H., Karelson, M., Saarma, M. Abstract: N6-Methyladenosine (m6A) is the most common mRNA base modification in eukaryotes. Methylation of adenosine residues to m6A contributes to the regulation of splicing, transport, stability, and translation of mRNA and two main classes of enzymes regulate it. The formation of m6A is catalysed by a methyltransferase complex containing methyltransferase-like 3 (METTL3), METTL14, and Wilms' tumour 1-associated protein (WTAP) as well as monomeric METTL16. Demethylation of m6A is catalysed by the fat mass and obesity-associated protein FTO and the RNA demethylase AlkB homolog 5 (ALKBH5). The m6A mRNA methylation dysregulation occurs in the nervous system and in Parkinson's disease (PD), but it remains poorly studied. Moreover, the role of m6A mRNA methylation in neuronal survival, neuroprotection, and neuroregeneration is unclear. We have earlier used high-throughput virtual screening of large compound libraries and identified four unique small-molecule ligands that activate m6A mRNA methylation by binding to the METTL3/14/WTAP complex and enhancing the binding of the methylation substrate SAM to nanomolar concentrations. Following this, we now discovered that two methyltransferase activators at 10 nM concentrations supported the survival and protected dopamine (DA) neurons in culture in growth factor deprivation and 6-hydroxydopamine (6-OHDA) neurotoxin models. In contrast, METTL3/14 inhibitor STM2457 triggered death of DA neurons. For clinical translation we also tested the most efficient compound C4 on induced pluripotent stem cell-derived human DA neurons and in animal model of Parkinson's disease (PD). C4 compound protected human DA neurons from 6-OHDA-induced cell death and increased neurite outgrowth and the number of processes demonstrating that it has both neuroprotective and neurorestorative properties. METTL3/14 activator C4 improved motor behaviour and protected DA neurons and their fibres faster and much more efficiently than GDNF in the rat 6-OHDA model of PD. These are the first specific activators of METTL3/14/WTAP and first demonstration that m6A regulators can protect and regenerate neurons. These data demonstrate that m6A mRNA methylation is a novel pathway regulating neuronal survival and regeneration. Copy rights belong to original authors. Abstract: N6-methyladenosine (m6A) methylation of RNA by the methyltransferase complex (MTC), with core components including METTL3-METTL14 heterodimers and Wilms' tumor 1-associated protein (WTAP), contributes to breast tumorigenesis, but the underlying regulatory mechanisms remain elusive. Here, we identify a novel cleaved form METTL3a (residues 239-580 of METTL3). We find that METTL3a is required for the METTL3-WTAP interaction, RNA m6A deposition, as well as cancer cell proliferation. Mechanistically, we find that METTL3a is essential for the METTL3-METTL3 interaction, which is a prerequisite step for recruitment of WTAP in MTC. Analysis of m6A sequencing data shows that depletion of METTL3a globally disrupts m6A deposition, and METTL3a mediates mTOR activation via m6A-mediated suppression of TMEM127 expression. Moreover, we find that METTL3 cleavage is mediated by proteasome in an mTOR-dependent manner, revealing positive regulatory feedback between METTL3a and mTOR signaling. Our findings reveal METTL3a as an important component of MTC, and suggest the METTL3a-mTOR axis as a potential therapeutic target for breast cancer. Copy rights belong to original authors. In this episode of the Epigenetics Podcast, we caught up with Dr. Chuan He, John T. Wilson Distinguished Service Professor at the University of Chicago, to talk about his work on the influence of dynamic RNA methylation on gene expression. More than 150 structurally distinct post-transcriptional modifications of RNA have been identified at thousands of sites in cellular RNA molecules. Some RNA modifications are dynamic and may have critical regulatory roles analogous to modifications of protein and DNA. Understanding the scope and mechanisms of dynamic RNA modifications thus represents an emerging research frontier in biology and medicine. The internal N6-methyladenosine (m6A) modification in messenger RNA (mRNA) is one of the most abundant RNA modifications in eukaryotes. On average, this RNA base modification is present in approximately 3 sites in every mammalian mRNA molecule. The m6A modification has been shown to be critically important for cellular differentiation, animal development, and a range of biological signaling and stress responses. In 2011, Chuan He's lab reported that m6A in mammalian mRNA can be oxidatively demethylated in vitro and inside cells by the enzyme FTO (a fat mass and obesity-associated protein), a major obesity factor This discovery identified FTO as the first known RNA demethylase. Chuan He’s lab also subsequently identified ALKBH5 as the second RNA demethylase that controls mammalian spermatogenesis. More recent work in the He lab has focused on characterization of the YTH family proteins as the “reader” proteins that detect and bind methylated RNAs in mammalian cells to regulate their stability and translation. Functional characterizations revealed that YTHDF2 regulates cytoplasmic localization and mediates the decay of methylated mRNA, YTHDF1 promotes translation of methylated mRNA by facilitating translation initiation, and other methylated RNA readers also contribute to the regulation of mRNA storage, transport, and cellular localization.   Research in the He lab also identified a core protein complex comprised of two subunits: METTL3 and METTL14, and an accessory factor WTAP, which mediates cellular m6A RNA methylation. Cellular RNA methylation levels can have profound impacts on normal cellular differentiation and cancer cell proliferation. Their current work focuses on how the methylation selectivity is achieved. All legends are local in a way, and when one local person tells a tale of a strange encounter, it can be a cause for skeptical snickers from the townsfolk. When more than a hundred people from the same region all see the same thing, it can lead to books, a major Motion Picture and even an annual festival. Such is the case with Point Pleasant, West Virginia and the Ohio River Valley's own "Mothman" — whose local sightings turned into national legend. There are several elements that contribute to the intense curiosity that still surrounds the stories of the Mothman, himself described as a winged, humanoid creature with glowing red eyes and a sympathetic, if mysterious nature. One of them is the sudden appearance of a friendly, inter-dimensional road tripper named Indrid Cold who just wants to get to know you better and may be searching for a cryptid on the loose . Another is the unsettling thought that these encounters may have portended a disaster that took the lives of 46 people exactly 13 months later.
