Engineered Proteins Show Promise in Stopping Glioblastoma Invasion

Scientists have engineered small, targeted proteins that can penetrate brain cancer cells and prevent them from invading healthy tissue, offering a promising new approach to treating glioblastoma multiforme (GBM), one of the deadliest forms of brain cancer. This strategy was developed by researchers at the University of Nevada, Reno, and published recently in Oncotarget. The Challenge of Treating Glioblastoma Multiforme Glioblastoma is an aggressive and fast-growing brain tumor that infiltrates healthy brain tissue, making complete surgical removal nearly impossible. Standard treatments like chemotherapy and radiation can slow its growth but rarely prevent it from returning. One major reason for this invasiveness is a group of enzymes known as matrix metalloproteinases (MMPs), which break down surrounding tissue to allow cancer cells to spread. Among these, MMP-9 plays a particularly important role in driving tumor progression and resisting existing therapies. Attempts to block MMPs using small-molecule drugs have failed in clinical trials due to problems like poor selectivity and harmful side effects. Researchers have been searching for safer, more targeted methods to interfere with these enzymes and limit glioblastoma's spread. The Study: Engineered Proteins to Inhibit Tumor Invasion In the study called “Effect of TIMPs and their minimally engineered variants in blocking invasion and migration of brain cancer cells,” researchers Elham Taheri and Maryam Raeeszadeh-Sarmazdeh investigated tissue inhibitors of metalloproteinases (TIMPs), which are natural blockers of MMPs, and their engineered modified versions made to work better. Specifically, the team studied TIMP-1, TIMP-3, along with two engineered molecules, mTC1 and mTC3, in laboratory cell models of GBM. I've always been a fan of MMP. And I still am, but there's one thing about it that I do struggle with – the list MPs. I know it took some of us a while to totally understand why we have list MPs. I understand – but I don't necessarily like it. Which is why I thought retiring Labour MP David Parker raised some good points about MMP in his valedictory speech in Parliament. He reckons we should replace it with something else. His preference is the single transferable vote system, and he thinks we should have another referendum to see if people want to stick with the MMP voting system. I don't think we need a referendum because I think most people are happy with it. I am, anyway. Where I am with David Parker is list MPs. He didn't say straight out that he thinks they're a waste of time. That's probably because he's a list MP himself – or he was, anyway. But he thinks that, even if you're a list MP, you should be connected to an electorate. I know you see some list MPs driving around the place with signage on their vehicle saying “so-and-so, so-and-so, your such-and-such MP in wherever you might be”. Example: Tracey McLellan bills herself as a Labour list MP based in Banks Peninsula. But that's just optics. It's just done to make us think that a list MP is just as approachable and interested in their local community as someone specifically elected to represent an area. And I think list MPs have got a big sell job on their hands to convince us of the value they bring. I know they say they work frantically. But how does that stack up when we've got the likes of Prime Minister Christopher Luxon dealing with everything that comes with being PM – plus he's also the MP for Botany. So David Parker is right to be saying that list MPs need to do more than just meat in the room at Parliament when it comes to voting in a pack. I disagree with him though when he says MMP is to blame for polarising people. He says MMP has based politics in New Zealand on identity, but I thought that's what MMP is all about. Because identity politics is when you get a political view based on things like ethnicity, race, nationality, religion, denomination, gender, sexual orientation, social background, political affiliation, caste, age, education, disability, intelligence and social class. My response is: what's so bad about that? It's not as if we didn't have special interest political parties before MMP. We had Christian Heritage and the Christian Democrats. And the Values Party was pretty much an environmental political outfit. Even