Proteomic Identification of the MYST Domain Histone Acetyltransferase TIP60 as a Coactivator of the Myeloid Transcription Factor C/EBPα

The transcription factor C/EBPα is a key player in granulopoiesis and leukemogenesis. In the present study, we sought to identify C/EBPα interacting proteins. A glutathione-S-transferase-C/EBPα fusion protein was used to pull down interacting proteins from U937 nuclear extracts. These proteins were analyzed by 2-D gel electrophoresis or 1-D nano LC and identified by mass spectrometry. The interaction between C/EBPand two novel interacting partners, the cell cycle regulator protein MCM5 and the MYST domain histone aceyltransferase TIP60, was confirmed by using pull-down and co-immunoprecipitation experiments. TIP60 was able to markedly enhance C/EBPα mediated transcriptional activation in reporter gene assays, suggesting that TIP60 is a co-activator of C/EBPα. This co-activator function of TIP60 was dependent on its intact histone aceyltransferase domain and on the C/EBPα DNA binding domain. TIP60 was found to be associated with the human C/EBPα promoter in vivo in a chromatin immunoprecipitation assay with a concomitant increase in histone H3 and H4 acetylation. Furthermore, we observed a lower expression of TIP60 mRNA in undifferentiated U937 CD11b- cells compared to retinoic acid induced differentiated U937 CD11b+ cells suggesting that higher TIP60 expression is associated with myeloid differentiation. Correlated expression between C/EBP and TIP60 was also observed in certain leukemia subtypes. These findings point to a functional synergism between C/EBP and TIP60 in myeloid differentiation and suggests that TIP60 might be an important player in leukemogenesis.

Functional inactivation of the transcription factor CAAT Enhancer Binding Protein Alpha (C/EBPα) either by mutation or direct protein-protein interaction leads to acute myeloid leukemia (AML), whereas the activation of C/EBPα restores normal myeloid cell differentiation. We and others have shown that protein-protein interactions of C/EBPα play a pivotal role in myeloid differentiation and AML. In the present study we applied proteomics based mass spectrometry to identify C/EBPα interacting proteins on a proteome-wide scale. For this, the GST and GST-DNA binding domain of C/EBP (GST-DBD) was incubated with nuclear extracts of U937 cells. Interacting proteins separated by two-dimensional gel electrophoresis were identified by MALDI-TOF mass spectrometry. Using this approach we could identify PAK6, MADP-1, ZNF45 and the c-Jun N-terminal kinase 1 (JNK1) as C/EBPα interacting proteins. Since JNK1 activates c-Jun, the contra-player for C/EBPα, we hypothesized that the JNK1 and C/EBPα interaction might have some biological relevance. We could show that JNK1 binds to the DNA binding domain of C/EBP in GST-pull-down and to the C/EBPα in co-immunoprecipitation assays in-vitro and in-vivo respectively. JNK1 phosphorylates and increases the half life of the C/EBPα protein via inhibiting its ubiquitination and thereby enhances its transactivation and DNA binding activity. Furthermore, JNK mRNA expression as well as its kinase activity is lower in the AML patients in comparison to normal bone marrow mononuclear cells which implicates a possible mechanism of C/EBPα inactivation in certain acute myeloid leukemia subtypes. Thus our data suggest that a JNK activity is required for C/EBPα activation in myeloid cells and a loss of JNK regulated C/EBPα expression may contribute to leukemogenesis.

Overexpression of proto-oncogene c-jun and constitutive activation of the Jun NH2-terminal kinase (JNK) signaling pathway have been implicated in the leukemic transformation process. However, c-jun expression has not been investigated in acute myeloid leukemia (AML) cells containing the most common chromosomal translocations. t(8;21) is one of the most common AML-associated translocation and results in the AML1-ETO fusion protein. Overexpression of AML1-ETO in NIH3T3 cells leads to increased phosphorylation of Ser63 in c-Jun, which is generally JNK dependent. The role of the JNK signaling pathway for the functional properties of AML1-ETO is, however, unknown. In the present study we found high expression levels of c-jun mRNA in t(8;21), t(15;17) or inv(16) positive patient cells by microarray analysis. Within t(8;21) positive patient samples, there was a correlation between AML1-ETO and c-jun mRNA expression levels. In myeloid U937 cells, c-jun mRNA and c-Jun protein expression levels increased upon induction of AML1-ETO. AML1-ETO transactivated the human c-jun promoter through the proximal AP-1 site via activating the JNK signaling pathway. JNK targets c-Jun and ATF-2, which also bind to the proximal AP-1 site in U937 cells, were also phosphorylated upon AML1-ETO induction. Furthermore, AML1-ETO induction increased the DNA binding capacity of c-Jun and ATF-2 to the proximal AP-1 site of the c-jun promoter, which might result in their enhanced transactivation capacities. Interference with JNK and c-Jun activation by using JIP-1 or a JNK inhibitor reduced the transactivation capacity of AML1-ETO on the c-jun promoter and the pro-apoptotic function of AML1-ETO in U937 cells. AML1-ETO seems to activate the JNK signaling pathway by inducing the expression of a cytoplasmic factor, possibly G-CSF, because supernatant of AML1-ETO expressing cells was sufficient to induce phosphorylation of JNK and c-Jun in wildtype U937 cells. This data demonstrates a novel mechanism of how AML1-ETO might exert positive effects on target gene expression and identifies the proto-oncogene c-jun as a common target gene in AML patient cells.

