※ 投稿邮箱:418150505@qq.com※ 本文章首发订阅号:百车全说,订阅号阅读更加方便,欢迎关注。你有没有发现,最近一年里,不光国产新能源的价格越来越卷,就连合资品牌的新能源价格也在慢慢放低身段。就像丰田的bZ4X,之前可是要25万多才能买到中高配,现在19.98万起,据说有的地方还降了6万,只要13.98万起。还有日产日产艾睿雅上个月的湖北大促销也是一下降了6万。但是今天要说的是一款全新车型,前段时间上市的别克E5,定位中大型纯电SUV,售价却只要20.89万起。这个价格一出来网友也是直呼真便宜,有诚意。事实真的如此吗?我们这期节目就来好好聊聊别克E5的价格到底贵不贵?新车都有哪些亮点?适合什么人买,以及配置又该怎么选?别克E5的价格贵不贵?直接说结论,确实很便宜。我就问你一个问题,如果买合资燃油车,你会拿别克对比哪些品牌?无非是大众、丰田、本田 、日产等等。那么,你放眼整个20万的合资纯电SUV市场,看看除了别克E5,还能买到哪些产品?日系你可以看三款:本田e:NS1、丰田bZ4X和日产艾睿雅。本田e:NS1就不用说了,定价17.5-21.8万,花20万就买个XR-V的油改电,还是个小型SUV,确实就是妥妥的韭菜。估计绝大多数人也压根都没听过这台车。本田都不愿意给这车做广告,完全是来打酱油的。丰田bZ4X定位中型SUV,但是车长还不足4米7,轴距2850毫米,起售价19.98万起。其实一开始bZ4X的售价非常贵,都要去到25万才能买到中配。最近迫于库存压力和内卷的大环境,才开始大幅降价,指导价降到了19.98万。但是听说最近这个售价又降到了13.98万。之所以这车即便降价也无人问津,大概率因为三点。丰田过于相信自己的品牌力,还是按照燃油车的思路销售,价高配低,电动车准用户不吃这一套。其次,有车主反映在0-4摄氏度的条件下,bZ4X的续航达成率只有约48%。作为参考,同样参与测试的特斯拉Model Y四驱版本的续航达成率为近七成,而奔驰EQA和大众ID.4也有六成以上的表现。同时,在低温环境下快充速度慢,也被众多车主吐槽。日产艾睿雅也是自恃清高的主,一个紧凑级纯电SUV定价却要28.48万起,车长才4米6出头,轴距2米8都没有,价格真的是虚高太多了。如果艾睿雅能有目前bZ4X的降价幅度,我猜销量应该会上来。因为同是日系纯电SUV,它可比bZ4X好看多了。并且,日产是真的在纯电技术上有积累,日产Leaf曾经常年位居全球电动车销量榜第一的位置,只不过中国消费者大多不知道而已。再看德系这边,大众的ID系列虽然定价高,但是人家早早地就把优惠放开了。比如ID.4系列虽然定价19万多起,但是ID.4中低配优惠完落地都在16万左右甚至更低,所以它的销量也不小。现在上汽大众ID.4 X加一汽大众ID.4 CROZZ一起每个月也有4000多台,在合资里绝对算卖得好的。但是ID系列里也只有ID.3和ID.4卖得好,同样是中大型SUV的ID.6优惠就很少,落地基本都要25万+,销量自然也不会太好。现在咱们再回头看看,合资品牌中大型SUV的别克E5,长宽高为4892/1905/1684毫米,轴距2954毫米。和日系比,那肯定是大了两圈都不止,即便对比德系ID.6 X,车长还长了16毫米,但是轴距也短了11毫米。而ID.6 X是7座,别克E5是五座,有点类似汉兰达与冠道的竞争关系。E5和ID.6 X比,E5属于是大两房,ID.6 X是小三房。更重要是E5比ID.6 X的起售价要便宜了五万。说完了合资,咱甚至可以对比国产,别克E5的尺寸也确实很能打。就比如一向以性价比著称的比亚迪,20万的纯电SUV你也只能买到宋PLUS EV。就算你再加上混动,也只能买买唐DM-i和护卫舰07。而唐EV的起售价也要去到28万多了。并且对比唐的尺寸,E5车长比它长了22毫米,轴距更是长了134毫米,而且唐还不是比亚迪最新的e3.0纯电平台,E5是通用最新的奥特能纯电平台。说句不该说的,别克E5甚至比自家燃油车定价还便宜,真的是别克狠起来连自己都打。先来看看花20万能买到哪些别克燃油SUV?昂科威S 定价20.99万起,昂科威PLUS定价22.59万起,这两车都比E5小不少。更夸张的是别克E5的尺寸都已经和29.99万起的昂科旗非常接近了。E5的车长比昂科旗短了99毫米,但是轴距却比昂科旗长了91毫米。所以理论上说,E5的车内空间基本和昂科旗差不多,但是售价却便宜了9万。即便优惠完,昂科旗的落地价也是要比E5高好几万,这还没算上E5有着更好的内饰设计以及更丰富的功能配置。此外,别克E5的同门师哥凯迪拉克锐歌,售价都要43.97万起,它俩可是出自同一平台,都是中大型SUV,只不过单电机版一个后驱,一个前驱。但是别克E5却比锐歌便宜了整整一半的价格,你说这个价格让以后的凯迪拉克锐歌还怎么卖?一个凯迪拉克标值20万吗?不过这也是玩笑话,锐歌的用料、质感以及外形设计肯定也要比E5强不少。但是网友都调侃, 别克E5就是凯迪拉克锐歌“青春版”,我觉得还挺贴切的。因此一句话总结别克E5,20万的定价,30万的尺寸。即使目前没有优惠,也有一定的性价比。别克E5有哪些亮点?既然说它的定价很良心,那它究竟有哪些亮点?第一,肯定是尺寸和空间。这个就不用多说了,刚才说的已经很透彻了。20万出头,基本没有比它尺寸还大的合资纯电SUV了,国产品牌也屈指可数。第二,和凯迪拉克锐歌一样,用的都是通用最新的奥特能纯电平台。这个平台有两大亮点,一是对电池的安全性确实下了不少功夫。直接说结果,奥特能平台的电池经过了很多测试,比如将电池包从5米的高空坠落,没起火也没冒烟,并且电池模组和冷却液管都完好。这个国家标准是1米。奥特能在诞生初期,通用内部的标准比国标还要高得多。而且电池壳体使用超高强度的钢井字形结构,大幅优化了电池的碰撞防护性能。其次,这个平台在智能化上也有创新。比如无线连接的电池管理系统(wBMS)。据说这个创新在布局上可以减少90%线束,以此来提升电池组的安全性。不过我觉得这个平台最大的特点就是下到20多万的别克E5,上到凯迪拉克锐歌,以及海外一千多匹马力的全新悍马EV,用的都是奥特能。所以不严谨的说一句,别克E5和悍马EV也是远房兄弟。第三,E5起步就是高配。要说别克E5内饰最大的亮点,一定是中控台上这块30英寸的一体式弧面6K屏幕。好像自从福特EVOS开始,美系车终于把内饰设计给重视起来了,之前屏幕要么不舍得给,要么杵着个竖屏。但是从福特的带鱼屏再到别克E5的曲面屏,不得不说,只要认真对待消费者的需求,就不会设计不出好看的内饰。E5上的这块30英寸的屏幕其实已经很大了,而且中间没有大黑框风格,看起来就是一整块。不过,锐歌的33英寸屏幕更大。但是E5的定价只有锐歌的一半,屏幕尺寸小了3英寸,可以理解吧。更关键的是,这块屏E5是全系标配。除了屏幕以外,像方向盘加热、百度地图、VCS智能座舱、全新eConnect系统、高通骁龙8155芯片、eCruise Pro智能驾驶辅助等也都是全系标配。是不是挺良心的?配置怎么选?目前别克E5售价20.89-27.89万,有五个配置,标准续航和长续航各有低配和高配,再加一个四驱艾维亚顶配。不过主流的四个单电机配置价格都在23.99万以下。但就像刚才说,别克E5入门即高配,所以如果你对续航没有要求,那么最低配的智享标准续航版就是最有性价比的配置,毕竟三元锂电池也有68度,CLTC续航545公里,日常也是完全够用的。并且两个标准续航的电机马力也要更大,标准续航的单电机最大马力245匹,最大扭矩330牛·米,售价20.89万。长续航的电机最大马力204匹,330牛·米,电池包也从68度涨到了80度,CLTC续航620公里,定价22.29万。因此最低配这2个配置最值得推荐。如果你觉得预算还没花完,那可以看看23.99万的臻享长续航。配置上还多了20英寸轮圈、电动后备箱+记忆、开门预警、360全景影像、泊车入位、真皮座椅+座椅通风、121色氛围灯等。至于艾维亚的四驱版,我觉得就没有必要了。虽然它是四驱双电机,但是系统最大马力287匹,,百公里加速6.7秒。但是放眼整个市场,27.89万的定价显得性价比不是那么高,毕竟25-30万可选的电动车可以太多了,而且很多都是三秒俱乐部成员。所以真正推荐入手,要么最低配,要么长续航的高配。别克E5适合什么人买?和自家的兄弟又该怎么选?20万这个价位能买的电车已经非常多了,有像smart精灵1号、极氪X这样的精品小车,也有像比亚迪汉EV、深蓝SL03这样的高性价比中型、中大型轿车,还有深蓝S7、小鹏G6、比亚迪宋PLUSEV等一众国产纯电SUV。但是20万想买台纯电SUV,还要合资,可选的真不多。所以别克E5的受众群体就很明显了,在这个价位,不仅想要一台空间大的纯电SUV,且更青睐合资品牌,那别克E5确实是个不错的选择。至于和自家兄弟E4怎么选,其实也很简单。别克E4新车最大的亮点就在它的外观设计。其实我也没想到,作为一款合资的纯电车,做得确实还挺好看。虽然前脸和E5差不多,但是侧面和车尾和E5完全就是两台车了,一个是正常的SUV造型,一个是溜背造型。并且车尾还有两个尾翼设计,看起来确实很动感,这个设计年轻人肯定喜欢。而且内饰标配的30寸曲面屏也是亮点之一。所以E4和E5最大的区别就在外观,E4更像是E5的轿跑版。另一个亮点肯定是它的空间和尺寸了,E4长宽高为4818/1912/1580毫米,轴距2954毫米。长度比E5短了74毫米,宽度相差7毫米,高度矮了101毫米,所以E4的造型更低趴一些。其他地方E4和E5基本就一样,不管是30寸的6K曲面屏还是方向盘,基本没区别。而且再到动力和续航,差别也不大,标准续航版搭载65度的弗迪磷酸铁锂电池,CLTC续航530公里,顶配的长续航四驱搭载79.7度的时代上汽三元锂电池,CLTC续航620公里。此外,E4最大的亮点还在于结合了GS套件。因为E4的定位明显要更年轻,所以比起E5,E4加入了GS的运动套件。不过GS在E4上就是个皮肤,类似宝马的M套件、奥迪的S-LINE等。但是有了这套运动皮肤的加持,E4确实要更有吸引力。所以综合来说,别克E4的消费者肯定要比E5更加的年轻化,而且更加追求整车的颜值和设计。写在最后不过,虽然别克E5的性价比很高,亮点也很多,但是不足之处依然有。比如在造型上,还是有些油车的影子。而且别克E5的动力比较保守,连四驱双电机的百公里加速也只有6.7秒。当然话分两头讲,照顾续航,就得牺牲动力,这是取舍问题。其次,虽然E5智能化速度还不够快。比如,如果你选装高阶智能驾驶辅助Super Cruise,暂时还不能用,得等到年中才能开放使用。而目前市面上能实现NCA城市智能驾驶的车都已经有很多款了。而且,现如今的消费者在买电车时,消费观念也已经很明确,很大一部分消费者在买车前,就清楚自己不仅仅是在买一台电车,更多的是在买一种全新的消费体验,想要花相同的价格来体验到同价位油车所没有的功能和服务。如果以这种眼光来看别克E5的话,别克E5更多的是一台空间很大、性价比很高的工具电车。车内功能很全,但是没有那种眼前一亮的感觉。而在未来的车主服务方面,别克E5不是独立一套服务体系,还是依托原有的燃油车经销商,这也是需要选择的难题。但无论怎么说,E5这个产品看得出是给出很大诚意了,后续再努努力,说不定别克在合资纯电市场还真能闯出一番天地。作者:三刀、新一编辑:新一可以添加微信46415254加入我们的社群音频图文更新在订阅号: 百车全说每期抽三条留言,每人赠168元的“芥末绿”燃油添加剂一瓶点击订阅,每周三,周六更新会有提醒新听友可以搜索:百车全说2014,百车全说2015,百车全说2016,往期300多个小时的节目可供收听