Social Credit had a particular identity –it wasn't just another mainstream political party. Another thing I disagree with is David Parker's view that MMP is bad because it polarises people. He says under the first-past-the-post system, New Zealand was one of the best countries in the world and that, with MMP, the place was meant to get even better. But he says it hasn't. He says politicians are more divided than ever and New Zealand society is more divided than ever. But I don't think MMP is to blame. People are polarised all over the world – more so after Covid. And there are many different voting systems all over the world. Nevertheless, David Parker thinks MMP has run its course and he reckons it's time to put it to the people and have another referendum to see if we're still happy with MMP, or whether we'd like to change to something else. Long serving Labour MP and former Minister David Parker gave his valedictory speech in Parliament last night, where he gave his colleagues across the House a list of things to do, among his thank yous and goodbyes. Chief among them was closing the gap between the very wealthy and the middle class. It could be done, he said, with a tax on capital income, a wealth tax, some form of interest deductibility ban with rules for deductions to avoid double taxation. He said this would pay for a tax-free threshold for income earners up to $10,000 with the next 10,000 subject to lower tax rates. Another was that he hoped freshwater standards would endure in some form, and called on future Ministers for the Environment not to become Ministers for Pollution. Looking at you, Shane Jones. Parker also suggested a way of future proofing New Zealand against future disasters by getting the Reserve Bank to use a quantitative easing scheme to purchase a long dated bond in the event the Alpine fault ruptures, which is not a bad idea because that would spread the cost of the disaster over generations, rather than have one generation deal with it. He wanted to see the government take on the tech giants with a proposal to make their social platforms liable for harmful content shared on their platforms. And he called out MPP as a political system that is becoming worse over time, that is fuelling culture war politics. “Under First Past the Post, New Zealand became amongst the best country in the world, but MMP was meant to be better. Perhaps Doctor Hooten is right and MMP gets worse over time. It's the people's system, not ours. As things polarise and the hard issues don't get fixed, we should allow the people to again make their choice. I'd vote STV. All 120 of us would have to serve in a seat - that drives behavioural change. I'd add in a small upper house, 30 people appointed as in Canada, or voted in STV and limited to two terms each.” That was David Parker last night in Parliament talking about the New Zealand he would have liked to have seen when he left politics. The thing is, the public have had their say and they have chosen and then reaffirmed MMP. My conscience is clear. Like David Parker, I preferred STV – that's what I voted for back in the 90s and I still think it's a better system today. I think he's right when he says that MPs need to have electorates to which they are accountable. And I think STV would be a fairer, less divisive system. First Pass the Post was undemocratic. There were times when New Zealand elected a government that only had around 38 - 40% of the vote, and 100% of the decision making, and that's not particularly fair. Some form of proportional representation is more representative, it's more democratic. If we're going to live in a democracy, we might as well behave as though we're living in a democracy and vote and get results as if we're living in a democracy. So from his to-do list, which would you like to see MPs pick up on? And specifically, when it comes to the voting system, I don't think we've got it right yet. There will be some of you who vote, who have grown up with the MMP and that's all you have known. As someone who knows First Past the Post and MMP, I think MMP is better than FPP and producing a more democratic and fairer result. Is it perfect? Nowhere near it. I think we need to keep refining it just because we've voted for it once, reaffirmed it once, doesn't mean we have to be stuck with it forever. See omnystudio.com/listener for privacy information.

