Olfactory neuroscience and psychophysics remain years behind the other senses in part because rich datasets linking olfactory stimuli with their corresponding percepts, behaviors, and neural pathways underlying this transformation, remain scarce. Here we present a concerted effort to unlock and unify dozens of stimulus-linked olfactory datasets across species and modalities under a unified framework called Pyrfume. We present examples of how researchers might use Pyrfume to conduct novel analyses uncovering new principles, introduce trainees to the field, or construct benchmarks for machine olfaction. Abstract: Mapping molecular structure to odor perception is a key challenge in olfaction. Here, we use graph neural networks (GNN) to generate a Principal Odor Map (POM) that preserves perceptual relationships and enables odor quality prediction for novel odorants. The model is as reliable as a human in describing odor quality: on a prospective validation set of 400 novel odorants, the model-generated odor profile more closely matched the trained panel mean (n=15) than did the median panelist. Applying simple, interpretable, theoretically-rooted transformations, the POM outperformed chemoinformatic models on several other odor prediction tasks, indicating that the POM successfully encoded a generalized map of structure-odor relationships. Here, we generated the most comprehensive murine olfactory ethological atlas to date, consisting of behavioral responses to a diverse panel of 73 odorants, including 12 at multiple concentrations. These data revealed that the mouse behavior is incredibly diverse, and changes in response to odor identity and intensity. Using only behavioral responses, ~30% of the 73 odorants could be identified with high accuracy (>96%) by a trained classifier. Mouse behavior occupied a low-dimensional space, consistent with analyses of human olfactory perception. While mouse olfactory behavior is difficult to predict from the corresponding human olfactory percept, three fundamental properties are shared: odor valence is the primary axis of olfactory perception; the physicochemical properties of odorants can predict the olfactory percept; and odorant concentration quantitatively and qualitatively impacts olfactory perception. Abstract: Neural codes for sensory representations are thought to reside in a broader space defined by the patterns of spontaneous activity that occur when stimuli are not being presented. To understand the structure of this spontaneous activity in the olfactory system, we performed high-density recordings of population activity in the main olfactory bulb of awake mice. We found that spontaneous activity patterns of ensembles of mitral and tufted (M/T) cells in the main olfactory bulb changed dramatically during locomotion, including decreases in pairwise correlations between neurons and increases in the entropy of the population. Maximum entropy models of the ensemble activity revealed that pair-wise interactions were better at predicting patterns of activity when the animal was stationary than while running, suggesting that higher order (3rd, 4th order) interactions between neurons shape activity during locomotion. Taken together, we found that locomotion influenced the structure of spontaneous population activity at the earliest stages of olfactory processing, 1 synapse away from the sensory receptors in the nasal epithelium.
