Abstract: As signalling organelles, primary cilia regulate their membrane G protein-coupled receptor (GPCR) content by ectocytosis, a process requiring localised actin dynamics at their tip to alter membrane shape.(1, 2) Mammalian photoreceptor outer segments comprise an expanse of folded membranes (discs) at the tip of highly-specialised connecting cilia (CC), in which photosensitive GPCRs like rhodopsin are concentrated. In an extraordinary feat of biology, outer segment discs are shed and remade daily.(3) Defects in this process, due to genetic mutations, cause retinitis pigmentosa (RP), an untreatable, blinding disease. The mechanism by which photoreceptor cilia generate outer segments is therefore fundamental for vision yet poorly understood. Here, we show the membrane deformation required for outer segment disc genesis is driven by dynamic changes in the actin cytoskeleton in a process akin to ectocytosis. Further, we show RPGR, a leading causal RP gene, regulates activity of actin binding proteins crucial to this process. Disc genesis is compromised in Rpgr mouse models, slowing the actin dynamics required for timely disc formation, leading to aborted membrane shedding as ectosome-like vesicles, photoreceptor death and visual loss. Manipulation of actin dynamics partially rescues the phenotype, suggesting this pathway could be targeted therapeutically. Wow! This episode delivers on many different levels from a history, artifact point of view. Gene is a wealth of knowledge when is comes to Bowhunting history and honestly this chat didn't even begin to scratch the surface. We talked about how for some hunters they are skipping the "romance" portion of being a hunter. The part where you become curious about the history, curious about your weapon, or maybe the landscape of where you are hunting. Curious as to who was doing it before you? How long ago? With what? And the most important the, why? Way before social media and "hero" shots, what drew them to the woods or mountains. Was it food? Maybe in some cases, but let's be honest. Food isn't the reason we hunt, I mean It may be "A" reason, but not "THE" reason. There's a much deeper interest than that for most hunters, I'm sure of itGene is one of those guys who in his 60 plus years of being arguably one of the greatest Whitetail Bowhunters of all time, he ask those questions. Then he seeked those with answers out- and later shared their stories. I'm fortunate enough to be one of the people Gene shared those stories with. Now I'm bringing them to you. This month, a whitetail hunting icon on the Jay's Sporting Goods Podcast....Barry Wensel. Barry has been chasing mature bucks with traditional gear for more than fifty years. He has great advice on targeting trophy bucks and says hunting them is different than other deer. Wensel talks about patterning big bucks and has tips on where to find them.

Meet April Wensel, founder of Compassionate Coding, which provides training curriculum for technical teams to build supportive, diverse, and inclusive cultures, implement better code review practices, and foster better communication between engineers and other employees. Wensel’s love of computer games during her youth led her to software engineering in college and her career. After attending Pomona College, she moved to Silicon Valley to work in the software development world for companies such as Sony, edtech startup Zoodles, AI startup Cognoa, and others. After witnessing a multitude of company cultures, she discovered how companies were ignoring empathy and only speaking to a specific type of engineer. With this in mind, she founded Compassionate Coding to effect positive change in a company’s culture. At the Tacos and Tech event in Mountain View in 2017, Wensel met a lot of San Diego companies who were very open to speaking about their culture and diversity. San Diego based April Wensel is the founder of Compassionate Coding. After a decade in software, she noticed that there was a lot of suffering in the industry. Rather than just assuming that that can't be changed, Wensel decided compassion was the answer. Phil Burgess introduces his guest April Wensel by telling us April is an international speaker and the founder of Compassionate Coding a social enterprise bringing emotional intelligence and social consciousness to the software industry. April is also a mentor and a keen advocate of diversity and inclusiveness in the industry. Phil agrees that identifying your core values is essential and a very good point and that if you establish what they are early, and what you want to do with your career as you mature, opinions will change but they are essential if you want a career in IT. Phil enquires of April about her worst IT moments and learning from that experience. April tells us about a job she loved and how she was upset on ethical grounds regarding how the company was progressing, she also tells us what happened to her. Phil shares with April an interview he did with David J Anderson ,and David made the point April was making, that in your career you have to make the choices between your job and what is ethically correct. Phil asks April about her career highlight or greatest success. April outlines how her view of compassionate coding spreading through the technical community has given her personal satisfaction. April had seen the lack of concern in human beings and knew she wanted to do something on her own. When she receives messages from people implementing or taking on board her ethics she says it light’s her up inside. April informs us that if we are looking to have a career in IT we need to have more that coding skills. We need to be able to manage our time well, able to communicate our ideas in an effective way, and work well with others. April assures us that the industry is not as it always was perhaps being shut away in a basement on our own, but that IT is now in this modern landscape and communication skills are going to become more important. BEST MOMENTS April tells us “I’ve matured I am not excited by technology for technology’s sake, for me the most exciting is social entrepreneurs in technology companies April explains “we are able to apply all the techniques established and apply them to social problems” Phil asks April about non technical skills. April believes listening is a very important skill to have in the industry especially when dealing with non-technical staff. “Ignore any advice that does not resonate with your core values “April says when Phil asks her to share any parting piece of advice April discusses what happened when she had to leave the company she loved working for over ethical issues. April says it was a problem with the culture of the company and its ethics.
