The cost of reworking a failed IHC slide due to poor tissue adhesion is estimated to be ~$80 per slide, considering the reagent cost and workload administration.1 Adhesion slides reinforce tissue adherence and integrity, minimizing the need to recut and restain the sample to ensure proper tissue morphological characteristics. Adhesion slides may also be used for H&E stains and special stains for added adhesion, but could retain excess reagent, or background staining, on the slide. In this study the differences in contact angle and in tissue adherence during microtomy were analyzed, investigation was done on whether different adhesion slides exhibit similar levels of background staining during histological staining procedures, and evaluation and comparison of the tissue adhesion properties of adhesion slide brands across different tissue types and applications was performed. After wide-ranging testing of adhesion slide characteristics, this study exhibited that not all adhesion slides are created equal. While water bath behavior showed to not be a relevant factor, there was considerable variation in background staining and tissue adhesion between slides. The results of this study suggested that it is important to determine what the needs are for your laboratory based on the types of staining done and tissue types used. It is also important to test adhesion slides to find the right slide or slides for your laboratory applications.

Franz at the Nano Life Science Institute (WPI-NanoLSI), at Kanazawa University.The research described in this podcast was published in the Journal of Cell Science in January 2024.Kanazawa University NanoLSI websitehttps://nanolsi.kanazawa-u.ac.jp/en/A novel role for S100A11 in focal adhesion regulationResearchers at Kanazawa University report in the Journal of Cell Science on a novel role of the small Ca2+ion-binding protein S100A11 [S one hundred A eleven] in focal adhesion disassembly.S100A11 is a small Ca2+ion-activatable protein with an established role in different cellular processes involving actin cytoskeleton remodeling, such as cell migration, membrane protrusion formation, and plasma membrane repair. It also displays F-actin binding activity and localizes to actin stress fibers, but its precise role in regulating these structures remained unclear.In their study, Tareg Omer Mohammed, You-Rong Lin, and Clemens M. Franz together with colleagues from the Nano Life Science Institute (WPI-NanoLSI), Kanazawa University, in Japan, and Karlsruhe Institute of Technology, in Germany, report a novel localization of S100A11 to disassembling focal adhesions at the end of contractile stress fibers in HeLa and U2OS cells. Specifically, S100A11 transiently appears at the onset of focal adhesion disassembly, reliably marking the targeted adhesion sites for subsequent disassembly. Interestingly, S100A11 leaves focal adhesion sites before the completion of disassembly, indicating that S100A11 plays a specific role in the initiation of adhesion site disassembly, rather than the disassembly process itself.So what are focal adhesions anyway and what can we learn from them?Focal adhesions are integrin-containing cell/matrix adhesion sites enabling cells to adhere to the cellular environment and to apply cellular contraction forces during extracellular matrix remodeling. Directed cell migration requires the coordinated assembly of new adhesion sites at the front, and disassembly at the rear of the cell, and better understanding mechanisms regulating focal adhesion turnover is, therefore, an important goal in cell migration and invasion research. The newly discovered role of S100A11 in focal adhesion disassembly extends insights into the molecular mechanisms underlying focal adhesion site disassembly.The authors furthermore delineate a force-dependent recruitment mechanism for S100A11 to adhesion sites involving non-muscle myosin II-driven stress fiber contraction, activation of mechanosensitive, Ca2+ ion-permeable Piezo1 channels, and intracellular Ca2+ ion influx at mechanically stressed focal adhesions. In turn, locally elevated Ca2+ ion levels activates and recruits S100A11 to the adhesion sites targeted for disassembly. So how did they work this out?The force-dependent recruitment of S100A11 to stressed focal adhesions was confirmed using a micropipette pulling assay able to apply pulling forces onto individual focal adhesion sites. Even when myosin II-dependent intracellular contractility was inhibited, external pulling forces still recruited S100A11 to stretched focal adhesion sites, corroborating the mechanosensitive recruitment mechanism of S100A11. However, extracellular Ca2+ ion and Piezo1 function was still indispensable, indicating that myosin II-dependent contraction forces act upstream of Piezo1-mediated Ca2+ ion influx, in turn leading to S100A11 activation and focal adhesion recruitment.Lastly, the authors show impaired focal adhesion translocation and disassembly ratNanoLSI Podcast website

