Dr. Pol Boudes, CMO of Rectify Pharmaceutical, highlights the importance of membrane proteins, specifically the role transporters play in how cells interact with their environment. Rectify is working on developing positive functional modulators (PFM) to address diseases related to dysfunctional transporters the first being primary sclerosing cholangitis, a rare liver disease with no current treatment options. The PFM can potentially restore the normal function of transporters involved in bile composition and secretion, which are key mechanisms in PSC.   Pol explains, "So you have different types of membrane proteins, but what we're working with are called transporters, so they're proteins that are based on the membrane but channel components out and in the cells. So basically, it's a way for the cell to interact with its environment, and you have multiple types of transporter proteins. We're specifically working or started to work with one group of family of proteins that are called the ABC transporters. They're a very important function, and they can either be disrupted because there is a mutation in the protein, so it's a genetic disease, or they are potentially disrupted because there is just a functional deficit of this transporter. The protein is normal, but it's not functioning properly. So it's a functional deficit of what we call the wildtype protein." "So with the PFMs, what we're doing is to use small molecules that you can administer orally, and they bind to the transporter, and by doing so, they can modify the three-dimensional structure of the transporter and consequently the way this protein behaves within the cellular environment. So we correct the function of the transporter by changing the size, if you want, of the shape of the transporter. So we started to focus on this type of proteins and you have many potential diseases due to transporter deficit."  "We're focusing on one disease, liver disease. That's our lead PFM for a disease called primary sclerosing cholangitis, which is a disease of the liver. This very severe liver disease is also a disease that has no treatment available, which is a little bit frustrating because the progression of this disease leads to liver cirrhosis. And the only thing you can do at this stage is liver transplantation. As you might know, liver transplantation is problematic because it's a very complex process, it's also very expensive, and unfortunately, there is a shortage of transplants. The thing is – these were primarily black-box technologies that seemed very abstract from daily life for consumers and for the teams of developers at banks tasked with managing them. ChatGPT changed that and with a slight change in interface, it's unlocked a lot of use cases for financial services. We're early for sure in this evolution but companies that have been studying the space are already beginning to make a move. In this episode of The Challengers, Josh Liggett and Zack Miller explore the pandemic's impact on incumbent banks as well as on challengers. Zack mentioned fears are high and that's creating rumors that certain challengers are failing. Josh believes it's a good time for incumbents to really get focused around digital. They have no other choice. Next, the hosts discuss OakNorth, which is still Europe's fastest growing startup. But growth of its loan book is slowing, whether by design or by market forces. The bank is profitable -- perhaps the only one of many challengers to reach this milestone. Square gets a license and Zack believes that we'll see Square move strongly into lending to merchants using its technology. Like PayPal, Amazon, and Shopify, this captured lending is powerful and makes a ton of sense from a service model perspective. It can also be quite lucrative. Credit Sesame, like Credit Karma a few months before it, is rolling out a banking product, Sesame Cash. PFMs want to own the money movement piece for their customers. Lastly, we mention Bill Harris' new firm, One, which just launched in private beta after a $17 million funding round. Harris was CEO at PayPal and Intuit, and founded Personal Capital before this. Sharing clean data between banks and apps may be somewhat of a boring business but it’s an important one. This is a very active area of fintech and this should help to propel things forward. In this presentation you will be introduced to a relatively new material called IPS e.max CAD. Unlike traditional PFM or zirconia-based restorations, IPS e.max CAD is milled from a single block that maintains a consistent shade throughout. With this material, there is no need to opaque out the dark metal substructure found in PFMs or keep the bright white zirconia substructure from bleeding through the ceramic. This presentation discusses the IPS e.max process used by the laboratory. It also features eight clinical cases showing how IPS e.max may be used for a variety of clinical needs.
