As the famous frog once said, it's not easy being green. And when it comes to decarbonising industry, indeed, reducing emissions of all sorts, the task is a complex one. Fossil fuels are used to manufacture some of mankind's most ubiquitous products, from plastics to cement to steel; and even in areas where we're trying to improve our footprint, there are repercussions. Mining lithium for electric car batteries isn't exactly without impact. Add to the mix stories of corporations prioritising profits, and governments focusing on short-term popular policies – and it would be easy to feel disheartened.Professor Anna Korre says her role is to be the champion of science in this debate: providing clear evidence to help reduce environmental impacts, while allowing vital production processes to continue. Anna is an environmental engineer at Imperial College London and Co-Director of the university's Energy Futures Lab. Her work has led to a risk model that's now used in mining operations around the world – and her current research into sub-surface CO2 storage could hold the key to decarbonising British industry. Welcome to the Design Thinking podcast! I'm Dawan Stanford, your host. My guest today is Paolo Korre, Director of Service Design and User Insight for SE Health, a not-for-profit healthcare company in Ontario, Canada. We'll chat today about Paolo's transition from industrial design to design thinking in the healthcare space, challenges he faces as a designer working in healthcare, and his experience with being the lone designer on an innovation team. Paolo reached industrial design through a love of fine arts, drawing, painting, and crafts. He did his undergraduate degree in industrial and product design. He assumed he would be working on making furniture or other “stuff,” but soon realized that this isn't what the world really needs. As he evolved beyond traditional design, he went back to school for further education. Eventually, he was able to bring design thinking back home. In our conversation today, he'll dig into how his skills in design relate to his work in healthcare (and how he convinced people to give it a shot!). For example, he explains that he worked on a project around improving the experience of patients receiving private care. As a result, they ended up launching Elizz, a whole brand dedicated to supporting family caregivers. Paolo has experienced being a lone designer as well as being part of a team of designers and part of an innovation team. It's all part of a learning journey, he explains, and speaks of trying to figure out how much design method he can apply in these various roles. I år för sjunde gången. Två gånger har Thomas och jag gjort pilgrimsresan till content marketings Mecka. Två gånger har vi varit deltagare på Content Marketing World. Första gången 2014 och andra gången 2016. Båda gångerna rapporterade vi hem till dig. Men inte i år. Vi var inte där. Det var däremot Mattias Brännholm – tidigare USA-korrespodent för SVT och tv-producent vid OS i Osaka med mera, numera content marketing geek vd för Brama Content. Vi ringde Mattias och bad honom berätta om sina lärdomar och intryck från Content Marketing World. Story var temat för Content Marketing World 2017, och det syns tydligt i rapporter från konferensen. Plasminogen wird durch verschiedene Proteasen proteolytisch gespalten. Zu den Enzymen, von denen bekannt ist, daß sie Plasminogen an definierten Peptidbindungen prozessieren können, gehören Elastase aus polymorphnukleären Neutrophilen (PMN) und Metallo-Elastase aus Makrophagen. Ein Spaltprodukt ist Miniplasminogen, das die proteolytische Domäne und den Kringel 5 umfaßt. Die Spaltstelle bei Val441 ist charakterisiert und ein Sandwich-ELISA gegen die Neodeterminante ist etabliert. Das dabei entstehende Counterpart besteht aus den Kringeln1-4. Es wird Angiostatin genannt, weil es hemmend auf die Neubildung von Gefäßen wirkt. Im Rahmen dieser Arbeit sollte untersucht werden, ob Miniplasminogen unter pathologischen Bedingungen wie Sepsis, Peritonitis und Tumorerkrankungen auftritt, um daraus einerseits den Einfluß der Elastase abzuleiten und Einblicke in die pathophysiologischen Abläufe bei Entzündung und Tu-morgeschehen unter Betrachtung der Plasminogenspaltprodukte in Korre-lation zu anderen Parametern zu gewinnen. In Vorversuchen konnte gezeigt werden, daß unter dem Einfluß von aktivierten polymorph-nukleären Neutrophilen Miniplasminogen aus Plas-minogen generiert wird. Von den potentiell an der Proteolyse beteiligten En-zymen konnte in vitro nur bei dem Einsatz von PMN-Elastase Mini-plasminogen nachgewiesen werden, nicht jedoch von MMP-2, -8 und -9. In Untersuchungen zur Stabilität von Miniplasminogen unter verschiedenen Blutabnahmebedingungen waren Citrat-Proben den anderen Systemen (EDTAPlasma, Serum) überlegen. In Proben von gesunden Probanden konnte in keinem Fall Miniplasminogen über der Nachweisgrenze des ELISAs gemessen werden.Es wurden Proben aus klinischen Studien zu Sepsis, Peritonitis und Mammakarzinom ausgewählt, die auf Grund einer hohen PMN-Elastase-Konzentration ein Entstehen von Miniplasminogen erwarten lassen konnten. Ein Ausscheiden von Miniplasminogen über die Niere konnte durch Messungen im Urin ausgeschlossen werden. In Citratplasma-Proben von Peritonitispatienten war kein Miniplasminogen nachweisbar, in Peritonitisexsudaten desselben Patientenkollektivs waren Miniplasminogenwerte bis 90 ng/ml meßbar. Es zeigte sich allerdings keine Korrelation zu anderen Parametern (Elastase-Konzentration, Plasminogenkonzentration). Signifikante Unterschiede der Miniplasminogenkonzentrationen konnten zwischen den Mittelwerten der Proben der Patientengruppe, bei der therapeutisch Fresh-Frozen-Plasma intraabdominell appliziert wurde, und der Kontrollgruppe, sowie zwischen den Gruppen mit und ohne Tumorerkrankung nachgewiesen werden. Bei der Evaluierung von Serumproben aus einem Mammakarzinom-Kollektiv wurden Werte bis 52 ng/ml gemessen. Eine Korrelation mit anderen Parametern oder signifikante Unterschiede in den verschiedenen Subgruppen konnten auch hier nicht gezeigt werden. Ein Zusammenhang zwischen der proteolytischen Kapazität in den Exsudaten und der MPlg-Entstehung ließ sich nicht zweifelsfrei beweisen. MPlg ist daher – im Gegensatz zu dem Elastase-spezifischen Spaltprodukt des Fibrinogens (FEP) (Gippner-Steppert, 1991) - als ein spezifisches Spaltprodukt des Plg nicht für den indirekten Nachweis der proteolytischen Aktivität der PMNElastase geeignet. Erfolgversprechend könnten ggf. immunhistochemische Untersuchungen von Tumormaterial in Hinblick auf das lokale Entstehen von Miniplasminogen sein.