Oncosuppressive properties and a low pathogenicity in adult animals make the two rodent parvoviruses MVMp and H1 promising vectors for cancer gene therapy. Recombinant vectors were developed from genomes of these viruses in which a part of the capsid-protein-coding sequence was replaced by a transgene. These recombinant viruses can replicate their genome and express their transgene, but they are unable to produce progeny viruses. Such vectors are produced by cotransfection of the recombinant virus genome and a helper plasmid which provides the capsid proteins. During this procedure, RCVs (Replication Competent Viruses) can be generated, probably by homologous recombination. As a consequence, RCVs can express their capsid proteins and produce progeny viruses. In the present work, second-generation H1- and MVMp- based recombinant virus genomes were developed in order to minimise RCV levels in the recombinant virus preparations. By constructing chimeras and pseudotypes between MVMp and H1 viruses, virus preparations with high titres were obtained which did not show any detectable RCVs in plaque-assays. This corresponds to a 100-17000fold decrease of RCVs for the chimeras and pseudotypes. In this work we also characterised the host-cell-tropism of the chimeras and the pseudotypes. The chimeric vectors and Pseudotypes allowed us to show that the restriction for viral replication of the rat H1 genome in murine A9 cells is not due to the H1 capsids, but can be ascribed to a 1800 bp long DNA region of the H1 genome, located in the NS coding sequence. To our knowledge this is the first report that convincingly shows that the tropism of a rodent parvovirus is determined by the viral genome. Since a recombinant MVMp genome packaged with either H1 or MVMp capsids led to viral DNA replication in A9 cells, virus entry is not responsible for the lack of recombinant H1 vector replication in these cells. After transfection of A9 cells with H1 viral DNA, replication and viral protein expression took place, whereas after H1 infection it did not. Thus we can conclude that the cause for the restricted H1 virus replication has to be found after virus entry but before the beginning of viral gene transcription. It is usually accepted that viral gene transcription needs the conversion of single-stranded Summary 134 DNA into double-stranded monomeric replicative form. Most probably this conversion reaction is disordered in A9 cells for the H1 genome. A defective double-stranded conversion might be due to an interaction of VP proteins with the viral single stranded DNA sequence. Another possibility is an interaction of a cellular protein with the viral single stranded DNA which then leads to an inhibition of the viral DNA conversion. It might be also possible that a cellular factor which is needed for the conversion reaction can not bind to the H1 genome in A9 cells. Interestingly, in most of the tested murine cell lines the tropism was determined by the MVMp genome, while for all human cell lines the tropism was determined by the virus capsid. The second part of this work focused on the effect that H1 virus infection on cellular gene expression. For this purpose, the cDNA array technique was used. It was previously shown that parvovirus H1 infection leads to the activation of caspase 3 and apoptotic cell death of the human monocytic U937 cell, in a way similar to the apoptotic cell death induced by TNFα. Moreover, the c-myc gene, overexpressed in these cells, is down-regulated during H1 infection. In agreement with these data, we were able to show -by comparing the gene expression profiles between buffer treated and H1 infected synchronised U937 cells- genes coding for caspases 2, 4 and 8 were upregulated after H1 infection. In addition, different genes associated with the TNFα complex were found to be up-regulated, as well as the mad4 gene, which is able to inhibit the transcriptional activity of c-myc by binding its main partner max. Moreover, 2 genes involved in oestrogen-synthesis were found to be up-regulated after infection. Oestrogen was shown to have some protective effect against TNFα induced apoptosis in U937 cells. Induction of these genes might reflect a defence mechanism of the cell. Also, a gene encoding for a subunit of the 26s proteosome and different genes involved in differentiation processes or in apoptosis were up-regulated in infected cells. Few genes were shown to be down-regulated after infection, such as the adenylate cyclase and cAMP dependent transcription factor 1. Altogether, our data give a better understanding of the molecular pathways which are used by the parvovirus and enlighten the diversity of cellular responses to a H1 virus infection in U937 cells.

Downregulation of Tumor Necrosis Factor Expression in the Human Mono-Mac-6 Cell Line

Mono-Mac-6 cells, but not U937 cells, can be Induced to rapidly express tumor necrosis factor (TNF) mRNA and protein when triggered with Ilpopolysaccharlde (LPS) at 1 pg/mI. Preincubatlon of the cells for 3 d with low amounts of LPS (10 ng/mI) results In nearly complete suppression of TNF secretion. This downreguiatlon appears to occur at the pretranslational level since specIfIc mRNA is virtually undetectable under these conditions. By contrast, the same prelncubatlon with 10 ng/mI LPS results in enhanced phagocytosls (28.6-67.2% for Staphylococcus aureus), demonstrating that not all monocyte functions are suppressed. While these results show that only stringent exclusion of LPS from culture media allows for Induction of TNF In the Mono-Mac-6 cell line, the pronounced effect of LPS preincubatlon may also provide a suitable model with which to study the mechanisms of LPS-lnduced desensitizatIon.