September 2022 wurde ein Druckverlust im Strang A von Nord Stream 2 gemeldet, später auch in beiden Strängen von Nord Stream 1. Die Ursache war eine Reihe von Explosionen, die seismographisch aufgezeichnet wurden. Um 02'03 Uhr verursachte die erste Explosion ein Leck in Strang A von Nord Stream 2, etwa 12 Seemeilen südöstlich der dänischen Insel Bornholm. Am selben Tag um 19'03 Uhr entstanden mindestens drei weitere Lecks zwischen Bornholm und Öland, von denen zwei in den Strängen von Nord Stream 1 liegen und das dritte erneut Strang A von Nord Stream 2 betraf. Das letzte Leck wurde erst am 29. September 2022 von der schwedischen Küstenwache entdeckt. Der Strang B von Nord Stream 2 blieb unbeschädigt. Die Lecks befinden sich in 70-80 Metern Tiefe und in einem Abstand von mindestens einer Seemeile voneinander. An der Wasseroberfläche waren Gasblasen zu sehen, die im Umkreis von einem Kilometer über den Lecks aufstiegen. Die Lecks liegen knapp außerhalb der Hoheitsgewässer in den ausschließlichen Wirtschaftszonen Dänemarks und Schwedens. Desinformationstechniken können eine ernsthafte Bedrohung für die Demokratie und die Gesellschaft als Ganzes darstellen, da sie dazu führen können, dass Menschen falsche oder irreführende Informationen glauben. 本文章发布于订阅号:百车全说,订阅号阅读更加方便,欢迎关注前37分钟聊丰田bZ4X,后半部分聊五一假期小故事。今天这期节目聊聊前不久刚刚预售的丰田bZ4X。由于刚开始预售,网络上软文太多,很多网友看得晕头转向,甚至有些都已经蠢蠢欲动准备下单了。但是看了看钱包,发现30万买台丰田的电动车,总感觉哪里不对。所以后台收到很多人私信,问我这车到底好不好,能不能买?我先说结论,如果你不差钱,信丰田,想玩个新鲜,可以买。但是你要谈性价比,谈科技感,谈娱乐和舒适性配置,市面上大把的车可以把它虐成渣渣。今天这期节目,我们就详细聊聊这款丰田bZ4X,与大家分享一些我的观点。现如今,买电动车的车主也是越来越挑剔了,因为中国应该是目前世界上可以选择的电动车品牌最多的国家。20-30w这个区间,我们也可以找到几十款电动车可以买。特斯拉的Model3、蔚来的ET5、小鹏P5/P7/G3、比亚迪汉/唐还有后续的一系列海洋系列,其他的像是吉利、奇瑞、长城、广汽埃安等等品牌,一堆的电动车躺着被你选,你为什么一定要买丰田bZ4X呢?是这台车长得太帅,让你无法自拔吗?这台车的整体造型设计,与丰田家族传统燃油SUV相比,并没有特别突出的亮点。反倒是前轮上方的两个黑色轮眉格外醒目。按道理说,你要突出越野车的狂放不羁气质,做个撞色处理我也能够理解。可是那两坨不规则的黑色轮眉设计,就像是涂口红涂歪了的感觉,或者说像是侧面的保险杠还没有喷漆就出厂的感觉。我看了一下玫瑰棕,魅力银都是这样两坨黑色在前轮上方,我是无法接受,不知道大家什么感觉。如果你非要送我一辆bZ4X,我会毫不犹豫地选择墨渊黑,用车身的黑色去遮盖这两坨黑色的轮眉。这台车的内饰设计,也没什么出彩的地方。对外展示的都是7英寸全液晶仪表+12.3英寸中控屏的版本。实际上22万和24万的版本,中控屏仍然是8英寸的小屏,如果是单独设计的8英寸小屏那还好点,可是丰田为了节省成本,直接在原有的大屏面板上嵌入了一块小屏幕,左右两边的黑框可以停下一排游艇,奇丑无比。不仅如此,22万和24万的版本还是塑料方向盘,没有倒车雷达,没有360全景影像,没有无钥匙进入。更夸张的是,主副驾座椅还是手动调节。20多万的电动车,丰田真的是心大,难道不看看周围的竞品吗?哪儿来的自信?要说这车唯一良心的,应该就是L2级的智能驾驶辅助全系标配吧。不过,在20万级的电动车里,它要连这个都不标配,估计要被网友喷出翔来。不仅同级别的电动车几乎都是标配,就连丰田自家10几万的卡罗拉和雷凌都是标配。所以毫无疑问,看bz4X的客户肯定第一时间pass掉了22万和24万的两个版本,那么就只剩下26万、28万和30万三个版本可选了。26万是两驱(前驱)顶配,28和30万都是四驱版本。26万和28万配置几乎一样,其实就是两驱和四驱的差别。可是丰田bZ4X有个很有意思的地方,它的两驱版本配备的是150kw单电机,峰值扭矩266牛米,零百加速7.5秒,CLTC续航615km;而四驱版本前后各有一个80kw的电机,综合最大功率160kw,峰值扭矩337牛米,零百加速6.9秒,CLTC续航560km。也就是说四驱版本的性能并没有增加多少,反倒是续航缩水了50多公里。我觉得没有谁会真的拿一台电动SUV去玩越野吧?虽然丰田与斯巴鲁共享x-mode技术,前后扭矩也会有动态分配,但毕竟是前后轴电机驱动,难不成还给加三把锁,然后搞个扭矩放大多少倍吗?这充其量算是个全轮驱动,说是四驱都算是给足面子了。所以,如果把四驱版本去掉,丰田bZ4X唯一值得入手的,就是两驱顶配26万这个版本。但是我看了一眼配置单,所谓顶配,轮毂还是和低配一样是18寸的(顶配20寸),没有并线辅助,没有电动后备箱,没有车内空气净化,音响全系都是6个扬声器,当然更不可能是什么品牌音响。可能有人会说,丰田bz4x的配置虽然不高,可是续航还行。你刚刚不是说了,两驱CLTC续航可以到615km吗?这里解释一下,CLTC续航是在去年7月1日被用来取代NEDC续航的一种标准,但是CLTC续航被很多专业人士质疑虚标太严重,甚至比NEDC还要严重。比如特斯拉Model 3 Performance,新款从76.8kWh增加到78.4kWh,只多了1.6度电,竟然续航从605km增加到了675km。特斯拉1.6度电能开70km吗?要知道,电动车15kWh/百公里已经算是非常优秀的能耗表现了。说白了,续航增加的原因就是之前用的NEDC,现在换成了CLTC。给大家一组数据对比:NEDC续航484km,CLTC续航就是509km,WLTP续航只有398km。换句话说,A厂家如果用CLTC续航,B厂家用WLTP续航,那肯定是B厂家吃亏。所以现在市面上大部分电动车对外宣传,都会用CLTC续航标准,比如凯迪拉克Lyriq的续航650km;福特Mustang Mach-E续航619km;BMW IX续航665km,这些都是CLTC续航。其实水分都很大,真实续航起码打8折。而且,丰田bZ4X最低配22万的版本,不仅配置丐中丐,甚至电池包都缩水了16度电,CLTC续航才400km,花22万买一台实际续航连国产10来万的电动车都比不过的合资车,我实在想不出来这样的客户图的是啥?有人可能要问,丰田bz4x难道一点亮点都没有吗?那个“高效太阳能充电穹顶”,难道不是丰田独有的技术吗?宣传说“每年吸收能量可行驶1800km呢。就算我每年就开1万公里,如果放在太阳底下充电就能续航1800km,那我岂不是1/5的日子都不用充电了吗?说实话,丰田bZ4X这个所谓车顶太阳能电池板,我看着都想笑,咱们比亚迪F3DM早在2010年就给安排上了,12年前咱们国产车玩剩下的,现在又被拿出来炒作。而且还是只能在最顶配30万的版本上选装。选装之后,分段式天窗就没了,车顶覆盖一片光伏板。光伏发电的效率有多低,这个可能需要专业人士来解释一下。根据我查到的相关资料,一平米的光伏板,如果日照充足的情况下 ,按200w的充电标准,一天日照按4个小时计算,也就是充了1度电左右。如果日照不充足,充电效率也就是50-100w之间,我看着我的65w手机充电器,笑而不语。之前如果关注过丰田bZ4X的朋友,应该知道海外版本还有类似F1赛车的异型方向盘,而且带线控转向。可惜到了国内,这两项都取消了。取消异型方向盘我能理解,毕竟法律法规各个国家和地区不一样,但是线控转向你可以不取消。这玩意虽然贵的要命,但的确是个好东西。英菲尼迪Q50L当年上市的时候,就大吹特吹了通线控转向,算是给老百姓普及了一下这个技术。可惜的是,路面上99%的Q50L都没有这个技术,因为只有售价39.98万的最顶配版本才有线控转向。