After replacing the rail, rings, and checking the scope, a gunsmith finally discovered the rifle's mounting holes were drilled at a 45° angle from the factory. The manufacturing defect that made proper alignment impossible.)Continuation of the story from where it cut off last week- We looked in there and about half the threaded hole in the receiver was visible through the rail. Well shit… We tried a different rail and it was the same. I decided to contact Bergara to see if they would fix it. I started a warranty case with them and they sent an RMA form and asked me to ship the rifle to them. I sent it over and assumed that everything would be straightened out. After several weeks with zero updates, I called and asked for some kind of information. I even gave them exact instructions on how to replicate the problem. They said they would update me when they got it into their system and when they got started on the inspections. After they had it for a month they finally shipped it back and their conclusion was that the rifle was in spec. About a million emails later and both sides being stubborn, I gave up. Obviously they weren't going to fix it and I was at my wit's end. I brought it back to the gunsmith and had him drill and re tap the receiver holes to a big enough size to line it up. In spec my ass. Ever since then the Bergara name has left a bad taste in my mouth. I'm not really on social media and don't know how to get through how bad of an experience I had with them because I think people need to know. I am sorry for the long story but I already left a lot of details out. Researchers Identify Key Signaling Pathway Driving Muscle Cell Aging

BUFFALO, NY — June 23, 2026 — A new #research paper was #published in Volume 18 of Aging on May 28, 2026, titled “p38MAP kinase regulates senescence in human iPS-derived myocytes.” The study was led by first author Hiroki Sato and corresponding author Ryuichiro Sato from The University of Tokyo, along with corresponding author Makoto Shimizu from The University of Tokyo and Ochanomizu University. As people age, skeletal muscle gradually loses strength, mass, and function, contributing to frailty, reduced mobility, and an increased risk of falls. Although cellular senescence is widely recognized as a major driver of aging, the mechanisms that promote aging within mature muscle fibers remain poorly understood. In this study, researchers developed a human cell-based model to investigate how senescence develops in skeletal muscle cells and identified a signaling pathway that appears to play a central role in the process. The research team used human induced pluripotent stem cell (iPSC)-derived myocytes, specialized muscle cells generated from stem cells. To mimic age-related cellular damage, the investigators exposed the cells to low-dose X-ray irradiation, which induced DNA damage without causing extensive cell death. The treated muscle cells developed several characteristics commonly associated with aging. They exhibited muscle fiber atrophy, reduced contractile activity, and increased expression of p21, a well-established marker of cellular senescence. The researchers also observed elevated production of senescence-associated secretory phenotype (SASP) factors, inflammatory and extracellular signaling molecules that are commonly released by senescent cells. To better understand the molecular changes involved, the investigators performed proteomic, transcriptomic, and functional analyses. These studies revealed activation of p38 mitogen-activated protein kinase (p38MAPK), a signaling pathway previously linked to cellular stress responses and aging in other tissues. Further experiments demonstrated that inhibiting p38MAPK partially attenuated several senescence-associated features in the muscle cells. Treatment with p38MAPK inhibitors reduced muscle fiber atrophy, improved contractile function, and suppressed the expression of several senescence-associated factors. Conversely, activating p38MAPK promoted aging-like changes in the cells, further supporting its role in regulating muscle cell senescence. The study also identified activation of an integrin–FAK/SRC–p38MAPK signaling axis following DNA damage. According to the authors, this pathway may help explain how cellular stress is translated into long-term functional decline in skeletal muscle. Importantly, the findings were supported by analyses of human muscle aging datasets, which showed increased activity of MAPK signaling, focal adhesion pathways, and cytokine-related signaling in older skeletal muscle tissue. Dr. Howard Federoff, Scientific Co-Founder, Chief Medical Officer, and Executive Vice President of Corporate Medicine and Science at Kenai Therapeutics, is developing a cell therapy for Parkinson's disease that involves transplanting IPSCs into the brain to replace lost dopamine neurons.  