Building a house begins with the foundation, its longevity depends on the foundation strength. With cutting tools, this foundation is the material. There are various cutting materials such as cemented carbide, PCD, HSS, ceramics and more. Each type contains different grades. Through history, the introduction of each material and its use has led to a significant change in performance. The last century was marked by the progress of material technology and now, we see fewer new solutions. Does this mean development of new tool materials has already peaked and is experiencing stagnation? No, it is simply that new developments are deep within the cutting material and are focused on its structure. This can be observed only with the help of scanning electron microscopy (SEM), X-ray diffraction (XRD), electron backscatter diffraction (EBCD), and other sophisticated methods. They cover a tremendously complicated world of coatings that is extremely diverse. The most common available cutting material today is cemented carbide, primarily coated. In terms of performance, it represents a balance between efficiency, tool life and cost. Cemented carbide is known also as ‘tungsten carbide’ or simply ‘carbide’. A combination of cemented carbide, coating, and post-coat treatment produces a carbide grade. Only one of these components, the cemented carbide, is an essential element in the grade. Cemented carbide comprises hard carbide particles cemented together by binding metal (mainly cobalt). Most cemented carbides integrate wear-resistant coatings. New developments in cemented carbide are concentrated in three directions, carbide production, advanced coating methods and innovative post-coating techniques. Considerable success has been achieved in each direction. This is reflected in the wealth of new products introduced to the market. Customers analyse grades using parameters such as productivity, tool life and performance. In upgrading carbide grades, ISCAR is very sensitive to the challenge faced by industry. In this context, ISCAR’s solutions are developed considering the trends of modern metalworking. For example, titanium and superalloy use has increased. The growing usage demands technological solutions. The new tools require an appropriate foundation, made of advanced materials to achieve the desired cutting geometry. And for the construction of this foundation, ISCAR offers its new effective ‘bricks’ – upgraded carbide grades developed by metallurgists in the last few years. In milling, ISCAR has developed PVD coated IC882 and CVD coated IC5820 grades– two chocolate-color carbide grades for cutting titanium, high-temperature alloys and stainless. An integral component is a post coating treatment that facilitates longer tool life, due to increased resistance to chipping, notch wear, and build-up edge formation. The IC882 grade demonstrates impressive performance when machining conditions are hard, and the advantages of the IC5820 are most fully manifested when applied to milling with pinpointed high-pressure coolant supply. The PVD coated carbide grade IC806 was designed especially for turning high-temperature superalloys. As a result of the high mark that the grade received from the manufacturers of aerospace components, ISCAR expanded the application range of IC806 for laydown threading inserts. In addition to cemented carbide, industry consumes other hard cutting materials such as ceramics, diamond and cubic boron nitride (CBN). Their application on difficult-to-cut materials facilitates considerable increases in productivity. In recent years, ISCAR has enriched its cutting material range by introducing several new non-carbide grades, including the SiAlON grades IS25 and IS35 for cutting high-temperature alloys and the CBN grade IB20H for hard part turning. At the same time, customer demand is not limited to the effective machining of exotics. Steel is still the main structural material and ISCAR’s last innovation was in the parting and grooving realm - two new PVD coated carbide grades, IC1010 and IC1030. These were utilised in inserts from the TANG-GRIP and DO-GRIP families (Fig. 2). The grades are intended for machining stainless steel and steel. While the hard submicron grade IC1010 is recommended for productive parting and grooving with high cutting speed, the tough IC1030 is more suitable for interrupted cut and unstable machining conditions. Upgrading grades is an essential component for tool manufacturers’ success, and innovations should have a strong foundation. This why ISCAR’s motto ‘to be on the upgrade’ guides and inspires new tool material developments.