丰田bZ4X不但顶配没有,选装都不给选。线控转向,简单理解就类似于赛车模拟器,它没有传统机械结构来得那么真实,电机无法模拟轮胎受力带来的方向回跳和小震动。但是线控转向最大的优势就是转向比可以自由变化,转向不再受到机械的限制,它可以让你手感重一些,也可以让你手感轻一些,可以让你在路口轻松调头,也可以让你左右晃动方向盘没有虚位。理论上讲,工程师想怎么调整都可以随心所欲。战斗机上用的就是线控转向,因为它不需要考虑成本问题,也不用考虑驾驶乐趣,电子信号的传递速度肯定优于液压结构。今后无人驾驶如果普及,线控转向大概率也会随之普及。但是目前来看,但凡敢用线控转向技术的,首先要配备3个独立的ECU行车电脑,如果一个出故障,另外两个要保证正常运转。不仅如此,还要配备一套传统的机械转向,如果三个ECU都出故障,系统会立即接合离合器,恢复机械转向连接。听起来是不是很无聊的一个设计?为了一个普普通通的转向功能,消费者要多花这么多冤枉钱。所以,丰田bZ4X更不可能在30万的电动车给你配备这种功能。不过,今后与bZ4X同平台的雷克萨斯RZ,说不定有可能配备线控转向,毕竟丰田都卖这个价了,雷克萨斯RZ不卖个40-50万怎么对得起观众呢?别说3个ECU,给你整6个也不是事。当然,按雷克萨斯对中国消费者双标的态度,能配足3个就不错了,而且就算给1个还是有人跪舔。我看到其他媒体稿件里提到,丰田bZ4X的轴距2850mm,空间堪比汉兰达。数据上看,的确没有错,汉兰达的轴距也是2850mm。可是汉兰达车长4965mm,这车才4690mm,也就比丰田Rav4荣放长了90mm,宽度和高度也和荣放差不多。说白了,就是一台紧凑级suv,为什么大家硬要说这是中型suv呢?电动车由于自身结构的特殊性,没有发动机和变速箱,所以前后轮都会顶边设计,这样一来,轴距自然就变长了。目前为止,同级别电动车的轴距都不会太差。就算有些竞品轴距看起来比丰田bZ4X小一些,你实际去体验看看,空间也不会有明显的感知小很多。还有一些网友猜测,这台车会不会是比亚迪贴了丰田标在卖?咱们有一说一,人家丰田自家的eTNGA架构,不至于还得去抄袭比亚迪吧。比亚迪现在纯电车型都在更新e3.0平台,两者之间还是有很大差别的。再加上丰田bZ4X的电池包用的是丰田和宁德时代共同开发的三元锂电池,这也与比亚迪的磷酸铁锂刀片电池不一样。丰田bZ4X的底盘架构也没预留发动机变速箱和油箱位置,也谈不上是油改电的产品。但是大家注意这个TNGA平台下的产品,今后会衍生出很多差半级的车型,而这些车型宽度不变,只是长度、高度、轴距会有变化。比如卡罗拉和亚洲狮,RAV4荣放和凌放(轴距都没变)。毫无疑问,eTNGA下的丰田电动车肯定也会玩这种手法,疯狂的制造差异化产品,搞得你眼花缭乱 ,光在丰田电动车体系里对比就花了N久时间,当然就没时间和精力去对比其他车型了。在充电方面,bZ4X也没什么亮点。最大150kw充电功率,在400v快充桩上跑满的话,0-80%大约30分钟时间,市面上大部分车都可以做得到。而丰田bz4x的充电口全部在车头两侧,左边是快充口,右边是慢充口。我特别不喜欢充电口在车头的电动车,这是厂家节省成本,但是麻烦了客户的行为。你想想,自家车位上好不容易装了个充电桩,结果每次还得拖根长长的充电线线,一直拉到车头位置。而那些在公用充电桩充电的车主更惨,公用充电桩的充电线很短,你只能车头扎进车位,且不说倒车出车位的时候,左右如果停有车辆,几乎全部都是盲区,非常危险。我们楼下就有一排充电桩,充电桩前面是双车道,很窄,我每次看网约车过来充电,为了让车头进入车位,前前后后倒车无数遍,后面堵着长长的一排车,我看着都心疼。至于丰田bZ4X上面的这块12.3英寸中控屏,以及它的车机系统,我相信大多数人看着就提不起兴趣。说来说去,也就是车家互联、QQ音乐、百度地图、语音控制这些功能,连OTA远程升级都没有,这套车机大家不用抱有太大幻想。车机系统方面,玩过国产新造实力的,保准你觉得这就是上个时代的产物。智能驾驶方面也是一样,虽然标配了L2级智能驾驶辅助,但是丰田bZ4X从头至尾没有宣传自己有多少个毫米波雷达,多少个摄像头,激光雷达更是想都不用想的。说了这么多丰田bZ4X缺这少那的地方,其实总结下来一句话,就是这车性价比太差,我相信稍微有些常识的客户应该是一眼就看出来了。但是丰田为什么有底气给这么一台低配电动车定这么高的价格呢?我觉得有两点原因,一是他的竞品大众ID.4不是也定的差不多价格吗?大众ID.4好歹上汽和一汽还换了个后缀,改改造型卖,丰田bZ4X干脆名字和造型都懒得改,直接放一丰和广丰卖。而ID.4的尺寸还比丰田bZ4X小一圈,各方面参数和配置都差不多,凭什么ID.4能卖这么贵,丰田就不行呢?ID.4一样也有很多人买。况且丰田bZ4X这只是预售价,比如我分析的两驱顶配26万的版本,如果最终上市25.5万,那么跟ID.4的两驱顶配24万多,也就差1万来块钱。丰田应该还是比较自信,这1万多差价,如果大众和丰田二选一,还是有不少人站队丰田的吧。另一点原因,就是丰田毕竟是全球销量第一的汽车公司,在咱中国有1400多家门店,很多老百姓就认这牌子。丰田吃准一部分人还是会把传统观念带到电动车里来,比如保值、耐用、经济、省心等等。所以,你看丰田bz4x的销售政策,第一条就是:10年20万公里免费基础保养。有人一看,这可比雷克萨斯混动的免费保养周期还要长。实际上,电动车保养一次就1-2百块钱,便宜的很。所有销售政策里,也就这一条最吸引人,其他的什么礼包,置换补贴,增购补贴,送电卡都没啥吸引力。可是我有个疑问,按理说不接受电动车的人,应该连丰田电动车也不接受。难道丰田电动车就没有续航焦虑了?就没有保值率问题了?就不会起火自燃了?而那些接受电动车的客户,早把市面上的车型比较了个遍了。他们还能接受丰田bZ4X这么没性价比的产品吗?所以,我觉得丰田bZ4X有点像BBA造电动车的思路, 本文章发布于订阅号:百车全说,订阅号阅读更加方便,欢迎关注前37分钟聊丰田bZ4X,后半部分聊五一假期小故事。今天这期节目聊聊前不久刚刚预售的丰田bZ4X。由于刚开始预售,网络上软文太多,很多网友看得晕头转向,甚至有些都已经蠢蠢欲动准备下单了。但是看了看钱包,发现30万买台丰田的电动车,总感觉哪里不对。所以后台收到很多人私信,问我这车到底好不好,能不能买?我先说结论,如果你不差钱,信丰田,想玩个新鲜,可以买。但是你要谈性价比,谈科技感,谈娱乐和舒适性配置,市面上大把的车可以把它虐成渣渣。今天这期节目,我们就详细聊聊这款丰田bZ4X,与大家分享一些我的观点。现如今,买电动车的车主也是越来越挑剔了,因为中国应该是目前世界上可以选择的电动车品牌最多的国家。20-30w这个区间,我们也可以找到几十款电动车可以买。特斯拉的Model3、蔚来的ET5、小鹏P5/P7/G3、比亚迪汉/唐还有后续的一系列海洋系列,其他的像是吉利、奇瑞、长城、广汽埃安等等品牌,一堆的电动车躺着被你选,你为什么一定要买丰田bZ4X呢?是这台车长得太帅,让你无法自拔吗?这台车的整体造型设计,与丰田家族传统燃油SUV相比,并没有特别突出的亮点。反倒是前轮上方的两个黑色轮眉格外醒目。按道理说,你要突出越野车的狂放不羁气质,做个撞色处理我也能够理解。可是那两坨不规则的黑色轮眉设计,就像是涂口红涂歪了的感觉,或者说像是侧面的保险杠还没有喷漆就出厂的感觉。我看了一下玫瑰棕,魅力银都是这样两坨黑色在前轮上方,我是无法接受,不知道大家什么感觉。如果你非要送我一辆bZ4X,我会毫不犹豫地选择墨渊黑,用车身的黑色去遮盖这两坨黑色的轮眉。这台车的内饰设计,也没什么出彩的地方。对外展示的都是7英寸全液晶仪表+12.3英寸中控屏的版本。实际上22万和24万的版本,中控屏仍然是8英寸的小屏,如果是单独设计的8英寸小屏那还好点,可是丰田为了节省成本,直接在原有的大屏面板上嵌入了一块小屏幕,左右两边的黑框可以停下一排游艇,奇丑无比。不仅如此,22万和24万的版本还是塑料方向盘,没有倒车雷达,没有360全景影像,没有无钥匙进入。更夸张的是,主副驾座椅还是手动调节。20多万的电动车,丰田真的是心大,难道不看看周围的竞品吗?哪儿来的自信?要说这车唯一良心的,应该就是L2级的智能驾驶辅助全系标配吧。不过,在20万级的电动车里,它要连这个都不标配,估计要被网友喷出翔来。不仅同级别的电动车几乎都是标配,就连丰田自家10几万的卡罗拉和雷凌都是标配。所以毫无疑问,看bz4X的客户肯定第一时间pass掉了22万和24万的两个版本,那么就只剩下26万、28万和30万三个版本可选了。26万是两驱(前驱)顶配,28和30万都是四驱版本。26万和28万配置几乎一样,其实就是两驱和四驱的差别。可是丰田bZ4X有个很有意思的地方,它的两驱版本配备的是150kw单电机,峰值扭矩266牛米,零百加速7.5秒,CLTC续航615km;而四驱版本前后各有一个80kw的电机,综合最大功率160kw,峰值扭矩337牛米,零百加速6.9秒,CLTC续航560km。也就是说四驱版本的性能并没有增加多少,反倒是续航缩水了50多公里。我觉得没有谁会真的拿一台电动SUV去玩越野吧?虽然丰田与斯巴鲁共享x-mode技术,前后扭矩也会有动态分配,但毕竟是前后轴电机驱动,难不成还给加三把锁,然后搞个扭矩放大多少倍吗?这充其量算是个全轮驱动,说是四驱都算是给足面子了。所以,如果把四驱版本去掉,丰田bZ4X唯一值得入手的,就是两驱顶配26万这个版本。但是我看了一眼配置单,所谓顶配,轮毂还是和低配一样是18寸的(顶配20寸),没有并线辅助,没有电动后备箱,没有车内空气净化,音响全系都是6个扬声器,当然更不可能是什么品牌音响。可能有人会说,丰田bz4x的配置虽然不高,可是续航还行。你刚刚不是说了,两驱CLTC续航可以到615km吗?这里解释一下,CLTC续航是在去年7月1日被用来取代NEDC续航的一种标准,但是CLTC续航被很多专业人士质疑虚标太严重,甚至比NEDC还要严重。比如特斯拉Model 3 Performance,新款从76.8kWh增加到78.4kWh,只多了1.6度电,竟然续航从605km增加到了675km。特斯拉1.6度电能开70km吗?要知道,电动车15kWh/百公里已经算是非常优秀的能耗表现了。说白了,续航增加的原因就是之前用的NEDC,现在换成了CLTC。给大家一组数据对比:NEDC续航484km,CLTC续航就是509km,WLTP续航只有398km。换句话说,A厂家如果用CLTC续航,B厂家用WLTP续航,那肯定是B厂家吃亏。