Ten people have died and about 30 inquired following an accident involving a truck and a bus on the N6 between Reddersburg and Springfield in the Free State. The road had to be closed and traffic diverted as a result. Free State Emergency Medical Services officials are attending to the trapped and injured passengers. The injured have been transported to hospitals in Bloemfontein and the nearby Springfield. Sakina Kamwendo spoke to SABC reporter, Ishmael Modiba in the Free State

Eastern Cape traffic officials have closed Barkly Pass, on the R58 route between Barkly East and Elliot, this morning due to heavy snowfalls. The Eastern Cape Transport Department says some vehicles are stuck in the snow on the R58. Authorities are also keeping an eye on snowfall-prone routes such as Penhoek Pass on the N6 between Komani and Jamestown and others mainly in mountainous areas. Abstract: The METTL3-METTL14 methyltransferase complex adds N6-methyladenosine (m6A) to mRNA with profound impacts on cell fate. Studies delete METTL3 or METTL14 interchangeably to define the role of m6A in target tissues despite a lack of data confirming that these deletions are equivalent. Intestinal epithelial METTL14 deletion triggers stem cell death in the colon with no overt phenotype in the small intestine. The effect of METTL3 deletion in the same tissues remains unknown. We report that intestinal epithelial METTL3 deletion caused unexpected severe defects in the small intestine, including crypt and villus atrophy associated with cellular senescence and death in the crypt transit amplifying zone. Ribosome profiling and m6A-sequencing demonstrated downregulated translation of hundreds of unique methylated transcripts, including genes essential to growth factor signal transduction, such as Kras. Our study suggests that METTL3 is essential for small intestinal homeostasis via enhanced translation of growth factor signaling in crypt transit amplifying cells. We now report on a double transgenic APPNL-G-F MAPTP301S mouse that at 6 months of age exhibits robust A{beta} plaque accumulation, intense MAPT pathology, strong inflammation and extensive neurodegeneration. The presence of A{beta} pathology potentiated the other major pathologies, including MAPT pathology, inflammation and neurodegeneration. However, MAPT pathology neither changed levels of amyloid precursor protein nor potentiated A{beta} accumulation. The APPNL-G-F/MAPTP301S mouse model also showed strong accumulation of N6-methyladenosine (m6A), which was recently shown to be elevated in the AD brain. M6A primarily accumulated in neuronal soma, but also co-localized with a subset of astrocytes and microglia. The accumulation of m6A corresponded with increases in METTL3 and decreases in ALKBH5, which are enzymes that add or remove m6A from mRNA, respectively. The Federal Government's expenditure in the first six months of 2022 exceeded revenue by N6.23tn. The Federal Government recorded N3.2tn and N2.74tn fiscal deficit in the first and second quarters of 2022. It however exceeded the Federal Government's fiscal deficit budget of N1.6tn for the period under review. A breakdown revealed that recurrent expenditure, capital expenditure, and transfers accounted for 81.0 percent, 13.3 percent, and 5.7 percent of total expenditure, respectively. It noted that the fiscal operations of the Federal Government in the second quarter of 2022 remained aligned with the extant fiscal frameworks, which sought to achieve macroeconomic stability, improve revenue generation, and create fiscal space to boost infrastructural development, among other objectives.

A new review paper was published in Aging (listed as "Aging (Albany NY)" by MEDLINE/PubMed and "Aging-US" by Web of Science) Volume 14, Issue 19, entitled, “RNA modifications in aging-associated cardiovascular diseases.” Cardiovascular disease (CVD) is a leading cause of morbidity and mortality worldwide that bears an enormous healthcare burden. Aging is a major contributing factor to CVDs. Functional gene expression network during aging is regulated by mRNAs transcriptionally and by non-coding RNAs epi-transcriptionally. RNA modifications alter the stability and function of both mRNAs and non-coding RNAs and are involved in differentiation, development and diseases. In this new review paper, researchers Xinyu Yang, Priyanka Gokulnath, H. Immo Lehmann, Zhitao Hou, Sun Yang, Liangzhen You, Guoxia Zhang, Yanwei Xing, Ji Lei, Guoping Li, Shuwen Guo, and Hongcai Shang from Fangshan Hospital and Dongzhimen Hospital (affiliated with Beijing University of Chinese Medicine), Massachusetts General Hospital, Harvard Medical School, Heilongjiang University of Chinese Medicine, and Chinese Academy of Chinese Medical Sciences reviewed major chemical RNA modifications on mRNAs and non-coding RNAs, including N6-adenosine methylation, N1-adenosine methylation, 5-methylcytidine, pseudouridylation, 2′ -O-ribose-methylation, and N7-methylguanosine, in the aging process with an emphasis on cardiovascular aging. The Senate rejected the 2023-2025 medium-term expenditure framework and fiscal strategy paper which proposed A N6 trillion tax and import duties waivers proposed for in the N19.76 trillion 2023 budget which also contained A deficit of N12.4 trillion. The chairman of the senate panel, senator Solomon Adeola says that both the projected N12.43 trillion budget deficit and the n6trillion tax and import duty waivers should be critically reviewed downward before sending the proposals to the national assembly for consideration and approval. He also told the minister to look into the list of beneficiaries of the waivers for the required downward review to N3 trillion in order to give room for the reduction of the N12.43 trillion deficit figure.