Cryo-electron microscopy (cryo-EM), a powerful biophysical technique that can analyse molecular assemblies, is changing the paradigm in structure-based drug design. As targets become more challenging, solving structures of drug target molecules or biologics becomes increasingly difficult. Cryo-EM empowers drug discovery teams to take advantage of rational drug design for many more major drug target classes, opening a pathway to best-in-class drugs. Cryo-EM allows the structural analysis of protein complexes flash-frozen in their near native states. One can now directly visualise not just large macromolecules, but also smaller proteins complexes, including membrane proteins. This powerful technique can be used to complement traditional methods, such as X-ray crystallography (XRD) or nuclear magnetic resonance (NMR) for structure-based drug discovery. Background: In recent years magnesium alloys have been intensively investigated as potential resorbable materials with appropriate mechanical and corrosion properties. Particularly in orthopedic research magnesium is interesting because of its mechanical properties close to those of natural bone, the prevention of both stress shielding and removal of the implant after surgery. Methods: ZEK100 plates were examined in this in vitro study with Hank's Balanced Salt Solution under physiological conditions with a constant laminar flow rate. After 14, 28 and 42 days of immersion the ZEK100 plates were mechanically tested via four point bending test. The surfaces of the immersed specimens were characterized by SEM, EDX and XRD. Results: The four point bending test displayed an increased bending strength after 6 weeks immersion compared to the 2 week group and 4 week group. The characterization of the surface revealed the presence of high amounts of O, P and Ca on the surface and small Mg content. This indicates the precipitation of calcium phosphates with low solubility on the surface of the ZEK100 plates. Conclusions: The results of the present in vitro study indicate that ZEK100 is a potential candidate for degradable orthopedic implants. Further investigations are needed to examine the degradation behavior.

This work is focused on the investigation of synthesis conditions for the direct growth of metal-organic frameworks (MOFs) on self-assembled monolayers (SAMs) used for surface functionalization. The characterization of the surface-grown crystals is a central part of this thesis, in order to learn more about the mechanism of MOF crystallization and the interesting properties of this class of materials leading to future applications. The tuneable oriented growth of thin films of the porous MOF HKUST-1 (Cu3(C9H3O6)2(H2O)3•xH2O) on different functionalized SAMs is demonstrated. Films grown on carboxylate-terminated SAMs are highly oriented along the [100] direction, while alcohol-terminated surfaces induce a completely different orientation along the [111] direction. Homogeneous but less oriented thin films are also obtained on methyl-terminated SAMs. A combination of X-ray diffraction and scanning electron microscopy was used to study the film growth, including the morphological evolution of the crystals on the SAMs. In situ atomic force microscopy was used to directly investigate the growth processes of the oriented MOF HKUST-1 grown on SAMs on gold substrates. This approach provides direct evidence for a layer-by-layer mechanism of the constituent 1.5 nm d111 crystal spacing step, its form during growth, and the influence of the step vertex on the rate of growth. The orientation and structure of porous MOFs based on iron terephthalate can be controlled by heterogeneous nucleation on self-assembled monolayers of mercaptohexadecanoic acid (MHDA). The framework MIL-53 is the product of homogeneous nucleation, whereas in the same crystallisation solution, oriented MIL-88B grows on the functionalized gold surface. A new sample cell was developed to allow for the investigation of the structural changes of flexible, porous materials during adsorption and desorption of guest molecules. Crystals of the flexible MOF Fe-MIL-88B were investigated as bulk material as well as surface-grown, oriented crystals. We were able to follow the structural changes of the Fe-MIL-88B crystals upon ad- and desorption of water. Due to the orientation of the crystals on the gold substrates, structural changes in [001] direction could be observed. For the randomly oriented bulk crystals the structural changes in all crystallographic directions are observable and the changes of the lattice constants a and c and the cell volume could be determined quantitatively by indexing of the complete diffraction patterns. The sorption isotherms recorded with the help of the sorption@XRD method show two distinct steps of structural changes. During adsorption of water the lattice parameter c is slightly but still detectably decreased, whereas the lattice parameter a shows a prominent two step-increase resulting in an increase of the cell volume about 40 %. The conventional volumetric water sorption measurement also shows two distinct steps of the amount of adsorbed volume that can be correlated to the structural changes observed in X-ray diffraction. We have demonstrated that advantages of functionalization can also be realized in oriented film synthesis of MOFs: We have shown for the first time that functionalized MOFs can be grown on self-assembled monolayers and that a preferred orientation of the crystals is achieved. This was demonstrated for NH2-MIL-101 and to a lesser extent for NH-MIL-88B. Importantly for potential storage applications, it was also shown that the adsorption capacity of NH2-MIL-101 was not significantly reduced by the amino functionalization.