所以现在市面上大部分电动车对外宣传,都会用CLTC续航标准,比如凯迪拉克Lyriq的续航650km;福特Mustang Mach-E续航619km;BMW IX续航665km,这些都是CLTC续航。其实水分都很大,真实续航起码打8折。而且,丰田bZ4X最低配22万的版本,不仅配置丐中丐,甚至电池包都缩水了16度电,CLTC续航才400km,花22万买一台实际续航连国产10来万的电动车都比不过的合资车,我实在想不出来这样的客户图的是啥?有人可能要问,丰田bz4x难道一点亮点都没有吗?那个“高效太阳能充电穹顶”,难道不是丰田独有的技术吗?宣传说“每年吸收能量可行驶1800km呢。就算我每年就开1万公里,如果放在太阳底下充电就能续航1800km,那我岂不是1/5的日子都不用充电了吗?说实话,丰田bZ4X这个所谓车顶太阳能电池板,我看着都想笑,咱们比亚迪F3DM早在2010年就给安排上了,12年前咱们国产车玩剩下的,现在又被拿出来炒作。而且还是只能在最顶配30万的版本上选装。选装之后,分段式天窗就没了,车顶覆盖一片光伏板。光伏发电的效率有多低,这个可能需要专业人士来解释一下。根据我查到的相关资料,一平米的光伏板,如果日照充足的情况下 ,按200w的充电标准,一天日照按4个小时计算,也就是充了1度电左右。如果日照不充足,充电效率也就是50-100w之间,我看着我的65w手机充电器,笑而不语。之前如果关注过丰田bZ4X的朋友,应该知道海外版本还有类似F1赛车的异型方向盘,而且带线控转向。可惜到了国内,这两项都取消了。取消异型方向盘我能理解,毕竟法律法规各个国家和地区不一样,但是线控转向你可以不取消。这玩意虽然贵的要命,但的确是个好东西。英菲尼迪Q50L当年上市的时候,就大吹特吹了通线控转向,算是给老百姓普及了一下这个技术。可惜的是,路面上99%的Q50L都没有这个技术,因为只有售价39.98万的最顶配版本才有线控转向。丰田bZ4X不但顶配没有,选装都不给选。线控转向,简单理解就类似于赛车模拟器,它没有传统机械结构来得那么真实,电机无法模拟轮胎受力带来的方向回跳和小震动。但是线控转向最大的优势就是转向比可以自由变化,转向不再受到机械的限制,它可以让你手感重一些,也可以让你手感轻一些,可以让你在路口轻松调头,也可以让你左右晃动方向盘没有虚位。理论上讲,工程师想怎么调整都可以随心所欲。战斗机上用的就是线控转向,因为它不需要考虑成本问题,也不用考虑驾驶乐趣,电子信号的传递速度肯定优于液压结构。今后无人驾驶如果普及,线控转向大概率也会随之普及。但是目前来看,但凡敢用线控转向技术的,首先要配备3个独立的ECU行车电脑,如果一个出故障,另外两个要保证正常运转。不仅如此,还要配备一套传统的机械转向,如果三个ECU都出故障,系统会立即接合离合器,恢复机械转向连接。听起来是不是很无聊的一个设计?为了一个普普通通的转向功能,消费者要多花这么多冤枉钱。所以,丰田bZ4X更不可能在30万的电动车给你配备这种功能。不过,今后与bZ4X同平台的雷克萨斯RZ,说不定有可能配备线控转向,毕竟丰田都卖这个价了,雷克萨斯RZ不卖个40-50万怎么对得起观众呢?别说3个ECU,给你整6个也不是事。当然,按雷克萨斯对中国消费者双标的态度,能配足3个就不错了,而且就算给1个还是有人跪舔。我看到其他媒体稿件里提到,丰田bZ4X的轴距2850mm,空间堪比汉兰达。数据上看,的确没有错,汉兰达的轴距也是2850mm。可是汉兰达车长4965mm,这车才4690mm,也就比丰田Rav4荣放长了90mm,宽度和高度也和荣放差不多。说白了,就是一台紧凑级suv,为什么大家硬要说这是中型suv呢?电动车由于自身结构的特殊性,没有发动机和变速箱,所以前后轮都会顶边设计,这样一来,轴距自然就变长了。目前为止,同级别电动车的轴距都不会太差。就算有些竞品轴距看起来比丰田bZ4X小一些,你实际去体验看看,空间也不会有明显的感知小很多。还有一些网友猜测,这台车会不会是比亚迪贴了丰田标在卖?咱们有一说一,人家丰田自家的eTNGA架构,不至于还得去抄袭比亚迪吧。比亚迪现在纯电车型都在更新e3.0平台,两者之间还是有很大差别的。再加上丰田bZ4X的电池包用的是丰田和宁德时代共同开发的三元锂电池,这也与比亚迪的磷酸铁锂刀片电池不一样。丰田bZ4X的底盘架构也没预留发动机变速箱和油箱位置,也谈不上是油改电的产品。但是大家注意这个TNGA平台下的产品,今后会衍生出很多差半级的车型,而这些车型宽度不变,只是长度、高度、轴距会有变化。比如卡罗拉和亚洲狮,RAV4荣放和凌放(轴距都没变)。毫无疑问,eTNGA下的丰田电动车肯定也会玩这种手法,疯狂的制造差异化产品,搞得你眼花缭乱 ,光在丰田电动车体系里对比就花了N久时间,当然就没时间和精力去对比其他车型了。在充电方面,bZ4X也没什么亮点。最大150kw充电功率,在400v快充桩上跑满的话,0-80%大约30分钟时间,市面上大部分车都可以做得到。而丰田bz4x的充电口全部在车头两侧,左边是快充口,右边是慢充口。我特别不喜欢充电口在车头的电动车,这是厂家节省成本,但是麻烦了客户的行为。你想想,自家车位上好不容易装了个充电桩,结果每次还得拖根长长的充电线线,一直拉到车头位置。而那些在公用充电桩充电的车主更惨,公用充电桩的充电线很短,你只能车头扎进车位,且不说倒车出车位的时候,左右如果停有车辆,几乎全部都是盲区,非常危险。我们楼下就有一排充电桩,充电桩前面是双车道,很窄,我每次看网约车过来充电,为了让车头进入车位,前前后后倒车无数遍,后面堵着长长的一排车,我看着都心疼。至于丰田bZ4X上面的这块12.3英寸中控屏,以及它的车机系统,我相信大多数人看着就提不起兴趣。说来说去,也就是车家互联、QQ音乐、百度地图、语音控制这些功能,连OTA远程升级都没有,这套车机大家不用抱有太大幻想。车机系统方面,玩过国产新造实力的,保准你觉得这就是上个时代的产物。智能驾驶方面也是一样,虽然标配了L2级智能驾驶辅助,但是丰田bZ4X从头至尾没有宣传自己有多少个毫米波雷达,多少个摄像头,激光雷达更是想都不用想的。说了这么多丰田bZ4X缺这少那的地方,其实总结下来一句话,就是这车性价比太差,我相信稍微有些常识的客户应该是一眼就看出来了。但是丰田为什么有底气给这么一台低配电动车定这么高的价格呢?我觉得有两点原因,一是他的竞品大众ID.4不是也定的差不多价格吗?大众ID.4好歹上汽和一汽还换了个后缀,改改造型卖,丰田bZ4X干脆名字和造型都懒得改,直接放一丰和广丰卖。而ID.4的尺寸还比丰田bZ4X小一圈,各方面参数和配置都差不多,凭什么ID.4能卖这么贵,丰田就不行呢?ID.4一样也有很多人买。况且丰田bZ4X这只是预售价,比如我分析的两驱顶配26万的版本,如果最终上市25.5万,那么跟ID.4的两驱顶配24万多,也就差1万来块钱。丰田应该还是比较自信,这1万多差价,如果大众和丰田二选一,还是有不少人站队丰田的吧。另一点原因,就是丰田毕竟是全球销量第一的汽车公司,在咱中国有1400多家门店,很多老百姓就认这牌子。丰田吃准一部分人还是会把传统观念带到电动车里来,比如保值、耐用、经济、省心等等。所以,你看丰田bz4x的销售政策,第一条就是:10年20万公里免费基础保养。有人一看,这可比雷克萨斯混动的免费保养周期还要长。实际上,电动车保养一次就1-2百块钱,便宜的很。所有销售政策里,也就这一条最吸引人,其他的什么礼包,置换补贴,增购补贴,送电卡都没啥吸引力。可是我有个疑问,按理说不接受电动车的人,应该连丰田电动车也不接受。难道丰田电动车就没有续航焦虑了?就没有保值率问题了?就不会起火自燃了?而那些接受电动车的客户,早把市面上的车型比较了个遍了。他们还能接受丰田bZ4X这么没性价比的产品吗?所以,我觉得丰田bZ4X有点像BBA造电动车的思路, Most people don't think too hard on how their system should gracefully bow out rather than the old school method of complete failures and horrible restarts. Resilient software engineering is the discipline of making software and systems fail in ways that minimize and isolate failures while continuing to deliver service and availability. Basically, it means if you have a failure from hardware or dependencies, like a database, your service continues to work correctly and the broken parts just get shut down and replaced.Cloud-native software using microservices or even dynamically deployed containers or systems is the perfect way to implement resiliency in your operations. Look toward the next development cycle of your software and systems to begin implementing this immediately if you don't already have this in place. None of this really makes sense until you see an example, so think of it this way: you have a web-based service for customers to see their account profile and order history. It's built to scale with containers using AWS Elastic Kubernetes service—or EKS—and it is designed so when a system throws errors of any kind, that container is closed down. Then the