Abstract: We have previously documented that in liver cells, the multifunctional protein scaffold p62/SQSTM1 is closely associated with I{kappa}B, an inhibitor of the transcriptional activator NF-{kappa}B. Such an intimate p62-I{kappa}Bassociation we now document leads to a marked 18-fold proteolytic I{kappa}B-stabilization, enabling its nuclear entry and termination of the NF-{kappa}B-activation cycle. In p62-/--cells, such termination is abrogated resulting in the nuclear persistence and prolonged activation of NF-{kappa}B following inflammatory stimuli. Utilizing various approaches both classic (structural deletion, site-directed mutagenesis) as well as novel (in cell chemical crosslinking), coupled with proteomic analyses, we have defined the precise structural hotspots of p62-I{kappa}B association. Accordingly, we have identified such I{kappa}B hotspots to reside around N-terminal (K38, K47 and K67) and C-terminal (K238/C239) residues in its 5th ankyrin repeat domain. These sites interact with two hotspots in p62: One in its PB-1 subdomain around K13, and the other comprised of a positively charged patch (R183/R186/K187/K189) in the intervening region between its ZZ- and TB-subdomains. APEX proximity analyses upon I{kappa}B co-transfection of cells with and without p62 have enabled the characterization of the p62 influence on I{kappa}B-protein-protein interactions. Interestingly, consistent with p62 capacity to proteolytically stabilize I{kappa}B, its presence greatly impaired I{kappa}B interactions with various 20S/26S proteasomal subunits. Furthermore, consistent with p62-interaction with I{kappa}B on an interface opposite to that of its NF-{kappa}B-interacting interface, p62 failed to significantly affect I{kappa}B-NF-{kappa}B interactions. These collective findings together with the known dynamic p62 nucleocytoplasmic shuttling, leads us to speculate that it may be involved in piggy-back nuclear transport of I{kappa}B following its NF-{kappa}B-elicited transcriptional activation and de novo synthesis, required for the termination of the NF-{kappa}B-activation cycle. Consequently, mice carrying a liver specific deletion of p62-residues 68-252 harboring its positively charged patch, reveal age-dependent enhanced liver inflammation. Our findings reveal yet another mode of p62-mediated pathophysiologically relevant regulation of NF-{kappa}B. Copy rights belong to original authors. Nel 1999, mentre si trova negli Stati Uniti per assistere alla finale di Campionato del Mondo della sua disciplina, scopre K38: una società di salvataggio acquatico su moto, utilizzato in situazioni di emergenza. Decide di importare questo sistema in Italia, e ci riesce: dal 2000 il nostro Paese è incluso nella rete di K38, ottenendo il riconoscimento di scuola affiliata. Annigoni, oggi presidente di K38 Italia, riesce a fondere l’amore per le corse sull’acqua al tema della sicurezza e della salvaguardia, condividendo un modello innovativo e pionieristico.

It's a terrific day here at TIBS, we're finally sharing the day we got down to Sin Fronteras (Without Borders) for a day long visit and recorded a show for you! If you have not had a chance to check out the burgeoning craft beer scene in Mexico, we suggest you investigate! When you decide to go south of the border for adventure, make sure you add Sin Fronteras Brewing to your mandatory list. Currently, they are not brewing at Sin Fronteras yet, but they have a great selection of Mexican and American indie beers for you. This is an amazing place to spend a day of drinking and fun! Sin Fronteras is located directly on top of a cliff, at the water. Calling this a beautiful place to drink is an understatement. There is also a very famous surf store (K38) and 2 food places all within the same complex. Sin Fronteras also has two suites you can rent out if you would like to spend the night. It's a very special place, that has no direct comparison, to anywhere in the States. We were lucky enough to take a bus down with 10 of our fans/friends, which made transportation a breeze, and added to how special the day was! A giant shout out to Robert Esparza, the owner of Sin Fronteras, for welcoming us into his establishment and setting up the bus, we truly appreciate how much fun we had!Out first guest on the show is Mariano from Armor Cerveceria, he breaks down their beers and where you can find them in Mexico. In contrast to SD breweries, their best selling beer have nothing to do with the hop profiles. They bring in their water in 5 gallon jugs, talk about brewing the hard way!Later in the show we are joined by Alonzo from Silenas Brewing, for a longer conversation. That has been around for about 6 years and share a brewer with Thr3e Punk Ales, Ivan Maldonaldo. Alonzo discusses how they started, the current beers available, their avoidance of hazy ("Make clears IPA again!"), and their focus on drink-ability. We break down our Top 3 beer drinking towns, of course, Brian's number 1 is no surprise to anyone! Grab an indie beer and hit the play button!
