Abstract: Learning to associate a cue with its aversive consequence enables animals to prevent harm. Associative conditioning relies on activity within the cerebellum, but little is known about how changes in synaptic inputs to its output neurons shape their membrane potential dynamics during learning. Here we studied these changes in inputs using whole-cell and juxtasomal recordings of cerebellar nucleus neurons in awake mice during eyeblink conditioning and doing post-learning structural analysis. We identified changes in mossy fiber- and Purkinje cell-, but not climbing fiber-, inputs, leading to changes in subthreshold processing that correlate with the level and direction of the conditioned eyelid movements. These changes appear sufficient, as optogenetic stimulation of the inputs evoke incremental eyelid closures over the course of conditioning. Our data reveal how electrophysiological and structural modifications at the level of synaptic inputs to cerebellar output neurons facilitate conditioning of eyelid movements. Abstract: There is a profound lack of electrophysiological data from the cerebellum in humans, as compared to animals, because the pervading perception is that it is difficult to record human cerebellar activity non-invasively using MEG or EEG. We used on-scalp magnetoencephalography based on optically pumped magnetometers to detect learning related cerebellar signals in a classical eyeblink conditioning paradigm. In four healthy human adults, we observed unconditioned stimulus-related evoked responses locating in the cerebellum. These evoked responses diminished during the acquisition of conditioned responses, which corresponds with previous observed changes of Purkinje cell activities in animals. We also observed evoked responses immediately before conditioned blinks in 3 out of 4 participants, which were also located in the cerebellum. We discuss the potential nature of these cerebellar evoked responses and future applications of OP-MEG to the studies of human cerebellar physiology. Copy rights belong to original authors. Effect of Diverse Temporal Recoding of Granule Cells on Pavlovian Eyeblink Conditioning in The Cerebellum

Abstract: We consider the Pavlovian eyeblink conditioning (EBC) via repeated presentation of paired conditioned stimulus (tone) and unconditioned stimulus (airpuff), and investigate the effect of diverse temporal recoding of granule (GR) cells on the EBC in a cerebellar network by varying the connection probability pc from Golgi to GR cells. For an optimal value of pc* (=0.029), individual GR cells exhibit diverse spiking patterns which are well- or ill-matched with the unconditioned stimulus. Then, these diversely-recoded signals via parallel-fibers (PFs) from the GR cells are effectively depressed by the error-teaching signals via climbing fibers (CFs) from the inferior olive. Synaptic weights at well-matched PF-Purkinje cell (PC) synapses of active GR cells are strongly depressed via strong long-term depression (LTD), while practically no LTD occurs at ill-matched PF-PC synapses. This kind of "effective" depression at PF-PC synapses coordinates firings of PCs effectively, which then exert effective inhibitory coordination on cerebellar nucleus neuron [which evokes conditioned response (CR; eyeblink)]. When the learning trial passes a threshold, acquisition of CR begins. In this case, the timing degree Td of CR becomes good due to presence of ill-matched spiking group which plays a role of protection barrier for the timing. With further increase in the trial, strength S of CR (corresponding to the amplitude of eyelid closure) increases due to strong LTD in the well-matched spiking group, while its timing degree Td decreases. In this way, the well- and the ill-matched spiking groups play their own roles for the strength and the timing of CR, respectively. Thus, the (overall) learning efficiency degree Le (taking into consideration both timing and strength of CR) for the CR increases with learning trial, and eventually it becomes saturated. By changing pc from pc*, we also investigate the effect of diverse spiking patterns of GR cells on the EBC. Plots of both the diversity degree D of spiking patterns of GR cells and the saturated learning efficiency degree Le* of CR versus pc are found to form bell-shaped curves with peaks at pc*, and they also have a strong correlation. Consequently, the more diverse in temporal recoding of GR cells, the more effective in motor learning for the Pavlovian EBC. Copy rights belong to original authors. Visit the link for more info

You must be a member of the group.Chat is very welcome over in the group.Done and DustedMy project for the #STSICAL was knit using 40% Alpaca/40% Merino/20% Silk from John Arbon that I spun as a worsted 2ply to get a heavy sock/4ply weight yarn. I got 740m from 216g. I made Eyeblink by Heidi Alander. Decided to unpick the bind off and the last wrong side row. I then knit 8 rows and then did a beaded bind off using the technique in this video, by YarnFairy. I like how this technique means there are no holes under the picots.  I used Toho Metallic Iris Purple size 6/0 beads from JillyBeads.Eclipse by Lara Smoot using Yeomans Yarn Sport in the Birch colourway and The Yarn Yard Clan in pink.Refraction Shawlette Plus in DyeForYarn Sock BFL Superwash in the Wooden Treasure Chest colourway. The centre panel is made using some of the charts from Refraction Shawlette, with a different final chart.Regia Snowflake Color Socks in the 07709 colourway.Superwash Merino fibre, from World of Wool, that I dyed using Wilton's Food Dyes, I spun it as a worsted 2-ply and the resulting yarn was 282m/308yds of sport weight yarn.KraftyKoala merino silk fibre, was spun as a worsted chain-ply yarn. It measures 124m/136yds of a worsted weight yarn. Fibre was bought in a de-stash and I don't think she is dyeing anymore.A LOT of blocked squares for my Sock Yarn Blanket, which is based loosely on Shannon Geddes' Sock Yarn Square.Nose to the GrindstoneKnitting Goddess Socks, my usual vanilla pattern using The Knitting Goddess 4ply merino nylon that I got at Fibre East 2014. I don't think she uses this base anymore.The Doodler by Stephen West was the 2015 MKAL. I am using Countess Ablaze Lady Persephone Sock in the Persia colourway, Eden Cottage Yarns Pendle in Steel and The Yarn Yard Clan in Waverley. I referred to a great set of tutorials by jkimisyellow, which were really helpful.I've been prepping some Heavenly Wools New Zealand Halfbred Wool Roving in the Dragon's Breath Over Natural Grey. I have 200g and want to spin it as a true 3-ply. I have run out of yarn!Hobbledehoy Merino Tencel in the Lagoon colourway. It's been spun as a worsted 2-ply and I'm currently plying. I used my new spindle from AaronMakesStuff.PlottingMaking a Hitofude for my Mum using Artesano Alpaca Silk 4ply in the Lily Pad colourway. The bag I showed was from Shop Louleigh.My project for the #STSICAL is going to be knit using 40% Alpaca/40% Merino/20% Silk from John Arbon that I spun as a worsted 2ply to get a heavy sock/4ply weight yarn. I got 740m from 216g. I plan on making either Eyeblink by Heidi Alander or Wintermute by Melanie Berg.Crafting Library2015 Autumn Essentials: Flatland by Katya Frankel. This is from the Glow Series.I also got Bye Brook Farm 100% Romney Batt from Sarah of FiberTrek ,Woolweb Farm Navajo Churro yarn and Starcroft Fiber Nash Island Light in the Lobster Bake colourway from my friend Debs.Online CommunityLara Smoot has kindly offered to give away a copy of one of her most recently released patterns, Journey to Atlantis, which was previously an exclusive club pattern on Miss Babs Knitting Tour. It is kind of like a kids version of TMZ or Entertainment Tonight mixed in with a little Wayne's World. Max recaps the week in entertainment and gives his take and review of what he liked and didn't like on TV, at the movies, video games, in books and in music and even restaurant reviews.