Aws Elastic Load Balancer—or ELP—service points all subsequent requests to a different container instance in EKS.In that scenario, if a container is breached in a security event, or if something simply fails due to a software bug or data corruption, the service recovers by tossing a new system while yanking out the old system. This is security by designing self-healing IT systems. You get both security and stability for the same effort. Explosive growth is scary; 30X growth in months is terrifying. Zoom did it. Can you? Very few companies can stay functioning, let alone secure in those situations.Software-Container Supply Chain Sees Spike in Attacks. I don't think I've beat the drum of supply-chain attacks enough. These are on the rise now that there is a great example of how effective these are. I sure hope you've secured your supply chain. I'm sure you haven't, but we can always hope.Four states propose laws to ban ransomware payments. This is a bit like making it illegal to pay kidnappers or terrorists. I know many companies will get owned and pay anyway, and regulations to stop money flowing to criminals is nothing new. There will be loopholes found and exploited, like in all things. Keep up with what laws affect your organization and how you perform security. To stop ransomware pandemic, start with the basics. We security people repeat ourselves constantly because implementing the basic security defenses mitigates most risk for most organizations. The US federal government is pushing hard on cybersecurity now that they were owned in front of the whole world by the SolarWinds and MS Exchange debacles. These things are driven by people who have studied your systems and might have been in them for weeks.What Lies Ahead for K-12 Cybersecurity? As the president and principal of a small elementary school and a technologist, I've implemented mostly cloud services to support the school operations and classroom work. Many of us in tech who work with large organizations in state, local, and higher ed—called SLED—national, or federal governments, and large corporations forget that there are small and mid-sized businesses—or SMBs—and K through 12 schools that also have the same concerns we do. After all, we all run Windows, Mac OS, Unix, and Linux, middleware and cloud services.How to Protect Healthcare Data from Ransomware Attacks. Abstract: Influenza A virus (IAV) is a highly contagious human pathogen responsible for nearly half a million deaths each year. Non-structural protein 1 (NS1) is a crucial protein expressed by IAV to evade the host immune system. Additionally, NS1 has been proposed to stimulate translation because of its ability to bind poly(A) binding protein 1 (PABP1) and eukaryotic initiation factor 4G (eIF4G). We analyzed the interaction of NS1 with PABP1 using quantitative techniques. Our studies show that NS1 binds as a homodimer to PABP1, and this interaction is conserved across different IAV strains. Unexpectedly, NS1 does not bind to PABP1 that is bound to poly(A) RNA. Instead, NS1 only binds to PABP1 free of RNA, suggesting that translation stimulation does not occur by NS1 interacting with the PABP1 molecule attached to the mRNA 3'-poly(A) tail. We propose that NS1 binds to the eIF4G complex at the 5'-end of the mRNA and recruits the RNA-free PABP1, which may stimulate translation initiation by promoting the association of the ribosomal subunits. Respiratory syncytial virus (RSV) is recognised as the most frequent cause of severe lung infections in infants and cattle worldwide. Currently, no effective treatments are available and the development of a successful vaccine has been hampered by the fact that natural infection does not provide complete and durable protection. RSV nonstructural proteins, NS1 and NS2, are strong inhibitors of IFN α/β-production by specifically preventing interferon regulatory factor (IRF)-3 phosphorylation. However, the exact mechanisms leading to NS protein-mediated inhibition of IRF3 remain to be unravelled. One of the objectives of this study was to identify amino acid domains in the human respiratory syncytial virus (HRSV) nonstructural proteins (NS) responsible for their ability to ablate the IFN-β signalling pathway. Furthermore, I wanted to find out at which level of this signalling pathway the NS proteins exert their suppressive activity and which are their major cellular targets. HRSV strains A2 and Long differ in