I'm Stacey Simms and every other Friday I bring you a short episode with the top diabetes stories and headlines happening now. XX Tandem gets FDA clearance for a new infusion set. The company shared in an SEC filing that its Capillary Biomedical (CapBio) subsidiary won clearance for its SteadiSet Infusion Set. SteadiSet features an integrated inserter with a hidden needle designed for one-handed insertion. It received clearance to deliver insulin for up to three days of use. However, Tandem plans to submit a separate request to extend the indicated use time to up to seven days. It doesn't expect to begin commercial activities for SteadiSet until after it receives clearance for the extended use time. Many of us have been following this since Tandem acquired CapBio back in 2022.. it's been pretty hush hush – CapBio has never accepted my requests for interviews, so I'm excited to see this approval, mostly because my son really really needs it! https://www.drugdeliverybusiness.com/tandem-capbio-fda-clearance-insulin-infusion-set/ XX Insulet just announced via an email to health care providers that Dexcom G7 compatibility with the Omnipod 5 iPhone app will be released IN THE COMING WEEKS! Great news for people with #t1d and XX Sernova announces positive interim data from its ongoing Phase 1/2 clinical trial in patients with T1D. this is their Cell Pouch Bio-hybrid Organ as a functional cure for type 1 diabetes (T1D), today Interim data from 12 patients with transplanted human donor islet cells show patients achieving insulin independence, islet cell engraftment in Cell Pouch, islet function, islet survival, improved glycemic control, improved patient reported quality of life (QOL) and improved awareness of hypoglycemia and increased sensitivity to severe hypoglycemic symptoms. Improvement in patient outcomes was correlated with a cumulative increase in the quantity of transplanted islets. Based on these findings, the study is on track to meet its primary and secondary endpoints, and the confirmatory Cohort C is expected to initiate in H2 2025. Clinical trials with iPSC islet-like clusters from Sernova's partner, Evotec, are anticipated to begin in 2026 after completion of Cohort C of the ongoing trial. These interim findings, with 8 of 12 patients achieving insulin independence so far, support the thesis that Sernova's high volume ten channel Cell Pouch, used in Cohort B, plus an optimised immune suppression regimen, has the potential to achieve insulin independence, without portal vein transplant, in our planned clinical trial with Evotec's high quality iPSC islet-like clusters. Measured HbA1c (a blood test that shows the average blood sugar levels over the past 2-3 months) in patients with Cell Pouch alone, showed that 9 of 12 patients had a reduced value within the American Diabetes Association (ADA) recommended range of
Riding Shotgun With Charlie #229 John Turrell Armed Lifestyle Magazine There are some nice perks about going to SHOT Show as media and having more than one media gig. After I was asked to write for Armed Lifestyle Magazine, I got to meet the other contributors at SHOT. One of the ALM writers is John Turrell. John, as you'll hear, is from England. This is my second international passenger! I'm glad we were able to connect with a crazy busy SHOT schedule. Not just mine, he keeps busy in Vegas, too. And not with the night life. With writing! He and his team put out daily reviews from SHOT. Like many folks, John started shooting as a young child, around 10 years old, with his father. He's worked his way up to competitive shooting in UKPSA and IPSC. UKPSA is the United Kingdom Practical Shooting Association. He enjoys shooting pistols competitively and even went up against Jerry Miculek, who got him into shooting revolvers. He is currently the sales manager at a gun range, where he offers a shooting experience for new folks. The hopes are to turn them into licensed individuals who get into firearms and shooting. The range is unique particularly in the UK.They have an indoor range with turning targets. With the range shooting experience, people can get a safety briefing, how to shoot session, then fire 50 rounds, ending with a little fun competition with turning targets. The range is very much like an indoor range here in the US. In 1996, there was a mass shooting. The government decided that it was time to ban all pistols from the “subjects”. There are several limitations…ahem…infringements. The barrel length must be 12 inches long and the overall length has to be at least 24 inches. That sounds like a rifle to me rather than a pistol. Obviously, that makes it hard to carry on one's body, which the government doesn't want them to do. To get a license in the UK, you have to attend a class, join a range, then you have a probationary period of at least three months. Then you apply to your local country police force for the license, with the usual background check and references. Of course, you're not guaranteed to get the license either. If all goes well, you get a certificate. There are two types of certificates. The