The International Monetary Fund has warned that the Nigerian government may spend nearly 100 per cent of its revenue on debt servicing by 2026. The IMF's Resident Representative for Nigeria, Ari Aisen, says the biggest critical aspect for Nigeria is that they have done a macro-fiscal stress test, and the interest payments as a share of revenue and as you see us in terms of the baseline from the federal government of Nigeria, the revenue, almost 100 per cent, is projected by 2026 to be taken by debt service. IMF also warned that with fuel subsidy payments averaging N500 billion monthly, total expenditure on subsidy could hit a record N6 trillion by the end of the year.

The Federal Inland Revenue Service has generated a record N6.4 trillion as revenue in 2021, the highest in its history. This year's revenue collection exceeded that of 2019 when the agency raked in N5.3 trillion under the administration of Babatunde Fowler. Spokesman to FIRS Chairman, Oluwatobi Wojuola, says that despite the limitations faced in 2020 and 2021, the service achieved over 100 per cent of its collection target. Wojuola stated that in line with the law, 2021 income tax revenue was a function of the outcome of business activities in 2020.

Aging-US published a Special Collection on Eye Disease which included "ALKBH5-mediated m6A demethylation of FOXM1 mRNA promotes progression of uveal melanoma" which reported that ALKBH5, a key component of the N6-methyladenosine (m6A) methyltransferase complex, was significantly elevated in uveal melanoma cell lines and inhibited tumor growth in vitro and in vivo. EP300-induced H3K27 acetylation activation activated increased ALkbH5 expression and inhibited UM cell proliferation, migration, invasion and increased apoptosis in vitro. Besides, it may promote UM metastasis by inducing epithelial-to-mesenchymal transition (EMT) via demethylation of FOXM1 mRNA, which increases its expression and stability. It is a potential prognostic biomarker and therapeutic target in UM. Dr. Jingxiang Zhong and Dr. Lian Liu said, "Uveal melanoma (UM), which originates from melanocytes, is the most common primary intraocular malignancy in adults." Up to 50% of patients with primary malignant meningitis (UM) will develop metastatic disease. The most common sites of UM metastasis are the liver (60.5%), lungs (24.4%), skin/soft tissue (10.9%), and bone (8.4%). Unfortunately, the 1-year survival rate of UM patients with metastases is only 15% [5]. There is a pressing need to find useful prognostic biomarkers and therapeutic targets for this disease. Alkhabh5 overexpression of ALKBH5 promotes EMT of UM by upregulating FOXM1 expression via demethylating the m6A modification and further increasing the stability of FOXm1 mRNA. The Zhong/Liu Research Team concluded in their Aging-US Research Output, "we demonstrate that ALKBH5, which is positively regulated by epigenetic modifications of H3K27 acetylation, promotes tumor progression by inducing tumor EMT and increasing FOXM1 expression via m6A demethylation (Figure 7). Therefore, ALKBH5 is a potential target of UM molecular therapy." Todd meets with Sammi, a friend and mother of a young man with autism, to talk about the mixed feelings that come with her son's transition to adulthood for s1. E6. N6. of the Otherness Podcast titled Part Boy, Part Man. A Kind of Transition to Autism Adulthood.Through the lens of a mother, Todd reflects on the fear and growth that come through the development from childhood to adulthood for a young man on the spectrum.As Todd was developing the Otherness Podcast, he reached out to Sammi, a friend and mother of one of his former patients. Sammi was intrigued by the first episodes and felt that the voice of those who spoke through the podcast resonated with her. Todd was also curious about how her son, Bailey, was feeling and functioning as he is now 21 and about to transition to adult services. This turned into a discussion that peels back some of the initial feelings of anxiety around “growing up” and finds richness in the acknowledgement that Bailey is not fully an adult despite the term “adult transition”. She describes how this transition is as much about missing what a wonderful community his school has been for him throughout his life, as it is about going into something new. Sammi is touring different day programs, wants Bailey to be involved in the work community, and wants him to have a sense of happiness and self. She shares her pain while seeing Bailey's non-autistic peers obtain goals that he may want, but may never have. These include driving, graduating high school and college, and forming romantic relationships. “There are always transitions for the neurotypical population that sometimes make the autism transitions a little painful.”Despite her heavy involvement in the autism community as an advocate and fundraiser, she feels there is so much to explore for Bailey in this new phase of his life. Some of these plans may work out, and some may not. The