their ability to block interferon type I synthesis. Sequence analysis of their NS proteins revealed the presence of an amino acid residue in the NS2 protein with a potential role for RSV IFN-inhibitory functions. Two recombinant bovine respiratory syncytial (BRSV) viruses harbouring HRSV NS1 and NS2 genes were generated and tested in their ability to restrict IFN-β synthesis. These recombinant viruses differed only in the identity of the residue at position 26 of the HRSV NS2 protein: rBRSVh1/2 has a Threonine as in the Long strain, while in rBRS h1/2*T26I this amino acid was mutated into an Isoleucin similarly to A2 virus. Sets of in vitro tests revealed that IFN-β induction was impaired by rBRSVh1/2*T26I when compared to rBRSV h1/2. Analysis of the transcriptional factors (AP-1, NF-kB and IRF3) involved in the activation of IFN-β synthesis provided evidence that the inhibitory ability of rBRSVh1/2*T26I was correlated to a selective block of IRF3. The mutation (T26I) in the NS2 protein did neither effect the NF-kB activation pathway nor perturbed the IFN-resistance characteristics of the chimeric viruses. IRF3 is activated upon phosphorylation mediated by IKK-related kinases (TBK1 and IKK-ε). TBK1 was therefore cloned from a human lung cDNA library and its biological activities regarding the induction of IFN-β were compared in mock-infected and infected cells. rBRSVh1/2*T26I and HRSV A2 precluded virus-induced IRF3 activation by interfering with TBK1 functions. No direct interaction between TBK1 and NS2 protein was demonstrated indicating that the kinase TBK1 may not be the sole target involved in RSV mechanisms of evasion of the innate immune response. Recombinant IFN-inducible rabies viruses expressing HRSV Long-derived (rGFP-Ph2/1) or HRSV A2-like (rGFP-Ph2*T26I/1) NS proteins were also generated. The HRSV NS2 protein expressing an Isoleucin (NS2*T26I) at position 26 was not able to suppress IFN-β induction and to rescue the growth of the recombinant eGFP-Ph2*T26I/1 in interferon-competent cells. A low expression of the mutated NS2*T26I protein was probably the reason of this failure. In summary, these results show that the HRSV NS2 protein possesses an intrinsic IFN-β inhibitory activity, which is achieved throughout a selective inhibition of the IRF3 activation pathway. The block appears to be exerted at the level of IRF3-kinase TBK1. Interferonantagonist functions of the HRSV NS2 protein are linked to a particular amino acid motif in the N-terminus of the protein. Identification of this amino acid domain and of TBK1 as the cellular target provide a better insight of how the HRSV NS2 protein prevents the establishment of the antiviral innate immune response and therefore it might contribute to the development of an effective vaccine.

HRSV ist eine häufige und weltweit verbreitete Ursache von Infektionen des Respirationstraktes. Es führt zu einer entzündlichen Erkrankung der respiratorischen Schleimhäute mit Mukosaödem, Hypersekretion und Bronchospasmus. Die Übertragung des viralen Erregers erfolgt durch Tröpfcheninfektion oder Kontakt mit kontaminierten Gegenständen. HRSV-Infektionen zeigen die höchste Inzidenz bei Säuglingen, vor allem in den ersten zwei bis sechs Lebensmonaten. Bei 25% bis 40% dieser Säuglinge nimmt die Erkrankung einen schweren Verlauf mit Befall des unteren Respirationstraktes in Form einer HRSV-Bronchiolitis oder -Pneumonie. Bei 0,5% bis 2,0% ist eine stationäre Behandlung im Krankenhaus erforderlich. Die Inzidenz nimmt wegen des zunehmend effektiveren Immunsystems mit dem Alter ab. Erwachsene und ältere Kinder zeigen meist keine Symptome bzw. Symptome einer leichten Erkältung. Reinfektionen im Laufe des Lebens sind häufig. Eine effektive kausale Therapie bei HRSV-Infektionen steht derzeit nicht zur Verfügung. Bei Patienten mit leichtem Krankheitsverlauf ist keine spezielle Behandlung erforderlich, therapiert wird symptomatisch. Aktuell ist keine spezifische Prävention in Form einer aktiven Impfung oder als effektive antivirale Therapie etabliert. Angesichts der hohen Inzidenz von HRSV-Infektionen und -Reinfektionen sowie der enormen gesundheitlichen und wirtschaftlichen Auswirkungen ist ein effektiver Impfstoff gegen HRSV als Forschungsziel vorrangig. Das Genom von HRSV, das zur Ordnung der Mononegavirales gehört, besteht aus einem negativ-orientierten RNA-Einzelstrang mit einer Länge von 15 222 Nukleotiden (beim A2-Stamm) und kodiert