the passage. Why? I was in a hurry. 75 questions in 45 mins. When I ask students in my workshop how many skip, over half agree. Many so called experts teach this. I figured out quickly I had to read the passage as I tested; otherwise, I was having to go back and reread. As you read from one underline to the next, remember this isn’t reading comprehension. You just need to think, “In general what is happening?” “What’s the purpose of the passage?” “What’s the author’s main point?” 3 Reasons why you should read as you go: Tough to summarize an essay you didn’t read. Context Some was/were eaten. For Cat’s first birthday her mother bought her 2 cakes. Some were eaten. There were two cakes. Changing tense of the verbs. If you read a sentence by itself, the present tense verb, for example, works great. But if you read the paragraph, every other verb was past tense, and you’ll immediately make a change. So read the passage as you go. That doesn’t mean read the entire essay, then go back and answer the questions. Just read to question one, then answer question one. Then read to question two, then answer question two. If you do this, you’ll finish even faster with a higher score than not reading it. Remember this is not reading comprehension. You don’t need to know all the details. Keep asking yourself as you are reading, “In general, what is happening here?” Special thanks to Ms. Julie Garner for a great review of the podcast. One of the things that Cat is - without a doubt - is fiercely independent. And now that independence may be limited depending on what happens with her PIP assessment. Some years ago, the government decided to replace the Disability Living Allowance (DLA) with a Personal Independence Payment (PIP).  It's introduction was staggered and everyone who wants to continue receiving this benefit has to apply and go for an assessment. For Cat, that time has come - the form has gone in, she's got her assessment date (which should have taken place by the time you listen to this). Cat talks openly about what it means to her and the potential impact if she loses her Motability allowance - which is key in maintaining her independence. She also talks about the upcoming Women in Tech Cyrmu and we hear a clip of her and her husband Dan having a meet & greet with a lovely deer who decided to join them in the car (well just it's head!) while on a day at Longleat We hear her and her husband Dan enjoying an in car visit from Bambi (aka a deer) on a day out at Longleat.

Please click the link to get your free trial and audiobook today! https://www.audibletrial.com/clearlyofftopic Classic Juliet tells a hilarious story about a wedding she attended and her less than graceful arrival and attempt at the worm! Seriously give it a listen its hilarious! Well needless to say our guest today is much more graceful when removing articles of clothing or slightly showing some skin. Cat Lacohie is a multi talented burlesque performer. World renowned. She specializes in Aerial Hoop, Fire eating, Glass walking and of course… MAGIC! Welcome to the show Cat Lacohie aka Vixen Deville! Cat always wanted to perform. She went to college for Performing Arts in London receiving a BA in Theater Arts. Cat was always super into the world of Burlesque but never saw herself as a Burlesque performer. She didn’t start burlesque due to some societal norms. She didn’t think anyone wanted to see her with no clothes on! Something we all go through. Battling a pull inside herself to be on that stage, she finally went to an audition. She knew how to keep the attention of the audience, she wanted this to be about her having the power instead of just dancing around for the audience and being their court jester. She learned about calling out her flaws she was able to take the power back! Cat’s burlesque name is Vixen Deville! Vixen’s show has been developing her act for years now! She has regularly updated and tried new things. The idea was always for her show to be something else other than just a variety show or just a burlesque. She wanted her show to be different. With Vixen, anything is possible! You know like the movie Splash… when he is with the mermaid he can breathe underwater! If I can do it, you can was what she wanted to portray in her performances. Something to take away from this part of the conversation is that you can do anything you set your mind to. DO THE THING! Try and try again, who cares if you fail. Hey friends, if you eat fire…. Please be careful! And make sure to not breathe in! Liquid encouragement is something that we rely on sometimes. We look at alcohol as an excuse. Juliet’s worm was excusable because she was drunk. Why do we do this? Why are we ok with doing something if we have some sort of encouragement. Give yourself the excuse instead of using alcohol as one. For Cat, being on stage is what she wants so encouragement comes from within. Being on stage gives her that permission. She wanted to wear those thigh high boots and those little mini skirts and garters, but didn’t have anywhere to wear it that she felt comfortable. The stage is that permission and comfort for her! Vixen Deville, who is she? Vixen is the same person as Cat. But it didn’t start that way. Vixen was a character she played. Now in her show she starts as Vixen, but she gradually reveals herself as Cat. As her show has morphed her character has morphed. Vixen didn’t have any worries at all when Cat was on stage. What Vixen is today has morphed over the years. Vixen has transformed as Cat has grown. And its so beautiful. Vixen says “Reality is Offensive” Vixen is honest and unfiltered and that is what Vixen has taught Cat over the years. It's common for many of us to dislike some things about our bodies. We often compare our bodies to that of others and think that life would be so much easier if only we looked like "so and so". But what happens when it becomes too much? For Cat, what started out as disliking some parts of her body ballooned into self-hatred and loathing. Her body began to develop earlier than most girls, so she was being sexualized by older men while still in her teens. She was body shamed by dance teachers and constantly reminded how much she "needed to change" from those around her. It's no surprise that over the years, this has all amounted to an intense feeling of not being enough and feeding her mind with vicious self-talk.Cat has finally begun to understand that this way of thinking is hurtful and self-limiting, and some changes have got to be made. There's NO WAY that Cat or anyone can expect to achieve the fullness of happiness and success with this kind of mindset. Along the way, Cat has discovered self-love boosters that have helped her tons during her lowest days and shares her TOP 5 faves with you! Reframing the mind to think positively when we look in the mirror after decades of only hearing the bad isn't easy, but as you'll soon hear, we can absolutely do it and each of us deserves that inner peace.2017 was the year of honest reflection and acknowledgment of the things we've kept buried, but 2018 and beyond will be filled with wholesome changes and unapologetic living! The 89,000-Mr murine cytomegalovirus immediate-early protein activates gene transcription

To study trans-activation of gene expression by murine cytomegalovirus (MCMV) immediate-early (IE) proteins, the IE coding region 1 (ie1), which encodes the 89,000-Mr IE phosphoprotein (pp89), was stably introduced into L cells. A cell line was selected and characterized that efficiently expressed the authentic viral protein. The pp89 that was constitutively expressed in L cells stimulated the expression of transfected recombinant constructs containing the bacterial chloramphenicol acetyltransferase (CAT) gene under the control of viral promoters. The regulatory function of the ie1 product was confirmed by transient expression assays in which MCMV IE genes were cotransfected into L cells together with recombinant constructs of the CAT gene. For CAT activation by the ie1 product, a promoter region was required, but there was no preferential activation of a herpes simplex virus type 1 delayed-early promoter. All plasmid constructs that contained the intact coding sequences for pp89 induced gene expression in trans. The MCMV enhancer region was not essential for the expression of a functional IE gene product, and testing of the cis-regulatory activity of the MCMV enhancer revealed a low activity in L cells. Another region transcribed at IE times of infection, IE coding region 2, was unable to induce CAT expression and also did not augment the functional activity of ie1 after cotransfection.