Central Bank of Nigeria has imposed a 6.98 naira service charge on Nigerians for all USSD transactions.A statement jointly signed by the apex bank and the Nigerian Communications Commission says Customers will pay a flat fee of N6.98 per transaction every time they use USSD services from March 16.The deal was part of an agreement reached with banks following a disagreement between banks and telecom firms over USSD and other text message transaction requests.The statement partly read that the approach is transparent and will ensure the amount remains the same, regardless of the number of sessions per transaction.The CBN noted that the charges were part of the agreements after banks and telecommunication operators met on Monday to discuss the ₦42bn debt owed mobile operators by banks.

Ejike dreams of becoming a governor. He arrived in Lagos in January 2020, leaving his parents in Ebonyi state. His brother promised to send him to school. But he’s in Lagos, selling CDs in traffic- just like his brother. Making between N6,000-N10,000 a day, Ejike supports his brother’s family of four. Ejike wants to become a governor in Lagos state. He says his brother promised to send him to school next year. He’s told he can’t go to school this year because of COVID 19. Ejike is 14 years old. Farouk is 17 years old. His mum sells herbs in Ibadan while his dad is an Alfa (Islamic scholar). He travelled to Lagos on his own and caters to himself, calling passengers at the motor park and sometimes bearing load for money in the market in Oshodi. During the COVID19 lockdown, Farouk says he barely survived. He couldn’t make money as markets were open only temporarily and buses were not moving. The Federal Executive Council on Wednesday approved N40 billion for the construction of road and ecological projects in different parts of the country.Minister of Works and Housing, Mr. Babatunde Fashola, said N14.528 billion was approved for the construction of Gasamu-Hamshi-Gogoram road in Yobe State. He said N991.851 million approved for the construction of a link road connecting Uneme-Tusamu-Odoga to Okpekpe in Etsako East Local Government Area of Edo State and N6.397 billion approved for the reconstruction of Mamabu-Donga road in Taraba South Local Government Area of Taraba State.

O. Abstract: Building and maintaining neuronal networks and cognitive functions require mRNA localization and regulated protein synthesis in neurons. RNA modification N6-methyl-adenosine (m6A) has recently been shown in axonal and synaptically localized mRNAs whose local activity is required for axon growth, synaptogenesis, and neuronal plasticity. However, no cellular pathways engaging local epitranscriptomic modulation are known to underlie these post-mitotic neuronal functions. Now we report that cytoplasmic m6A reader YTHDF1 is enriched in neurons and required for axonal, dendritic, and spine development. We show that m6A and YTHDF1 are part of a microtubule plus-end associated RNA granule that contains extensive networks of mRNAs organized by autism risk gene adenomatous polyposis coli (APC). Disrupting m6A signals by knocking down methyltransferase METTL14 or YTHDF1, or overexpressing autism or schizophrenia-associated missense mutations I311V or S399L in human METTL14, reduce expression of APC granule and tubulin, disrupt microtubule assembly and function. These results reveal a novel neuronal subcellular locus for epitranscriptomic regulation to promote post-mitotic neurodevelopment. Copy rights belong to original authors. Using a structure-based drug discovery approach, we identified a METTL3 inhibitor (UZH1a) with potency in a biochemical assay of 280 nM, while its enantiomer UZH1b is 100 times less active. The crystal structure of the complex of METTL3 with UZH1a illustrates the interactions that make it selective against protein methyltransferases. We observed a dose-dependent reduction in m6A methylation level of mRNA in several cell lines treated with UZH1a already after 16 h of exposure, as determined by triple-quadrupole LC mass spectrometry, while its enantiomer UZH1b was essentially inactive at concentrations up to 100 M. Interestingly, the kinetics of m6A level reduction in mRNAs followed a first-order reaction model, with a half-decay time {tau} of 1.8 h and a maximum m6A inhibition level of 70%, which is in line with the previously observed shorter half-life of m6A-modified mRNAs. Notably, treatment with the compounds did not alter cellular METTL3 levels, ruling out indirect effects on m6A levels. The effect of the m6A level depletion by UZH1a directly translated into growth inhibition of MOLM-13 leukemia cells, under short-term and long-term culture. Incubation of the MOLM-13 cells with UZH1a, but not with UZH1b, resulted in increased cell apoptosis and cell cycle arrest already after 16 h of incubation. Interestingly, other cell lines sensitive to METTL3 level (U2Os, HEK293T) did not reveal statistically significant differences between UZH1a and UZH1b in