für zehn subgenomische mRNAs in der Reihenfolge 3’-leader, NS1, NS2, N, P, M, SH, G, F, M2(1+2), L, trailer-5’, die zur Expression von elf viralen Proteinen führen: fünf RNP-assoziierte Proteine, das sind das Nukleoprotein N, das Phosphoprotein P, die große katalytische Untereinheit L der RNA-Polymerase und der Transkriptionselongationsfaktor M2-1 sowie das nicht essentielle M2-2-Protein; vier Hüllproteine, dazu zählen das nicht-glykosylierte Matrixprotein M und drei Oberflächenproteine, im Einzelnen das Fusionsprotein F, das Anheftungsprotein G und das kleine hydrophobe Protein SH; zwei Nicht-Strukturproteine NS1 und NS2. NS1 und NS2 zeichnen die Pneumoviren vor allen anderen Viren der Ordnung der Mononegavirales aus. Beide NS-Proteine sind im Virion nur in Spuren nachweisbar, während sie in infizierten Zellen akkumulieren. Die beiden für die Proteine NS1 und NS2 kodierenden, nichtüberlappenden Gene liegen am 3‘-Ende des Genoms direkt im Anschluss an die leader-Region. NS1 und NS2 stimmen in den vier carboxyterminalen Aminosäuren überein, ansonsten weisen sie keine Sequenzähnlichkeiten auf. Das NS1-Gen hat eine Länge von 552 nt und kodiert für ein leicht saures Protein von 139 AS und 15,7 kD. Das NS2-Gen ist 503 nt lang und kodiert für ein basisches Protein von 124 AS und 14,7 kD. Die für die Ordnung der Mononegavirales charakteristische progressive Attenuation der Transkription sowie die Genlokalisation von NS1 und NS2 am 3‘-Ende lassen auf die höchste Transkriptionsrate für NS1- und NS2-mRNA unter den zehn HSRV-mRNA schließen, was auf eine bedeutende Rolle der NS1- und NS2-Proteine in infizierten Zellen hindeutet. NS1 und NS2 antagonisieren im Zusammenwirken die durch alpha-IFN und beta-IFN induzierte antivirale Antwort des Wirtsorganismus. Hierfür ist eine Koexpression beider NS-Proteine unbedingt erforderlich, ein NS-Protein allein zeigt keine derartige Aktivität. Der Mechanismus, mit dem HRSV die IFN-Antwort des Wirtsorganismus umgeht, ist unklar. In dieser Arbeit wurde die Funktion der NS-Proteine von klinischen HRSV-Isolaten aus fünf bis fünfzehn Monate alten Kindern untersucht. Durch die Anzucht der klinischen HRSV-Isolate in HEp-2-Zellkultur unter identischen Bedingungen wurden zunächst patientenabhängige Faktoren ausgeschaltet und damit die Grundlage für die Vergleichbarkeit der Wachstumseigenschaften der Isolate geschaffen. In den daraufhin erstellten Wachstumskurven konnten deutlich voneinander abweichende Wachstumverhalten der Isolate aufgezeigt werden. Der Befund, dass 3/4 der Bronchiolitis hervorrufenden HRS-Viren hohe infektiöse Titer (>106 infektiöse Viruspartikel/ ml an Tag 3) erreichten, während dies nur bei 1/3 der Bronchitis verursachenden Viren zu beobachten war, könnte auf eine Korrelation zwischen Wachstum in vitro und Pathogenität in vivo hindeuten. Um dies zu belegen, müsste eine größere Zahl von klinischen Isolaten analysiert werden. Die beiden Nicht-Strukturproteine versetzen HRSV in die Lage, die antivirale IFN-Antwort der Wirtszelle zu umgehen. Durch Behandlung von Virus-infizierten Zellkulturen mit IFN ließ sich nachweisen, dass alle klinischen HRSV-Isolate die Eigenschaft der IFN-Resistenz gleichermaßen besitzen und erst durch unphysiologisch hohe IFN Dosen eine wesentliche Inhibierung der Virusreplikation erreicht werden kann. Die in gleicher Weise ausgeprägte α-IFN-Resistenz bei den in Virulenz und Wachstumsgeschwindigkeit unterschiedlichen Viren deutete bereits darauf hin, dass diese Resistenz essentiell für alle klinischen RSV-Isolate ist, und dass zusätzliche Faktoren für das Maß der Aggressivität der Erreger verantwortlich sind. Mittels Nukleotid- und Aminosäuresequenzanalysen von NS1 und NS2 konnte dies weitgehend bestätigt werden. Anhand von RNA aus den HRSV-Isolaten wurde mit Hilfe des Enzyms Reverse Transkriptase cDNA von NS1 und NS2 synthetisiert, die nach dem Prinzip der PCR in vitro amplifiziert wurde. In anschließenden Klonierungsarbeiten wurden aus dem Vektor pBluescript II SK (–) und NS1-DNA bzw. NS1+NS2-DNA als Insert Plasmide konstruiert, in denen die Gensequenzen von NS1 und NS2 ermittelt und rechnergestützt in die entsprechenden Aminosäuresequenzen translatiert wurden. Die Analyse der NS-Sequenzen zeigte eine überraschend hohe Konservierung. Die Isolate waren einschließlich des Long-Stamms diesbezüglich untereinander sehr ähnlich. Diese Beobachtung stimmt mit der IFN-Resistenz überein und zeigt die Bedeutung der NS-Proteine. Die Ergebnisse dieser Arbeit deuten darauf hin, dass Abweichungen in den Sequenzen der übrigen Gene sowie patientenbezogene Faktoren wie Abwehrlage und anatomische Beschaffenheit des Respirationstraktes als Grund für die Unterschiede im Schweregrad der HRSV-Infektion eine Rolle spielen. Angesichts der stabilen Koexpression beider Nicht-Strukturproteine und des dadurch bedingten effektiven IFN-Escape sichern die Gene NS1 und NS2 die Überlebensfähigkeit von HRSV in vivo und stellen ebenso geeignete wie interessante Angriffspunkte in der Entwicklung eines attenuierten Lebendimpfstoffs dar.