The goal is not to cure the underlying cause of the disease, but to restore motor function and reverse the disease's progression. Initial clinical focus was on patients with moderate to moderate-severe idiopathic Parkinson's, showing efficacy and safety, leading to expanding the study to include earlier-stage and familial forms of the disease. Howard explains, "Kenai was formed about three and a half years ago, and the intention is to develop a cellular product, which means that what is manufactured will be eligible in the right patient groups who have a diagnosis of Parkinson's to be placed into the brain. And consequent to its placement, the cells will then form new circuits, dopamine circuits that are lost owing to the disease diagnosis that will have occurred years earlier." "The induced pluripotent stem cell approach is one we favor for several reasons. The nature of what then becomes the IPSC, as we like to call it, starts with a normal human volunteer whose medical history is very detailed and does not contain any familial personal history of neurodegeneration. That cell then undergoes a process called reprogramming. And in our case, it's done with a slightly different approach than many others. And the reprogramming effectively creates a cell that is pluripotent, meaning it can become any cell of the body. Initial clinical focus was on patients with moderate to moderate-severe idiopathic Parkinson's, showing efficacy and safety, leading to expanding the study to include earlier-stage and familial forms of the disease. Howard explains, "Kenai was formed about three and a half years ago, and the intention is to develop a cellular product, which means that what is manufactured will be eligible in the right patient groups who have a diagnosis of Parkinson's to be placed into the brain. And consequent to its placement, the cells will then form new circuits, dopamine circuits that are lost owing to the disease diagnosis that will have occurred years earlier." "The induced pluripotent stem cell approach is one we favor for several reasons. The nature of what then becomes the IPSC, as we like to call it, starts with a normal human volunteer whose medical history is very detailed and does not contain any familial personal history of neurodegeneration. That cell then undergoes a process called reprogramming. And in our case, it's done with a slightly different approach than many others. And the reprogramming effectively creates a cell that is pluripotent, meaning it can become any cell of the body. The National Institute of Environmental Health Sciences (NIEHS) Superfund Research Program (SRP) is hosting a Risk e-Learning webinar series focused on the use of innovative, human-relevant technologies to better characterize the biological effects of chemicals. New technologies, including advanced cell-based assays, organoids, and computational modeling approaches, are expanding the toolbox researchers use to answer previously difficult or unanswerable questions. Presenters will discuss how these emerging methodologies are being applied to uncover mechanistic insights, improve predictive accuracy for human health outcomes, and refine risk assessment frameworks. The third and final session, titled Innovative Methods for Understanding Chemical Toxicity, will feature three speakers discussing innovative approaches to understanding the dose at which chemicals trigger biological responses and the mechanisms behind them. Speakers include:Ana Maretti Garcia, Ph.D., University of Southern California: In this presentation, Dr. Maretti Garcia will discuss recent advances using advanced 3D human liver spheroid models to characterize the biological effects of multiple per- and polyfluoroalkyl substances (PFAS) on human liver metabolism and their contribution to metabolic dysfunction-associated steatotic liver disease (MASLD). She will highlight key findings recently published in Environment International (PMID: 40914107) and Communications Medicine (PMID: 41162609), focusing on molecular and cellular mechanisms identified through the integration of in vitro models with human-relevant data.Guru Ulaganathan, Duke University: Developmental neurotoxicity (DNT) is challenging to study due to the intricacy of human brain development, the dependence on the timing of exposure, and the emergence of effects long after the initial exposure. This presentation will discuss advances in human iPSC modeling using three-dimensional brain models that can be leveraged to mechanistically investigate neurodevelopmental perturbations at both a molecular and functional level.Weihseuh A. Chiu, Ph.D., Texas A&M University: Chemical contamination after disasters presents a plethora of challenges to risk assessment and risk management. Dr. Chiu will present several case studies from the Texas A&M Superfund Research Center of new approach methodologies (NAMs) to address this challenge by rapidly characterizing hazards and risks from individual chemicals and environmental mixtures. Lessons learned from these studies inform a general strategy for NAMs-based solutions for next generation risk assessment. Why is a broad approach that works with iPSC-derived cardiomyocytes so important to understanding molecular mechanisms underlying calcium homeostasis and arrhythmias? In this episode, Associate Editor Dr. Petra Kleinbongard (University of Duisburg-Essen) interviews author Dr. Katrin Streckfuss-Bömeke (University Medical Center Göttingen) and expert Dr. Melanie Paillard (Université Claude Bernard Lyon 1) about the new study by Ajmail et al. While arrhythmias in calcium transients are easy to detect by human perception, quantifying arrhythmias in a computer-readable manner is often difficult. To tackle the challenge, the authors developed an open-source program called Arrhythpy that quantifies and classifies confocal microscopy-based Fluo-4 Ca2+ transients to generate a measure of arrhythmia. What is unique about Arrhythpy is that it directly measures the shape and frequency of transients. The program includes two parts: frequency profiling using a method called wavelet transformation, and measuring transient peak shape using a method called autocorrelation function. After combining these measurements, Arrhythpy classifies each transient into one of 6 different subgroups. No specialty training is needed, and no specialized platforms are required to use Arrhythpy.
