BUFFALO, NY- May 1, 2024 – A new #researchpaper was #published in Aging (listed by MEDLINE/PubMed as "Aging (Albany NY)" and "Aging-US" by Web of Science) Volume 16, Issue 8, entitled, “A novel deep proteomic approach in human skeletal muscle unveils distinct molecular signatures affected by aging and resistance training.” The skeletal muscle proteome alterations to aging and resistance training have been reported in prior studies. However, conventional proteomics in skeletal muscle typically yields wide protein abundance ranges that mask the detection of lowly expressed proteins. In this new study, researchers Michael D. Roberts, Bradley A. Ruple, Joshua S. Godwin, Mason C. McIntosh, Shao-Yung Chen, Nicholas J. Kontos, Anthony Agyin-Birikorang, Max Michel, Daniel L. Plotkin, Madison L. Mattingly, Brooks Mobley, Tim N. Ziegenfuss, Andrew D. Fruge, and Andreas N. Kavazis from Auburn University, Seer, Inc., and The Center for Applied Health Sciences adopted a novel deep proteomics approach whereby myofibril (MyoF) and non-MyoF fractions were separately subjected to protein corona nanoparticle complex formation prior to digestion and Liquid Chromatography Mass Spectrometry (LC-MS). “Specifically, we investigated MyoF and non-MyoF proteomic profiles of the vastus lateralis muscle of younger (Y, 22±2 years old; n=5) and middle-aged participants (MA, 56±8 years old; n=6). Additionally, MA muscle was analyzed following eight weeks of resistance training (RT, 2d/week).” Across all participants, the number of non-MyoF proteins detected averaged to be 5,645±266 (range: 4,888–5,987) and the number of MyoF proteins detected averaged to be 2,611±326 (range: 1,944–3,101). Differences in the non-MyoF (8.4%) and MyoF (2.5%) proteomes were evident between age cohorts, and most differentially expressed non-MyoF proteins (447/543) were more enriched in MA versus Y. Biological processes in the non-MyoF fraction were predicted to be operative in MA versus Y including increased cellular stress, mRNA splicing, translation elongation, and ubiquitin-mediated proteolysis. RT in MA participants only altered ~0.3% of MyoF and ~1.0% of non-MyoF proteomes. “In summary, aging and RT predominantly affect non-contractile proteins in skeletal muscle. Additionally, marginal proteome adaptations with RT suggest more rigorous training may stimulate more robust effects or that RT, regardless of age, subtly alters basal state skeletal muscle protein abundances.” DOI - https://doi.org/10.18632/aging.205751 Corresponding author - Michael D. Avast, ye space mateys! Con and Dan haul anchor and set sail for the 80s yet again for a slice of semi-spoof space operatics in the form of The Ice Pirates (1984), which stars Robert Urich, Mary Crosby, Michael D. Roberts and features early appearances from Anjelica Houston and Ron Perlman. Set in another galaxy far, far away, where water has become a rare commodity, the film starts out as a heist caper, then takes a weird left-turn into castration and slavery before ending with a battle in a time warp on the way to finding a fabled water-rich planet. It has lasers and robots, but also knee boots, chain mail, cutlasses and an alarming amount of swinging on ropes. Not to mention something called 'space herpes'. The Ice Pirates is a 1984 American comic science fiction film directed by Stewart Raffill, who co-wrote the screenplay with Krull writer Stanford Sherman. The film stars Robert Urich, Mary Crosby and Michael D. Roberts; other notable featured actors are Anjelica Huston, Ron Perlman, Bruce Vilanch, John Carradine and former football player John Matuszak. The film was made at MGM, then under David Begelman, with John Foreman as producer. It was originally called The Water Planet and had a $20 million budget. However, as MGM was in financial difficulty, its bankers put a limit of $8 million on all films. Raffill said he would have to rewrite it and make it more comic, and they agreed. Rafill and Foreman said pirate movies like The Crimson Pirate were the main inspiration and they deliberately did not watch Star Wars. This was Trent's pick. Trent had not watched the movie since it's literal release in 1984 when he was 13 years old. Needless to say, it wasn't the same viewing experience this time around... No, really! Well, the sort of actual zombification possible through drugs. Mama Lorraine has an amount of the antidote, but will she have to use it on MacGyver when he gets caught by Colonel Devraux, the man terrorizing the neighborhood? Patron Jed's unspecified request for a MacGyver episode leads to the worst rated episode on the IMDB list and, of course, racism! Erik offers some praise for Mass Effect: Andromeda even with its story issues and the lack of the Quarian Ark. Justin unveils his "Bane 2020" initiative. The episode's fictional Little Haiti leads to another geography corner. Manimal's Black Partner -- actor Michael D. Roberts -- makes a surprise appearance. Justin and Erik debate whether or not zombies can be part of the Ninja Empire. A real Space Opera with sword fights, explosions, fighting robots, monsters, bar fights and time warps. In this undistinguished parody of the sci fi genre, Robert Urich is Jason who leads a band of pirates in redistributing the wealth of the few to the coffers of the needy. He also joins up with Princess Karina Mary Crosby in searching for her father and a possible source of water in the next galaxy. Meant to be a campy romp through the sci fi genre, the film stops short of achieving a goal that should have been effortless.