Among the Paramyxoviridae, only members of the subfamily Pneumovirinae like Respiratory Syncytial Virus (RSV) encode two nonstructural proteins, NS1 and NS2. These two proteins cooperatively mediate type I interferon resistance and prevent induction of interferon in infected cells. Interactions of NS1 and NS2 proteins in every combination were shown by using the yeast-two-hybrid system. Therfore, NS1 and NS2 are able to form homo- and hetero(oligo)mers in infected cells. Although RSV replicates exclusively in the cytoplasm, NS-Proteins are localized in the cytoplasm as well as in the nucleus. Expression of an enlarged NS1 fusion protein, EGFP-NS1-BRSV N, resulted in the same nuclear and cytosolic localisation indicating that nuclear localisation is not due to diffusion but rather to an active transport. Thus, NS-Proteins should have particular functions in the nucleus of infected cells. Furthermore, yeast-two-hybrid screening of a lung cDNA expression library using NS1 of bovine RSV (BRSV) as a bait, identified cDNA clones encoding several nuclear proteins and one cytosolic protein. BRSV NS2 protein and NS-Proteins from other pneumoviruses (HRSV, PVM) were also able to interact with the identified cellular proteins in yeast. The isolated cDNAs encode the nuclear proteins CDK4BP (p34SEI-1 or TRIP-Br1), RanBP16, MM-1, DEAD Box Helicase p68 and the cytosolic β-COPI. Specific interactions were determined by mutational analysis of BRSV NS1 in yeast. Co-immunoprecipitation from lysates of eukaryotic cells confirmed the interaction of both BRSV NS-Proteins with the cellular proteins. The interaction of MM-1 and p68 with both NS-Proteins was also shown in GST pull down assay in vitro. Engineered BRSV encoding a truncated p68 showed accelerated replication in MDBK and Vero cells, whereas growth of NS1/NS2 deletion mutants expressing the truncated p68 was unaffected. This indicates that the presence of NS-Proteins is a prerequisite for the acceleration of BRSV growth by truncated p68. Furthermore, replication of BRSV was attenuated on HeLa cells in which expression of p68 was knocked down by specific siRNA, whereas replication of the unrelated Rabies virus was not. Thus, p68 is a nuclear target protein for the NS-Proteins and supports BRSV replication in vitro. Growth and division of host cells is necessary for optimal BRSV replication and like p68, most of the identified nuclear protein interactors are related to regulation of the cell cycle and cell division, respectively. Therefore, NS-Proteins appear to influence the cell cycle for optimal replication of BRSV by targeting such proteins. Hence, with the yeast-two-hybrid system, the first cellular interaction partners were identified indicating new functions of NS-Proteins in the viral replication cycle.

Bovine respiratory syncytial virus (BRSV), family Paramyxoviridae, subfamily Pneumovirinae, is a major etiological agent of respiratory tract disease in calves. A peculiarity of pneumoviruses is the presence of two unique nonstructural protein genes, NS1 and NS2. We show that both BRSV NS proteins are necessary and sufficient to mediate IFN resistance. This function can also be transferred to another, unrelated virus, Rabies virus. The mechanism of IFN resistance is conserved among the Pneumovirinae since the NS proteins of human RSV (HRSV) and of Pneumoniavirus of Mice (PVM) are also able to mediate IFN resistance in a similar manner. Furthermore, both BRSV NS proteins are able to block the induction of IFN by inhibiting the activation of interferon regulatory factor 3 (IRF-3). As a result, BRSV is able to block the establishment of a cellular antiviral state. The characterization of interactions between virus (NS proteins) and the host cell (IFN system) will have important implications in the development of HRSV/BRSV vaccines
