Abstract: Patterns of activity across many neurons are fundamental units of neural computation. Two-photon holographic photostimulation allows both delivering input to, and imaging responses from, patterns or populations of neurons. However, to make this method an easily-deployable tool, simple methods are needed to robustly and stably express opsins and indicators in the same cells. Here we describe a bicistronic adeno-associated virus (AAV) that in transfected cells expresses both the fast and bright calcium indicator GCaMP8s, and a soma-targeted (st) and two-photon-activatable opsin, ChrimsonR. With this method, in the visual cortex of mice, stChrimsonR stimulation with two-photon holography drives robust spiking in targeted cells, and neural responses to visual sensory stimuli and spontaneous activity are strong and easy to measure. stChrimsonR is a good choice of opsin when a balance is needed between stimulation-laser activatability and avoidance of imaging laser activation. Abstract: There is no consensus on the best optogenetic tool for inhibiting neural activity. Lamprey parapinopsin (Lamplight) is a Gi/o-coupled bistable animal opsin that can be activated and deactivated by short and long wavelength light, respectively. Since native mechanisms of neuronal inhibition frequently employ Gi/o signalling, we asked here whether Lamplight could be used for optogenetic silencing. We show that brief short (405nm) and long (525nm) wavelength pulses repeatedly switch Lamplight between stable signalling active and inactive states, and that combining these wavelengths can be used to achieve intermediate levels of activity. We demonstrate that these properties can be applied to produce switchable and scalable neuronal hyperpolarisation, and suppression of spontaneous spike firing in the mouse hypothalamic suprachiasmatic nucleus. We show that expressing Lamplight in (predominantly) ON bipolar cells can photosensitise retinas following advanced photoreceptor degeneration, and that 405 and 525nm stimuli can produce responses of opposite sign in output neurons of the retina. Lamplight-driven responses to both activating (405nm) and deactivating (525nm) light can occur within 500ms. We conclude that Lamplight can co-opt endogenous signalling mechanisms to allow optogenetic inhibition that is scalable, sustained and rapidly reversible. Copy rights belong to original authors. Hey everyone welcome to the third episode for Life and Times of The Blind. This episode explores light theory and the cones biological structure.  Author Note:  I did leave out that the dominant opsin found in each of your cones is what actually determines what cone it is.  I meant to explain that but realized by the end I had forgotten.  So please keep that tidbit of information handy while you are listening.     

Opsins are the photosensitive proteins in the eye, responsible for converting a photons of light into an electro-chemical signals. Different opsins react to different wavelengths of light, each corresponding to a different band of colour. In humans, the 'visible spectrum' of light (a very anthropocentric term) is covered by three opsins, receptive to red, green and blue wavelengths. Other animals have opsins that are capable of subdividing the 'visible spectrum' and responding to a large number of very specific wavelengths of interest. All in all, the ability to detect light and recognise colour is not the same throughout the animal kingdom. In this episode, we are joined by Dr James Fleming of Keio University, Japan to discuss the evolution of opsins in the ecdysozoa (the group containing arthropods and a fair few worms). We talk about the fundamentals of light detection and how, using phylogenetics, we are able to tell which colours certain extinct animals were capable of detecting.

The International Institute for Species Exploration has selected it's "Top 10 New Species" from the approximately 18,000 new species named during 2014. The list includes cartwheeling spiders, feathered dinosaurs and strange multicellular organisms that could be an entirely new phylum - a new branch on the tree of life. Two separate studies have explored how octopuses and squid change their skin colour to rapidly camouflage themselves. They found that the skin (on squid and cuttlefish) and tiny hairs called cilia (on octopuses) have cells that are used in vision. In 2013 some research suggested that the blood from young mice can rejuvenate older mice. Well new findings cast doubt on those results, and things are a little more complicated. British and US scientists have published the first comprehensive map of genetic mutations linked to different strains of prostate cancer. They describe the map as "prostate cancer's Rosetta stone" and say it will guide future treatments and trials.