David Seymour's calling for fat to be cut from multiple areas of government. The Act Party leader wants a limit on the number of ministers – with no ministers outside of Cabinet, and no associate ministers other than in Finance. He wants 30 total government agencies, down from 41. Executive Director of business think tank New Zealand Initiative, Oliver Hartwich told Mike Hosking the current model doesn't make sense. He says there's a Minister of Housing but also a Minister of Building and Construction, as if they have nothing to do with each other. Hartwich also says the Ministry of Business, Innovation and Employment is a prime example of a bloated cabinet. It is being reported as a scrap, which I'm not sure is accurate. But if it is, it is a shame and probably a lesson for the combatants, who are both supposed to be on the same side. But under MMP, is anyone on the same side? Act are concerned about ACC and their current desire to solve safety issues in the manufacturing sector. But by putting a race-based lens across it, ACC want people who have answers, or programmes, for injuries to Māori and Pasifika. This of course is not what the Government is supposed to be about and a Cabinet edict says so. The shame of this is both sides, in fact all three sides, are at one on the issue. They campaigned on the issue and they campaigned on it because the last Government got so obsessed with Māori issues and language and acquiescing to everything cultural that a large swath of voters got thoroughly sick and tired of it. So why we are here after a year-and-a-half and a Cabinet instruction is beyond me. The fact ACC, or any Government department, are still trying this on is the real problem. It goes to a theme we have highlighted too often this term and that is that a public service don't appear to be neutral, or operating under the instruction of the Government of the day, but rather to their own beat. There is no good news in two parties seemingly debating a formally agreed approach with each other and there is no good news in a department continuing to do something they shouldn't. We have enough to deal with at the moment without previously agreed approaches being re-litigated or disavowed. Obviously work safety is not a race-based problem. It is an industry or sector problem and ACC should know this. Even if they didn't, they should be following instruction. Act are on the right side of this. You've got to hand it to Winston Raymond Peters. On this his 80th birthday... he's jetting round the Pacific representing his country. Keeping us close to our friends in the region as China circles and America wanders. He's visited more than 40 countries since Luxon took over. The stamina. The discipline. The confidence. The work ethic... all things to be admired. Doesn't matter what side of politics you come from, hats off to you Winston. And Happy birthday. And what a contrasted pictures he paints in Parliament... One side of the house... draped in performative costume's... wearing badges and head pieces... and snapping their fingers... and doing TikTok videos.... And not bothering to learn the standing orders... Not bothering to turn up to work. Not bothering to engage in meaningful debate so that we might all get to a better place... you know... the essence of democracy. Winston is not perfect... he's flip-flopped and u-turned more times than you can count. He angered the right picking an inexperienced Labour party over the high-polling Bill English in 2017. Some will never forgive him for that. BUT. he's a man with a strategy. He's a man who doesn't mind hard work. He's a man whose politics you may not like, but who's character modern society cannot afford to lose or forget. He was born in 1945 - just a few weeks before Germany surrendered to the Allies in WWII. He's been in politics for nearly half a century. The Kingmaker... punching well above his weight in MMP system. In fact... a teacher of mine used to say the P in MMP was Peters. He's loves a bit of drama... he loves a fight... he's not above performative politics too, by the way.. remember the NO sign? Remember the Winebox inquiry? The donations sagas? BUT... and this is why kiwis keep giving him a chance... he takes seriously the things and people's he promised to represent... and he works day and night to get the political deals done to make them happen. As exercised as some have been this week about social media, the Greens and their behaviour, the bigger issue is not the Greens, but Labour and all who might support them into Government. As much as we may froth and hyperventilate about any given issue of the day or small firestorm, who runs the country and how is what we all ultimately should be focused on. We will not be going back to a single large party ever again, for two reasons. One is because Covid is not repeating and, even if it did, I doubt we would panic again electorally the way we did. Two is given when Labour was left to their own devices, they literally destroyed the place, most of us, for good or bad, have come to the conclusion that a mix of parties under this MMP system we seem to have voted for an accepted is here to stay. So National need at least one player, probably two, and the same applies to Labour. For now, National seem to have got lucky and/or well organised. Whether you support it or not, broadly speaking National, Act, and NZ First get along fine. The media try to stir bits of trouble around minor matters periodically and we are yet to see a full-blown campaign post a first term, where the