a cell viability assay, confirming that the degree of reliance on m6A signalling for survival can vary between cancers/cell types. The National Bureau of Statistics, in its report released on Wednesday stated that the total value of transactions through electronic payment channels recorded in the second quarter of 2020 stood at N263.78 trillion with a transaction volume of 2,160,436,659. The report was titled Selected Banking Sector Data: Sectorial Breakdown of Credit, e-Payment Channels and Staff Strength (Q2 2020)’. The report stated that online transfer transactions dominated electronic payments with N72.25 trillion with 1,317,621,686 in volume of transactions. The value of cheque transactions stood at N2.29 trillion while NEFT transfers accounted for N59.55 trillion. Transactions via Automated Teller Machines (ATMs) amounted to N37.44 trillion while Point of Sale (PoS) transactions stood at N999.91 billion. The latest data from Nigeria’s statistics agency shows that the demand for goods and services from China is still on the high as the Asian country tops the list of import trading partners by 31.41% in the second quarter of 2020. This is despite the sharp fall in the value of the country’s total trade to N6.24 billion, representing 27.30% in Q2, against the 27.46% level recorded in the corresponding quarter, and 27.46% year-on-year. In this episode of the Epigenetics Podcast, we caught up with Dr. Chuan He, John T. Wilson Distinguished Service Professor at the University of Chicago, to talk about his work on the influence of dynamic RNA methylation on gene expression. More than 150 structurally distinct post-transcriptional modifications of RNA have been identified at thousands of sites in cellular RNA molecules. Some RNA modifications are dynamic and may have critical regulatory roles analogous to modifications of protein and DNA. Understanding the scope and mechanisms of dynamic RNA modifications thus represents an emerging research frontier in biology and medicine. The internal N6-methyladenosine (m6A) modification in messenger RNA (mRNA) is one of the most abundant RNA modifications in eukaryotes. On average, this RNA base modification is present in approximately 3 sites in every mammalian mRNA molecule. The m6A modification has been shown to be critically important for cellular differentiation, animal development, and a range of biological signaling and stress responses. In 2011, Chuan He's lab reported that m6A in mammalian mRNA can be oxidatively demethylated in vitro and inside cells by the enzyme FTO (a fat mass and obesity-associated protein), a major obesity factor This discovery identified FTO as the first known RNA demethylase. Chuan He’s lab also subsequently identified ALKBH5 as the second RNA demethylase that controls mammalian spermatogenesis. More recent work in the He lab has focused on characterization of the YTH family proteins as the “reader” proteins that detect and bind methylated RNAs in mammalian cells to regulate their stability and translation. Functional characterizations revealed that YTHDF2 regulates cytoplasmic localization and mediates the decay of methylated mRNA, YTHDF1 promotes translation of methylated mRNA by facilitating translation initiation, and other methylated RNA readers also contribute to the regulation of mRNA storage, transport, and cellular localization.   Research in the He lab also identified a core protein complex comprised of two subunits: METTL3 and METTL14, and an accessory factor WTAP, which mediates cellular m6A RNA methylation. Cellular RNA methylation levels can have profound impacts on normal cellular differentiation and cancer cell proliferation. Their current work focuses on how the methylation selectivity is achieved. The Petroleum Products Pricing Regulatory Agency on Tuesday increased the ex-depot price of petrol by N6 to N138.62/litre for the month of August 2020.Oil marketers stated that based on the latest increase in ex-depot price, the pump price of Premium Motor Spirit, popularly called petrol, could be between N149/litre and N150/litre this month.The Vice President, Independent Petroleum Marketers Association of Nigeria, Abubakar Maigandi, told our correspondent that the PPPRA, however, did not tell marketers the new pump price for petrol.He said the Pump price might be announced today.Maigandi added that with the N6/litre increase in ex-depot price, the projection is that the pump price will be around N149/litre or N150/litre, based on our usual computations.”Learn more about your ad choices. Oil marketers under the aegis of the Major Oil Marketers Association of Nigeria have said access to the dollar at N306 will enable them to resume importation of Premium Motor Spirit (petrol). The marketers also said there was an urgent need for the Federal Government, through the Petroleum Products Pricing Regulatory Agency, to increase the margins on petrol to encourage them to make more investments. Forte Oil Plc will be getting a new name very soon. This is