Near Falls Creek in Jefferson County is a region known as Beechwoods, a locality which includes some of the finest farmland in Washington Township. The area was settled in 1824 by Henry Keys, Alexander Osborn, John McIntosh, John McGhee and Thomas Moore. These early settlers, mostly of Scotch and Irish origin, named the community after the large number of beech trees they found there. It is one of these founders, Thomas Moore, who played a role in one of the most colorful chapters in the history of Jefferson County. It is a story that seems ripped right out of the pages of a novel, and features murder, suicide, a haunted farm and a buried treasure of silver and gold.

We kick off season 2 with a bang, talking about complex molecule synthesis with John McIntosh and Nastaran Salehi. Cyclic dinucleotides (CDNs) are a total synthesis fan's dream with 10 stereocenters (including 2 at phosphorus) arranged around a 13 membered macrocycle formed by 2 non-canonical nucleosides. These endogenous secondary messenger molecules have attracted attention because of their activation of the immune system via the STING pathway. We go into all aspects of the innovative biocatalytic cascade that the team designed for their synthesis, including: 1) How a bald eagle cyclic guanosine-adenosine synthase (cGAS) was engineered to form the macrocycle; 2) How cGAS was leveraged by medicinal chemists to greatly accelerate drug discovery; 3) How the team strung together and optimized a 4-enzyme cascade to deliver a CDN directly from nucleotide building blocks. "About 12 miles south of Tupelo, Mississippi is an exhibit called Tockshish. This exhibit tells about a site on the Old Natchez Trace known as Tockshish. This was McIntoshville, a community of white men and Indians built up around the home of the British Indian agent, John McIntosh, who settled there before 1770. When the old Indian trail was established as a national road in 1801, Tockshish became a relay station where post riders carrying mail between Nashville and Natchez could exchange weary horses for fresh ones. The post riders would then move on with their mail bags. For them it was a 5 day journey from there to Nashville and 7 days to Natchez. Girish Menon joined STiR Education as CEO in January 2021 after five years as Chief Executive at ActionAid UK. He brings more than 30 years' experience as a leader in the international development sector, having previously held roles as International Programmes Director and Deputy CEO at Water Aid UK, where he was responsible for programmes in 22 countries across sub-Saharan Africa and south Asia. Girish was born and brought up in India, and started his career with the Aga Khan Rural Support Programme. He has also worked at Action Aid, Plan International and DFID in India. Since moving to London in 2005, he has served on the boards of various non-profit organisations. He is currently on the board of Hope and Homes for Children. John McIntosh is the Director of Design and Programme Readiness and oversees the design of STiR's programmes for teachers and government officials. Today’s featured guest is John McIntosh. John is 60 years old and is from Glasgow, Scotland, and has stammered for as long as he can remember. After graduating with an honors degree in English Literature, John worked in social care for many years, mainly because he could not envisage entering a profession that made big demands on his speech. At the age of 35, he was married with two young daughters and decided to become a high school English teacher which he did until last August, when he retired. In 2004, when he was 45, he completed the McGuire Program Course, and was active in that program for many years. He currently is on the management committee of the Scottish Stammering Network, the main support group for PWS in Scotland, and helped establish the podcast “And Oh And On That Note”; which aims to get stammering voices out there in the public domain.