What Causes Frozen Shoulder?It is not known exactly what causes this condition. Immobilization of the shoulder (after an arm injury, for instance) can lead to a frozen shoulder. Inflammation of the muscles and/or tendons, as with rotator cuff tendinitis or bursitis, can also cause the shoulder joint to become frozen. The glenohumeral joint is surrounded by muscles and tendons and held together by ligaments. When the balance between the ligaments, tendons, and muscles is interrupted the tissue form what are called adhesions. Adhesions turn fascia, the covering of the muscles into raisins. Raisins? Are you crazy?What's am Adhesion?Adhesions are easy to visualize as raisins. Muscle tissue should resemble grapes. When the fascia becomes irritated the covering shrivels up and causes pain that would limit motion. The more something hurts the less likely you are to use it. Adhesions in my estimation are a big cause of adhesive capsulitis. Like any muscles that are not used they will atrophy. So now when I mention atrophy the raisin-to-grape analogy would make more sense. There are many nerves in the fascia which is another reason shoulder pain due to adhesions is so painfulMuscles That Cause Frozen ShoulderIf you have had trauma to your shoulder there can be other muscles involved. What I am going to tell you is that there are 4 primary muscles involved in adhesive capsulitis. BY addressing these muscles first we find the most success in treating shoulder pain.SubscapularisThe Subscapularis the muscle found in front of your scapula internally rotates the humerus and brings it closer to the midline of the body. It also protects the shoulder from moving out of place in a forward direction.The Latissimus DorsiThe latissimus dorsi is responsible for a lot of things. It helps to stabilize your pelvis, it goes all the way down from your shoulder to your lumbar spine. The lats play an extremely important role in stabilizing the neck, shoulder, back, and hips. Think of the latissimus as the anchor of your back. An unstable shoulder needs to be rectified when you have a frozen shoulder.The Middle DeltoidOf the three Deltoid muscles,the Middle Deltoid is the one we see most often involved with cases of frozen shoulder. There are three heads of the deltoid muscle group. They help lift the arm front, side, and backward. The anterior and posterior portions are stronger and are usually safe from strains. It is interesting to note that the deltoid muscle is the largest arm muscle there is, even larger than the pectoralis muscles. The supraspinatus is most commonly injured because there are three different positions where it can become compressed. Overuse of the shoulder can cause the muscle and tendons to become inflamed. The supraspinatus stabilizes the shoulder joint by keeping the head of the humerus firmly pressed medially against the glenoid fossa of the scapula. The supraspinatus moves the shoulder inwards and allows you to lift the arm. Dr. Dan Regan discusses his OutSmarting Osteosarcoma funded work on Focal Adhesion Kinase (FAK) inhibition to improve losartan-sunitinib immunotherapy in metastatic osteosarcoma. - Dr. Dan Regan is an Assistant Professor at the Flint Animal Cancer Center at Colorado State University and he is one of our OutSmarting Osteosarcoma 2022 grant recipients. Dr. Regan received his DVM degree from the University of Georgia and subsequently completed his residency training in veterinary anatomic pathology and PhD in the Department of Microbiology, Immunology, and Pathology (MIP) at Colorado State University. In 2018 he joined the Flint Animal Cancer Center and MIP in the College of Veterinary Medicine and Biomedical Sciences at Colorado State University. The focus of Dr. Regan's laboratory is to increase our understanding of the interplay between the immune system and (non-immune) tumor stroma, and how these compartments of the tumor microenvironment promote metastasis as well as respond to and mediate extrinsic mechanisms of resistance to anti-cancer therapy. To investigate this area of cancer biology, his laboratory utilizes a combination of in vitro 3-dimensional tumor co-culture models and animal models, focusing on breast and bone cancer (osteosarcoma). Dr. Regan also has a strong interest in comparative oncology and leveraging naturally occurring cancers in dogs as both a surrogate and intermediary model to evaluate and validate his laboratory's investigations into the tumor microenvironment. In collaboration with the laboratory of Dr. Steve Dow, his lab conducted immunotherapy clinical studies in dogs with spontaneous osteosarcoma and these study results have led to a phase I clinical trial in children with osteosarcoma.