following the shareholders’ resolution to change the name of the oil company to Ardova Plc. The company made this disclosure in a notification sent to the Nigerian Stock Exchange (NSE), where it announced its forthcoming Extraordinary General Meeting (EGM), scheduled to hold on Tuesday, December 17, 2019. The Directors of the company were empowered to draw up, sign documents and appoint advisers regarding the matter where necessary. They were also expected to comply with the directives of regulatory authorities to make the name change possible. In addition, the shareholders intend to close the Register of Members and transfer of Books of the Company from 6th December 2019 to 15th December 2019 for the purpose of attending the meeting. Revenue: Forte Oil generated N123.54 billion in revenue as of the end of the nine-month period of 2019 compared to N94.81 billion in revenue as of September 2018. Also available on the ASM Podcast Network app. Julie’s Biggest Takeaways Citrus greening disease, or huanglongbing, is a disease of citrus trees causing a major epidemic among citrus farmers around the world. The disease causes trees to sicken and eventually die, and is best diagnosed by PCR amplification of the bacterial DNA from the bacterium that causes the disease, Liberibacter asiaticus. Because the disease spreads through the tree at different rates, it’s important that many samples be tested for accurate diagnosis. Quarantining the disease has proved difficult, as undiagnosed roots can transmit the disease if they are used to hybridize with canopy plants. The disease becomes even harder to contain under bad weather conditions: the high winds of recent hurricanes can scatter the insect vector, the Asian citrus psyllid, leading to infection of new orchards. Although L. asiaticuscan’t be cultured, Graciela performed a screen on L. asiaticustranscription factors that were produced by E. coli. These were tested for inhibition by a chemical library, and discovered that a common treatment for gout, benzbromarone, inhibited protein activity. This discovery was confirmed using in vivoinfected plants and by expressing the gene in related bacterial species, Graciela and her team predict the protein plays a role in responding to osmotic stress. The protein target of the chemical differs widely between citrus greening disease and gout, but the protein-chemical interaction is similar enough to allow protein inhibition. Is there a link between the microbiome and diabetes? 10 years ago, Lactobacillus johnsoniican rescue animals that are predisposed to diabetes. L. johnsoniiinactivates a host enzyme, IDO, which regulates proinflammatory responses. Activated immune cells can travel to the pancreas and attack beta cells, leading to diabetes. Nigeria Disbursed 679.7 Billion Naira to The Three Arms of Government in June

Federation Account Allocation Committee (FAAC) disbursed the sum of N679.7bn to the three tiers of government in June 2019 from the revenue generated in May 2019. The amount disbursed comprised of N571.73bn from the Statutory Account, N106.83bn from Valued Added Tax (VAT) and N1.41bn exchange gain differences. Federal Government received a total of N284.16bn from the N679.70bn. States received a total of N187.61bn and Local Governments received N140.10bn. The sum of N40.44bn was shared among the oil producing states as 13% derivation fund. Revenue generating agencies such as Nigeria Customs Service (NCS), Federal Inland Revenue Service (FIRS) and Department of Petroleum Resources (DPR) received N5.55bn, N11.03bn and N5.92bn respectively as cost of revenue collections.  Ahead of the deadline given by the National Insurance Commission (NAICOM) for insurance firms to recapitalize, Consolidated Hallmark Plc has revealed its plan to raise its capital base from N6.1 billion to N10 billion. It is seeking an additional N3.9 billion to meet the new capital requirement, after it's capital base has been increased from N3 billion to N10 billion. While maintaining that its first line of action would be to issue additional share by way of a rights issue, Consolidated Hallmark made known that it is important it shares its plans with shareholders and stakeholders to get their support to realize the new target. According to the insurance firm, the rights issue would give existing shareholders the opportunity to increase their stakes in the company and take the benefit of a lower offer price than that of a public offer. Cries for better services by thousands of jobless young people within the Amahlathi Municipality in the Eastern Cape have finally been heard. The municipality has been placed under provincial administration following months of violent protests which led to the torching of municipal buildings and a clinic. Officials within the municipality are accused of providing jobs in exchange of sexual favours, nepotism, fraud and corruption. The municipality, based in Stutterheim is situated along the N6 connecting East London and Johannesburg.