gloves are off a bit and a few punches might get thrown. But overall, things are cordial and well managed. Labour on the other hand need certainly the Greens and possibly the Māori Party. Both are increasingly ropey. Both are increasingly belligerent, both are increasingly fringe and both are an electoral nightmare for a so-called mainstream, left-wing party. The Greens of James Shaw, Rod Donald, and Jeanette Fitzsimons are long gone. The Māori Party of Pita Sharples and Tariana Turia are long gone Both the current Greens and the Māori Party are anti-establishment disruptors who revel in a type of anarchy, almost as though they are outside the system. All of that is fine if you like that and want to vote for it. But it is completely incompatible to running a country, even for a Labour Party that has become increasingly left-leaning and socialist. If you don't need, or want, to be in power, and I don't think half the Greens or the Māori Party actually do, you can say what you want. But Labour do want power, and their problem is they are going to end up having to answer for an increasingly unhinged rabble or at least try to dress it up as something they can handle. Engineered TIMP Molecules Show Potential to Slow Glioblastoma Brain Cancer Spread

BUFFALO, NY - March 3, 2025 – A new #research paper was #published in Oncotarget, Volume 16, on February 28, 2025, titled “Effect of TIMPs and their minimally engineered variants in blocking invasion and migration of brain cancer cells." Elham Taheri and Maryam Raeeszadeh-Sarmazdeh from the University of Nevada, Reno, explored a new approach to slowing the spread of glioblastoma multiforme (GBM), the most aggressive and deadly form of brain cancer. Their study highlights the potential of both natural and engineered molecules to block cancer cell movement, offering a promising strategy to combat this challenging disease. Glioblastoma multiforme is difficult to treat because it quickly spreads into healthy brain tissue, making complete surgical removal nearly impossible. A major driver of this invasive behavior is a group of enzymes called matrix metalloproteinases (MMPs), which break down surrounding tissue and create space for cancer cells to spread. Among them, MMP-9 plays a particularly significant role in GBM progression and resistance to current treatments. To address this challenge, the researchers investigated tissue inhibitors of metalloproteinases (TIMPs), natural MMP blockers, and specially engineered versions designed for better effectiveness. The study used cell line models of GBM to test both TIMP-1 and TIMP-3 and their engineered counterparts (mTC1 and mTC3), specific blockers of MMP-9. “Our study focused on minimal TIMP variants, due to their small molecular size and potential in higher cellular uptake and delivery, to assess their potential in cell-based assays.” The results indicated that the engineered TIMPs were just as effective as, or even better than, the natural ones at reducing cancer cell migration and invasion. These findings are particularly promising because previous attempts to block MMPs with small-molecule drugs faced challenges such as poor selectivity and unwanted side effects. In contrast, these engineered TIMPs offer a more targeted and potentially safer approach. One of the greatest obstacles in treating brain cancer is delivering drugs across the blood-brain barrier, a protective layer that prevents many therapeutic compounds from reaching the brain. To address this, the researchers used cell-penetrating peptides to help the TIMP variants reach and enter cancer cells more effectively. Their results confirmed that the engineered TIMPs successfully reached tumor cells, further increasing their potential as a treatment. Additionally, the study found that these engineered TIMPs did not significantly affect healthy cells at lower doses, suggesting they could be used safely. This makes them strong candidates for further drug development. These findings could lead to new treatment options for GBM, a cancer with very few effective therapies. Future research will focus on testing these TIMP variants in animal models to evaluate their long-term effects and safety. Researchers also plan to investigate whether combining these engineered TIMPs with existing treatments, such as chemotherapy or immunotherapy, could improve outcomes. In summary, given the aggressive nature of GBM and the urgent need for better therapies, this study represents an important step forward. If further research confirms these results, engineered TIMPs could become a valuable tool in the fight against brain cancer, offering new hope for improved treatments and patient survival. Like most things in life, there is nuance and subtlety that is lost along the way. David Seymour is of a personality that undoubtedly gets up the noses of some. He might even bother the Prime Minister periodically. But his Land Rover Escapade is not a sackable offence. Neither is his letter written, not as a minister, for Polkinghorne a sackable offence. Even if you want to combine them and throw in the Treaty Principals Bill because he's agitated people with it, he is still not in sackable territory, nor indeed anywhere close. Here is the simple truth about MMP: why do we still report it like FPP and they're all in the same party? Could the Prime Minister sack David Seymour from Cabinet? I guess, but then what would happen? The end of the Government. Is he going to do that? No, he is not. When companies take over other companies there is often a clean out of