Die humanpathogenen Bakterien Yersinia enterocolitica und Yersinia pseudotuberculosis sind die Verursacher von Darminfekten wie Enteritis und Enterokolitis. Das äußere Membranprotein YadA stellt bei Y. enterocolitica einen essentiellen Pathogenitätsfaktor dar. Seine virulenzassoziierten Eigenschaften sind die Fähigkeit zur Autoagglutination, die Bindung an Moleküle der extrazellulären Matrix, an Epithelzellen und Granulozyten, und die Vermittlung von Serumresistenz. Welchen Teil die verschiedenen Domänen des Proteins zur Struktur und Funktion beitragen, war bisher kaum bekannt. Auch der Versuch, dem Adhäsin durch einen Domänenaustausch völlig neue Bindungs- oder Reaktionseigenschaften zu vermitteln, war bislang nicht unternommen worden. Darum wurden In-Frame-Deletionsmutanten, mit FLAG-Epitopen oder FaktorXa-Proteaseschnittstellen versehene YadA-Mutanten erstellt und FimH-YadA-Hybridproteine konstruiert. Die verschiedenen YadA-Mutanten wurden strukturell auf ihre Außenmembranlokation, Oberflächenexposition und Oligomerisierung untersucht, funktionell auf ihre Adhäsionseigenschaften und Serumresistenz. Dabei konnten folgende Erkenntnisse gewonnen werden: Kopf- und Stieldomäne bilden zusammen mit der interponierten, stark konservierten Neck-Region die translozierte, oberflächenexponierte Passenger-Domäne. Die C-terminale Membranankerregion (as 353-422) ist ausreichend für Insertion in die Außenmembran und Bildung eines trimeren YadA. Die Linker-Region vermittelt die Translokation der Passenger-Domäne durch die Außenmembran. Somit zeigte sich, dass YadA alle Kriterien für einen Autotransporter erfüllt. Der Versuch, ein Hybridadhäsin mit Mannosebindungsfähigkeit durch Austausch der Kopfdomäne mit der Lektin-Domäne von FimH zu erzeugen, schlug fehl. Dies zeigt, wie empfindlich Passenger-Domäne und Membrananker von YadA aufeinander abgestimmt sind. Die funktionelle Untersuchung der Mutanten ergab, dass die hochkonservierte Neck-Domäne zusammen mit der Kopfregion ein Bindungsmodul für Kollagen und Epithelzellen darstellt. Im Serumresistenztest erwiesen sich Kopf-, Neck- und auch Teile der Stiel-Region für ein Überleben entbehrlich. Es zeigte sich, dass keine Domäne für die Serumresistenz von YadA entscheidend ist.