I Naimakka var det minus 39,3 grader i veckan, ett köldrekord för i år. Men är norra Norrland och södra Sverige som två olika världar? Vad är det mer än vädret som skiljer tycker du som bor i övre halvan? Vad fick dig att flytta till Norrland? Vad fick dig att flytta därifrån? Norrland utgör 59,6 procent av Sveriges yta och det är faktiskt 80 mil mellan Gävle och Kalix. Men bara omkring 12 procent av Sveriges befolkning bor i de nio norrländska landskapen, från Gästrikland i söder till Lappland i norr. Hur är livet i där du bor? Vad gör det med samhällsservice och livsvillkor när det är så glest befolkat? Vad är charmen med att bo i Offerdal, Abborträsk, Vittangi eller Sveg? Och vad är utmaningen? Norrlänningar framställs ofta som omoderna och bakåtsträvande människor som bor i en avfolkningsbygd. Coming up we have some inspiring research from the University of Alberta regarding viral pathogen treatment; McMaster researchers have a new hypothesis on Huntington’s Disease; RepliCel solidifies co-development deal in Greater China; scientists discover a way to genetically screen for acute myeloid leukemia; and putting Canada in the limelight when it comes down to artificial intelligence and deep learning.   Keep on listening to find out more of the details!  +++++  The University of Alberta appears to have hit the nail on the head yet again. With so much inspiring research coming out of this campus, it should come as no surprise that they have made a significant discovery that has the potential to treat viral pathogens such as the Zika virus and respiratory syncytial virus (RSV).  Scientists from UofA discovered a new and promising class of chemical compounds that is comparable to the naturally occurring isatisine A, an antiviral originally found in traditional Chinese herbal medicine.  Fred West, a professor in the Department of Chemistry, who led the discovery along with RSV researcher David Marchant, a professor in the Department of Medical Microbiology and Immunology, says, “This is both a remarkable scientific discovery and also something that has the potential to positively affect not only global health but also the economy of Canada.”   West and Marchant worked in conjunction with Zika expert and cell biologist Tob Hobman, who is also a professor in the Faculty of Medicine & Dentistry at the University of Alberta. They tested the compound against potent viruses, such as Zika and respiratory syncytial virus , that yielded remarkable results. The compound was active and effective against both viral infections.  The Zika virus, which is a mosquito-borne pathogen, began wreaking havoc in May of 2015 after it had been identified as the culprit behind an outbreak of prenatal defects across South America. It coincided with a 2,700 per cent increase in Brazil from reported cases of microcephaly, an often fatal congenital condition associated with incomplete brain development in newborns.  Whereas, relatively unheard of respiratory syncytial virus poses a greater threat to infants, the elderly and those with compromised immune systems. This virus is responsible for up to 30 per cent of hospitalized respiratory cases in any given year.   West adds, “What we aim to do is further refine this compound to keep the elements that make it medically active and build in the structural components that make it possible for patients to consume in drug form. We are approaching that point.”  The next stage of drug development is already underway, and with Marchant’s new company Antibiddes Technologies Inc. ready to license the intellectual property and begin commercialisation, this is a promising development that could change the face of viral pathogen treatment.  ++++++  McMaster researchers develop a new theory regarding Huntington’s Disease that may shape the future of drug development for the disease.  A team of researchers from the university found that a unique type of signalling found in damaged DNA signals huntingtin – a harmful mutant protein found in the genes of those diagnosed with Huntington’s Disease – to aid in DNA repair is defective for those with the condition.  The new hypothesis was published in the Proceedings of the National Academy of Science (PNAS).  Laura Bowie, a PhD student in the Department of Biochemistry and Biomedical Sciences at McMaster, says, “The concept was that if we applied the signalling molecule back in excess, even orally, this signalling can be restored in the Huntington’s disease mouse brain. The net result was that we fixed the modification of huntingtin not seen in mutant huntingtin in Huntington’s disease.”  Using this hypothesis, the study team discovered a molecule called N6-furfuryladenine, derived from the repair of DNA damage, which corrected the defect seen in mutant huntingtin.  “Based on dosing by different ways of this molecule in mouse Huntington’s disease models, Huntington’s disease symptoms were reversed,” says Bowie. “The mutant huntingtin protein levels were also restored to normal, which was a surprise to us.”  Ray Truant, senior author on the study, has dedicated his career to Huntington’s disease research and how mutation leads to Huntington’s disease. It was his lab that was the first to demonstrate that normal huntingtin was involved in DNA repair.  