As a result, many HTT-lowering therapies are being pursued in clinical studies, including those that reduce HTT RNA and protein expression in the liver. To investigate potential impacts, we characterized molecular, cellular, and metabolic impacts of chronic HTT lowering in mouse hepatocytes. Lifelong hepatocyte HTT loss is associated with multiple physiological changes, including increased circulating bile acids, cholesterol and urea, hypoglycemia, and impaired adhesion. HTT loss causes a clear shift in the normal zonal patterns of liver gene expression, such that pericentral gene expression is reduced. These alterations in liver zonation in livers lacking HTT are observed at the transcriptional, histological and plasma metabolite level. We have extended these phenotypes physiologically with a metabolic challenge of acetaminophen, for which the HTT loss results in toxicity resistance. Our data reveal an unexpected role for HTT in regulating hepatic zonation, and we find that loss of HTT in hepatocytes mimics the phenotypes caused by impaired hepatic {beta}-catenin function. Back to the medieval! We talk about Saint Francis of Assisi and his Canticle of the Creatures, one of the first examples of written Italian. Anne once fantasized about a sitcom based on his life called Everybody Loves Francis! (Mostly just for the title.) And then take a trip to his hometown of Assisi in Umbria: how to get around, what to see, where to stay, and (as always!) what to eat! Pax et bonum! Pace e bene! Peace and Goodness!Patron saint of animals and ecology as well as the country of Italy (along with Catherine of Siena), Francis did quite a bit of traveling, preaching as he went. Yet he always came back to his hometown of Assisi. Well worth a visit today not only for its connection to its famous native son, Assisi is a marvelous example of a medieval Italian hilltop town. Francis is buried in the lower basilica of San Francesco in Assisi, where you can also admire the frescoes attributed to Giotto in the upper church that depict Francis's life and miracles. Also in the historic center of Assisi you can visit the Basilica of Santa Chiara, which contains the saint's crypt as well as the crucifix, which, according to the legend, first called Francis to "repair my church."As we mention in the episode, Francis died in the lower part of the town of Assisi, in this church of the Porziuncola, today enclosed by the Basilica of Santa Maria degli Angeli. We also talked about hiking up to the Eremo delle Carceri, a lovely place in the woods where Francis and his followers would retreat for solitude and prayer.And we also talked a lot about eating! A great place to eat (and stay) about a kilometer outside of the city walls is the Ristorante La Stalla at Fontemaggio, which, as the name suggests, is housed in a converted stall. You can try local favorites like pasta or meats cooked over an indoor enormous grill. Abstract: Progressive striatal gene expression changes and epigenetic alterations are a prominent feature of Huntingtons disease (HD), but direct relationships between the huntingtin (HTT) protein and chromatin remain poorly described. Here, using chromatin immunoprecipitation and sequencing (ChIP-seq), we show that HTT reproducibly occupies specific locations in the mouse genome, including thousands of genomic loci that are differentially occupied in striatal tissue from a knock-in mouse model of HD (B6.HttQ111/+) versus wildtype controls. ChIP-seq of histone modifications, generated in parallel, revealed genotype-specific colocalization of HTT with trimethylation of histone 3 lysine 27 (H3K27me3), a repressive chromatin mark. Close to genes that are differentially regulated in HD, greater HTT occupancy in HttQ111/+ vs. wildtype mice predicted increased H3K27me3, reduced histone 3 lysine 4 (H3K4me3, a marker of poised and active promoters), and down-regulated gene expression. Chris Cantle is one of those rare journalists who not only does everything but is good at everything. His career, first with Cycle World then Road & Track and now The Drive, has been filled with incredible adventures and we, as readers, have been lucky enough to vicariously take part with him. He took us along when he raced a 300+ horsepower jet-ski from Long Beach to Catalina and when he attempted to scientifically determine whether wearing one's facial hair in the style of Graham Hill would make one faster around a race track. This was Chris' first visit to CarStories but we had a blast and we can't wait to have him back!
The concept of “community cohesion” rose to prominence in the detritus of Bradford and Harehills, Burnley and Oldham, Northern English towns where 14 years ago rioting broke out between Asian and white communities. Called on by the Home Office to investigate the roots of the riots, sociologist Ted Cantle – until then the chief executive of Nottingham City Council for more than a decade and before that director of housing in Leicester City Council –led an investigation that produced Community Cohesion: The Report of The Independent Review Team, a document better known now as the Cantle Report. The report introduced two terms into the public conversation, “parallel lives” to describe how communities could exists side by side and yet in mutual exclusion and incomprehension, and “community cohesion,” which in its most general sense is the idea of not living parallel lives. In this Social Science Bites podcast, David Edmonds discusses one key component of parallel lives – segregation – that prevents cohesion. “[P]eople who lived in these parallel lives,” Cantle explains, “had no understanding of the other, they could easily be dealing with prejudices and stereotypes, they had no opportunities to disconfirm them, they had no opportunity to really challenge their own race’s views, or their own views about another faith.” In the podcast, Cantle adds that approaching these issues from several perspectives, specifically through different disciplines, leads to a better understanding on the underlying dynamic than any 'siloed' approach.
