In this 3rd episode of The Alzheimer's Solutions Revolution Podcast, I, Ralph Sanchez, talk with my good friend and co-host Susan Brender, as we begin this special podcast series designated as the Think Ahead podcast series. In the Think Ahead series, which will be published here over the coming weeks, I'll provide an extensive overview on the numerous risk factors associated with late-onset Alzheimer's disease (LOAD), and I expand on the research that sheds light on how you can modify and reduce your risk for LOAD and dementia as you age. In the first Think Ahead episode, I lay down the foundation for understanding the genesis and difference between early onset Alzheimer's disease (EOAD) and LOAD, and the science-based findings that illuminates the association between many risk factors—including genes and genetic variants, that are linked to Alzheimer's disease. Additionally, I'll begin the overview on the hallmark protein-based lesions (amyloid plaques and neurofibrillary tangles) that are associated with Alzheimer's is discussed. EOAD Gene Identified in Rhodesian Ridgebacks Adam Boyko, co-founder of Embark, and Rhodesian Ridgeback breeder Denise Flaim join host Laura Reeves to discuss EOAD (Early Onset Adult Deafness) in Ridgebacks and Embark's discovery of the genetic cause that can identify affected and carrier dogs before the condition develops. “EOAD is early onset adult deafness,” Flaim said. “It's a form of deafness that's not related to color. Many breeds, like dalmatians, the way the white overlays the cochlea impacts deafness. This is just a simple autosomal recessive, inherited the same way brown nose color is. If you have two copies of this recessive gene, if you are a Ridgeback, you become deaf. “The interesting thing in Ridgebacks is these puppies are born hearing. So if you are a breeder who wants to do your due diligence and BAER test your puppies at 8 weeks, they'll all hear. What then begins to happen is they start to go progressively deaf. The males quickly, usually by six months are completely deaf and the bitches can take from 12 to 18 plus months. “(This test) identifies if your dog is a carrier or not. If your dog is a carrier, you simply don't breed it to another carrier and you're free and clear. This is a really important point because the tendency among dog breeders, especially those who want to be really, really virtuous and really, really ethical, is to say, ‘oh, I'm going to identify all these carriers and get them out of my breeding program.' Which is, of course, what you don't wanna do. “You certainly don't want to increase the frequency of this gene in the population, but what you want to do is manage it. The thing is it's never what you know, it's always what you don't know. It's never what you worry about, it's always what you don't worry about. So yes, we've got this marker for deafness, but that deafness carrier you're throwing out of your breeding program may not carry for a really devastating disease for which we don't have a test. So, like anything, moderation and taking the bigger view is really important.” Teamwork for the win… “Project Dog started working with breeders like Denise,” Boyko said, “and recruited a whole bunch of samples and was able to find an associated region. But sequencing of a deaf dog didn't yield any candidate variants that were causing the mutation. So, you're sort of stuck in this world, do we want to offer a linkage-based test, which we know probably isn't going to be 100% accurate, or do we wait until we can find a mutation test. For a while, there was just a linkage test was all that could be offered. “So, Embark came onto the scene. We, of course, have a large database of dogs, of Ridgebacks and others, and so we were able to recruit more cases, more controls. We were able to verify the association Project Dog found. It's like ‘yeah, this is definitely on chromosome 18, right here. There had been advances in the genomics. We put some scientists on it …. and sure enough a mutation did find itself. It was in gene EPS 8L2, which was a fantastic gene because it's also associated with early onset deafness in humans, in recessive forms.” Hear the REST of the story on today's episode. Adam Boyko Adam is an associate professor in Biomedical Sciences at the Cornell University College of Veterinary Medicine, focused on the genomic investigation of dogs. Adam's research has addressed fundamental questions of dog evolution and history, disease and trait mapping, and advancing genomic tools for canine research. Adam has coauthored over 40 peer-reviewed scientific papers, including research in Nature, Science, and the Proceedings of the National Academy of Science and is a member of the Board of Trustees for the Morris Animal Foundation. He is a graduate of the University of Illinois, Urbana-Champaign and received an MS in Computer Science and a PhD in Biology from Purdue University before his postdoctoral work at Cornell and Stanford....