Truant argues that the traditional and controversial amyloid/protein misfolding hypothesis, where a group of proteins stick together forming brain deposits, is likely the result of the disease, rather than its cause.  He also stated that he considers this paper the most significant of his career, and that “This is an important new lead and a new hypothesis, but it is important for people to know this is not a drug or cure. This is the first new hypothesis for Huntington’s disease in 25 years that does not rely on the version of the amyloid hypothesis which has consistently failed in drug development for other diseases.”  Huntington’s Disease is an inherited, neurodegenerative illness that comes with dire physical, cognitive and emotional symptoms that often hit around middle age. The mutant huntingtin protein causes certain parts of the brain to die – specifically the caudate, the putamen and, as the disease progresses, the cerebral cortex.  Bev Heim-Myers, CEO of the Huntington Society of Canada adds “Innovative research initiatives, such as the work led by the team in Dr. Truant’s lab, including PhD student Laurie Bowie, has the potential to transform HD research. The answers we find for Huntington’s disease will likely lead to better understanding of treatments for other neurological diseases and it is important that we continue this cross-talk amongst neurodegenerative diseases.”  The study was conducted in partnership with the University of Alberta, Western University, Johns Hopkins University, and a collaboration with a U.S. biotech firm, Mitokinin LLC. Their work now continues in developing better derivatives of N6-furfuryladenine towards developing a drug.  This study was funded by the Canadian Institutes of Health Research, the Krembil Foundation, and the Huntington Society of Canada.  +++++  Vancouver-based Replicel Life Sciences announces that they have signed definitive agreements with YOFOTO, a Chinese company, solidifying their partnership to commercialise three of Replicel’s programs in Greater China.    The collaboration focuses on the development and commercialization in Greater China of RepliCel’s tendon regeneration cell therapy (RCT-01), skin rejuvenation cell therapy (RCS-01), and its injection technology in development for dermal applications (RCI-02) (excluding hair-related treatments).  YOFOTO Chairman states, “YOFOTO is a fast-growing company built on values related to beauty and healthy, active lifestyles. As a key part of developing and commercializing products related to these core values for our consumers, we are committed to being a leader in China in the commercialization of regenerative medicines.  The RepliCel cell therapy and injection technologies focused on skin rejuvenation and tendon repair are important building blocks in YOFOTO’s strategic healthcare vision. We are pleased to have structured a deal with RepliCel which results in YOFOTO not only being a development partner and commercial licensee but also an investor committed to contributing to RepliCel’s global success.”  YOFOTO’s investment in Replicel will include milestone payments, minimum program funding commitments, and sales royalties in exchange for an exclusive 15-year license to three of RepliCel products for Greater China (Mainland China, Hong Kong, Macau, and Taiwan). Additionally, YOFOTO commits to spending a minimum of C$7 million on Replicel’s programs and associated cell processing manufacturing facility over the next five years in Greater China.  RepliCel president & CEO, Lee Buckler says, “In 2017 RepliCel delivered successful phase I data in all three of its cell therapy programs and functioning prototypes of its next-generation dermal injector,” says “We were committed to delivering a landmark partnership to RepliCel shareholders in 2018. The partnership with YOFOTO represents such a deal and provides RepliCel with not only an outstanding partner in Greater China but capital to move our programs forward in Europe and North America,” he adds.  The deposit of over $5 million has been paid by YOFOTO, but remains in escrow following the closing of the transaction. Once YOFOTO has met certain conditions and once relevant Chinese patents are issued in China, they will be assigned a YOFOTO-owned Canadian subsidiary.  This is very exciting news for Replicel and Canada alike, as Canadian biotech companies begin to spread across the Pacific, with more foreign companies seeking to invest in Canadian innovation and research.  +++++  An international team of scientists discovers a technique that predicts healthy individuals who are at risk of developing acute myeloid leukemia (AML), which is an aggressive and often deadly form of blood cancer.  The findings, published in Nature, illuminate the ‘black box of leukemia’ and answer the question of where, when and how the disease begins, says co-principal investigator Dr. John Dick, Senior Scientist at Princess Margaret Cancer Centre, University Health Network.  