section one certificate is for centerfire rifles and long barreled pistols and shotguns with a more than three round capacity. The type of action doesn't matter, just the capacity. The section two license is for shotguns which hold two rounds in the magazine and one in the chamber. Despite the limitations, John enjoys having to do all the reloading. Seeing some Mossberg's at SHOT, he'd really love to have a 590, but due to the barrel length, he wouldn't be able to get home. We also talk about suppressors. As I've learned the original name was a silencer, but in the UK, they call them moderators. First, in order to purchase one, it has to be on your license that you can have them. To have it added to your license, it's only about $25. At the gun store, you tell them which one you'd like, then add the serial number to the license, you pay for it, and then go home with it. Boom. Easy! The main reason these are legal is for hearing protection. There is a possibility that the moderators will become an item that they don't keep track of on your license. However, if you buy one for your air rifle, no license information is required. He says it could be possible that it gets as easy as going to the store to buy groceries and then picking up a moderator like it's any other item. Very different than it is here. I really enjoyed getting to know John better. He's a great guy with a lot of knowledge. While at SHOT, he and his team crank out daily reports of the guns and gear they see.So he's there truly for work and not necessarily pleasure. The study investigates the K210del mutation in genetic cardiomyopathy, which impairs myocardial contractility due to calcium discoordination in the troponin complex. Using a structure-based drug repurposing approach, researchers identified risedronate, an FDA-approved bisphosphonate, as a structural corrector that restores the calcium-binding domain's normal configuration. Experiments with patient-derived iPSC cardiomyocytes and a mouse model demonstrated that risedronate normalized calcium sensitivity, improved contraction velocity, and restored left ventricular function. The findings highlight the potential of repurposing existing drugs to treat genetic cardiomyopathies.

Looking to the future, Jorge envisioned iPSCs becoming a cornerstone of standard medical practice, while acknowledging the need to address safety, scalability, and regulatory hurdles to fully realize their potential. What are Induced Pluripotent Stem Cells (iPSCs)? "Induced pluripotent stem cells are a type of stem cell created by reprogramming adult somatic cells, like skin or blood cells, back into an embryonic-like state," explains Jorge. This process involves introducing specific transcription factors, often called Yamanaka factors, to transform these cells into a versatile state. Once reprogrammed, iPSCs can differentiate into almost any cell type, making them invaluable tools for research, drug development, and potentially life-changing therapies. The Growing Importance of iPSCs iPSCs offer a range of advantages, particularly their ability to sidestep ethical concerns tied to embryonic stem cell use. “What makes iPSCs so important today,” Jorge notes, “is their versatility and potential applications. Researchers can create patient-specific cell lines, which are essential for drug screening, disease modeling, and personalized medicine.” This technology is pivotal for regenerative medicine, offering hope for repairing damaged tissues and organs. “From neurodegenerative diseases to heart damage, iPSCs open the door to innovative treatment possibilities,” he adds. Mastering the Production Process Producing iPSCs is a meticulous endeavor. "Consistency is key," emphasizes Jorge. Researchers must ensure that each batch of cells meets strict criteria to avoid unpredictable outcomes, especially when precision is vital in both research and therapeutic applications. Standardized protocols and quality control measures are essential to achieve consistency. These involve monitoring for contamination and verifying the cells' ability to differentiate into various cell types. “Imagine developing a therapy based on a specific batch of cells, only to find that subsequent batches behave differently,” he warns. “Such inconsistencies can jeopardize patient outcomes.” Tackling Challenges in Culturing iPSCs Culturing iPSCs presents its own set of challenges. High cell numbers are often needed for large-scale research or therapeutic applications, but scaling up production without compromising quality is no small feat. Maintaining the cells' pluripotent state is another hurdle, as they can easily differentiate prematurely under certain culture conditions. "Environmental parameters like temperature, pH, oxygen levels, and nutrient availability must be rigorously controlled," Jorge explains. “Even minor fluctuations can negatively impact cell health and their ability to remain pluripotent.” Innovations Addressing Culturing Hurdles To overcome these challenges, researchers are turning to advanced techniques like 3D culture systems and bioreactors.
