This episode: A bacteriophage that overcomes the bacterial CRISPR/Cas immune system by interrupting the CRISPR DNA with its own genome! Download Episode (6.8 MB, 10 minutes) Show notes: Microbe of the episode: Wenzhou mammarenavirus Takeaways Bacteria have many ways to resist being exploited by bacteriophage viruses, including the adaptable CRISPR/Cas system that uses a piece of viral nucleic acid sequence to target and destroy incoming phages. But phages also have many ways to evade and disrupt bacterial defenses. In this study, a phage is discovered that inserts its own genome into the CRISPR/Cas sequence in the bacterial genome, disrupting the bacterial defenses. To escape the defenses while it is doing this insertion, it carries genes for previously-unknown anti-CRISPR proteins. But inserting and removing a viral sequence from the bacterial genome is not always a clean procedure.   Journal Paper: Varble A, Campisi E, Euler CW, Maguin P, Kozlova A, Fyodorova J, Rostøl JT, Fischetti VA, Marraffini LA. 2021. CRISPR is the cutting edge of genetic-modification techniques and will likely transform scientists ability to change the DNA of virtually any organism on the planet, including humans. How will this technology be used? What should it be used? Where will this usage of the technology take humanity? Correction: During the video, Andy says the Cas9 protein reads the CRISPR DNA and makes RNA. This is not correct. The RNA is made by the RNA polymerase of the cell. Cas9 will bind to THAT RNA after it is cleaved. It is that Cas9 / RNA complex which then binds the DNA and clips it. What’s Left? What will be some of the ethical implications along with that and what exactly is genetic engineering technology like CRISPR actually being used for today?    On this episode Cormac speaks to former NASA employee and biohacker Josiah Zayner who sells CRISPR DNA kits for use at home, Director for the Centre for Genetics and Society Marcy Darnovsky talks about the ethical implications involved, Director of Communications from the Centre for Genetics and Society Kevin Doxzen about gene editing to save chocolate and assistant Professor from Trinity College Dublin's Immunology and Biochemistry Department Dr. Vincent Kelly, about where and how the technology is actually being used today.

* If scientists cure aging, you could periodically go on a pleasure blitz, and then voluntarily go into a coma while your dopamine levels recover or while your recreational drug tolerances go back down to base levels. Since you're not aging, there's no need to worry about wasting time. Plus, you could lose weight while in the coma * If they made a CRISPR DNA-editing device which connected to a cell phone and allowed you to design changes to your body using an app, you'd be trusting a company like Apple to edit your genes. Think about all of the update errors that might happen (to your body) * If it takes a few days for your brain to replenish a pleasure chemical like dopamine, in the meantime, you could schedule leisure activities which require different pleasure chemicals, such as a rollercoaster which requires lots of adrenaline but not much dopamine * An alarm clock that could shake your bed or shock you to wake you up could also defibrillate you, scare you during horror movies, and pleasure you. But don't use it if you're pregnant (wink wink) Shake and Shock Alarm Clock (@LordBluntsWorth aka "Spruce Rice" 00:39) CRISPR DNA Editing App (Avery Sommerfeld 12:14) Happiness Coma (@thomaswalma 21:51) Pleasure Chemical Vacation Planning (@thomaswalma 33:16) Recorded at PodcastDetroit.com