Dr. Dick, who is a professor, Department of Molecular Genetics, University of Toronto, holds the Canada Research Chair in Stem Cell Biology, and is co-leader of the Acute Leukemia Translational Research Initiative at the Ontario Institute for Cancer Research, says, “We have been able to identify people in the general population who have traces of mutations in their blood that represent the first steps in how normal blood cells begin on a pathway of becoming increasingly abnormal and puts them at risk of progressing to acute myeloid leukemia. We can find these traces up to 10 years before acute myeloid leukemia actually develops. This long-time window gives us the first opportunity to think about how to prevent acute myeloid leukemia.”  Study author Dr. Sagi Abelson, a post-doctoral fellow in the Dick lab, says: “acute myeloid leukemia is a devastating disease diagnosed too late, with a 90 per cent mortality rate after the age of 65. Our findings show it is possible to identify individuals in the general population who are at high risk of developing acute myeloid leukemia through a genetic test on a blood sample. The ultimate goal is to identify these individuals and study how we can target the mutated blood cells long before the disease actually begins.”  The study stems from Dr. Dick’s 2014 discovery that a pre-leukemic stem cell could be found hiding amongst all the leukemia cells that are present in the blood sample taken when a person is first diagnosed with acute myeloid leukemia. The pre-leukemic stem cell still functions normally but it has taken the first step in generating pathway of cells that became more and more abnormal resulting in acute myeloid leukemia (Nature, February 12, 2014).  Dr. Dick says, “Our 2014 study predicted that people with early mutations in their blood stem cells, long before the disease appears and makes them sick, should be able to be detected within the general population by testing a blood sample for the presence of the mutation.”  The team extracted the data from more than 100 participants who developed acute myeloid leukemia six to 10 years after joining the study, plus the data from an age-matched cohort of more than 400 who did not develop the disease.  Dick adds, “We wanted to know if there was any difference between these two groups in the genetics of their ‘normal’ blood samples taken at enrollment. To find out, we developed a gene sequencing tool that captured the most common genes that get altered in acute myeloid leukemia and sequenced all the 500 blood samples.”  The gene sequencing tool was a success and picked up mutations years before an individual was diagnosed with acute myeloid leukemia to accurately predict those at risk. Moreover, the team used advanced computational technology to assay the information obtained from routinely collected blood tests taken over 15 years in Israel and housed in a massive database of 3.4 million electronic health records.  The study has linked acute myeloid leukemia with a common feature of aging called ARCH-age related clonal hematopoiesis, whereby blood stem cells acquire mutations and become a little more proliferative. The majority of people that have ARCH will not develop acute myeloid leukemia. It is a requirement to have acute myeloid leukemia, but not the other way around.  +++++  Machine learning is augmenting human ability and drastically changing possibilities. The growth and brilliant minds from around the country have led the way for Canada to place its mark with AI on the world and build a more promising ecosystem for the future.  Across the country, there have been several companies lending a hand in this newer and multi-faceted industry that will reshape history. Among those, innovative researchers are developing imaging devices for skin cancer, diagnostic platforms that analyse natural speech to detect dementia and mental decline, advanced signal analysis to diagnose coronary artery disease, just to name a few, and numerous others that stem to topics unrelated to health care, but keep Canada at the forefront of change.  This year Canada had its 31st Canadian Conference in Artificial Intelligence that stands to show how long Canada has been involved with this game-changing technology. Events like this bring together hundreds of leaders in research, industry, and government that provide a melting pot of inquisitive and like-minded people.  Life sciences is an industry that is filled with innovation and offers much economic promise for governments who desire to grow a knowledge-based economy. Therefore, Biotechnology Focus, in support of Global Biotech Week, is conducting a survey to gauge Canada’s innovation culture. Last month, live on Facebook, self-styled biohacker, Tristan Roberts injected himself with a gene called N6. He hopes it will help his body produce an antibody that fights the virus that causes AIDS. But not everyone is on board. Why? Because it's untested and unregulated. Roberts is part of a growing community of bio-hackers. They believe they can get medical cures to the market, faster and cheaper than others held back by regulations. But many in the medical community are outraged. They call Robert's behaviour delusional, risky and frightening.