Die Alzheimer Demenz (AD) ist eine degenerative zerebrale Erkrankung, die klinisch durch den Verlust vielfältiger kognitiver Fähigkeiten gekennzeichnet ist. Ein geringer Prozentsatz (65 Jahre) erfolgte im Sinne einer homogeneren Subgruppierung und sollte eine aussagekräftigere Assoziation der genetischen Vulnerabilitätsmarker ermöglichen. In die Kontrollgruppe wurden ausschließlich gesunde Probanden ohne demenzielle Erkrankungen in der Familie eingeschlossen. Neben einer grundsätzlichen Assoziation mit der AD wurde in einem zweiten Schritt der Zusammenhang zwischen den 19 Polymorphismen und der individuellen Gedächtnisleistung als intermediärer Phänotyp untersucht. Die Gedächtnisleistung wurde mittels standardisierter Tests erhoben. Die 19 SNPs (rs4412638, rs12024093, rs12043872, rs3009113, rs913858, rs12113404, rs12672536, rs4722589, rs10998072, rs10998076, rs1162757, rs16925347, rs1162756, rs1683152, rs784566, rs11170562, rs784563, rs2249381, rs1716966) wurden mittels iPLEX-Verfahren genotypisiert und die massenspezifischen Produkte im MALDI-TOF Massenspektrometer ausgewertet. Signifikante Assoziationen konnten dabei insbesondere zwischen den drei Genen HNRNPA2B1, PCBP2, ELAVL4 und der Alzheimer Demenz, sowie der Gedächtnisleistung festgestellt werden. Auf diesen drei Genen zeigten sich fünf Marker mit dem Phänotyp der AD bzw. insbesondere mit der EOAD assoziierbar: Auf ELAVL4 ließen sich rs3009113 und rs12024093 in der Patienten-Gesamtgruppe, sowie im EOAD-Kollektiv mit der AD assoziieren; auf PCBP2 waren rs784563 und rs2249381 in der Gesamtgruppe der Patienten auffällig; auf HNRNPA2B1 ließ sich rs4722589 mit der EOAD assoziieren. Diese und weitere Polymorphismen der angeführten drei Gene konnten sowohl in der EOAD- als auch in der LOAD-Gruppe mit einer verminderten Gedächtnisleistung assoziiert werden. In der Haplotypanalyse ließen sich diese Befunde grundsätzlich bestätigen. Zusammenfassend lassen die Resultate der vorliegenden Untersuchung einen Zusammenhang zwischen drei der untersuchten RBPs (HNRNPA2B1, PCBP2 und ELAVL4) und dem Phänotyp Alzheimer Demenz, sowie dem intermediären Phänotyp Gedächtnisleistung vermuten. SNPs in intronischen Bereichen können regulatorische Sequenzen wie die Polyadenylierung- oder Spleißfaktorbindungsstellen beeinflussen und zu Veränderungen in der Transkriptionsrate oder im Spleißprozess führen. Zudem kann durch Muationen die natürliche Aggregationsbereitschaft RNA-bindender Proteine, sich in RNA-granules zusammenzulagern, verstärkt werden und damit direkt zellschädigend wirken. Da über Funktion und Struktur der untersuchten RBPs gegenwärtig noch relativ wenig bekannt ist und es bislang keine vergleichbaren Referenzstudien bezüglich Alzheimer gibt, ist weiterführende Forschungsarbeit zur abschließenden Interpretation der Ergebnisse notwendig. Aufgrund der bis dato vorliegenden Ergebnisse stellen RNA-bindende Proteine insgesamt einen interessanten Kandidaten für die Alzheimerforschung dar.

Kick Off Show with Rick Phelps Founder of Memory People Rick has Early Onset Alzheimer's Disease, known as EOAD. Instead of fading away with this disease Rick has done something different by raising awareness about the disease and giving a face to it by making a video log of his life with the disease. He has also created Memory People a closed group on Facebook for people who are memory impaired, their caregivers both family and professional, and advocates of the disease.