talent. When a new CEO arrives the same thing often applies. The business of running a country in an MMP environment is unique. You don't merge or take over, you coalesce. You are individual entities who agree on a series of ideas and a level of cooperation. It won't go perfectly. It might not even go swimmingly, because at no stage did you ever merge into one. You always remained, in this case, as three. When Chris Hipkins calls yet again for a sacking —and surely we are bored witless with that tactic— he tells us that not since the 80's and Lange and Prebble have we seen in-fighting like this. He is of course wrong. He forgets Peters and Shipley, and Peters and Bolger, and Anderton and Clark, and Kopu and Shipley, and the NZ First Tight Five. He forgets a vast swathe of our local and recent history and, not just that, he forgets Prebble and Lange were in the same party. Seymour and Luxon are not. In many respects we are lucky with this current line up. In a small country coalition choice is limited. In Spain and Germany where they have recently stretched the bounds of credibility in forming deals, they have collapsed. This deal won't collapse. The majority of the time there is cordiality, respect, and productivity. But reportage doesn't appear to feature those aspects. The great frustration I have with the Hipkins approach and the coverage of this frippery is that this is a time of tremendous importance on serious matters. This country is a mess, and it is in desperate need of addressing. The side show game increasingly looks from another age and also childish. If Seymour, Peters, or Luxon is on the phone to the Governor General to dissolve arrangements then come back to me. Kiko Loureiro is a Brazilian Grammy award winning heavy metal guitarist, best known as a member of the bands Angra and Megadeth. His new solo album "Theory Of Mind" is a tour de force of strong melodies, incendiary guitar playing, and great arrangements!  Alongside his band Angra, he has released eight studio albums, received two gold records, and is considered one of the most important Brazilian musicians of all time. His solo career started with the album "No Gravity". His second album was "Universo Inverso", recorded in only three days, featuring some of the best jazz musicians in Latin America. Three years later, Kiko released, "Fullblast" , a heavy and progressive piece. In 2008 he also presented his fusion project: Neural Code, in which the band explores rock and jazz, fueled with Brazilian rhythms. "Sounds of Innocence", his fourth solo album, was released in 2012, combining metal, jazz and Brazilian rhythms. The year 2015 came with big news: Kiko was invited to join Megadeth and recorded the guitars for Megadeth's fifteenth studio album "Dystopia" It was in early 2017 that he got the greatest recognition in his career. He became the first Brazilian to receive a Grammy Award by playing in a rock band. Megadeth got the award for "Best Metal Performance" with the album "Dystopia." Open Source released in July 2020 presents new musical ways in Kiko's compositions. The good news about the arrival of the protest is that after today, it's over. This has been one of the more misrepresented activities in recent years. It's hard to know what it is actually about, or whether those taking part even know what it's about. Is it about the Treaty Principles Bill? Is it about a general attitude towards the Government and their Māori policies? Is it a PR stunt for the Māori Party? And perhaps most importantly of all - what does it achieve? The answer, of course, is nothing. If it is about the bill, it dies after the select committee process and, even if it didn't, although you are more than justified to protest, a protest doesn't change the right of a Government to do what they promised they were going to do. It's called democracy. Democracy is slightly complicated by MMP, which may or may not produce what you thought it might. But democracy was what led to MMP in the first place because we voted for it. The two winners out of this so far are the Māori Party, who have got a lot of attention and drummed up good numbers in various parts of the country for their campaign. They have of course alienated themselves from many, but they don't care, and you were never voting for them anyway. The other is David Seymour who, whether you agree with him or not, has argued coherently and eloquently. Not performing so well includes the media who, yet again, have sided too often with the protest. I saw a reporter literally singing as part of her reportage. I saw a reporter dispute official figures from the police for the turn out, as though a dispute is fact. Jenny Shipley talking of civil war was as astonishing as anything. At least Chris Finlayson, in weighing in, kept his head. In a country with real and serious problems, a bill destined for the waste bin is not worth the energy this has been given. Our priorities are all over the place. We have looked shabby, unhinged, violent, and low rent. We are not living up to expectation, or our potential. This is not the country I grew up in, or indeed love. It is a wayward place in desperate need of some serious leadership and even more discipline. The Treaty Principles Bill gets debated this week, and then off it goes to the committee that will hear a lot of fear mongering and whinging about how it is the end of the world. After that it will come back to Parliament, a vote will be had, it won't get the numbers, and it will be dead. This is unusual as Governments normally put bills in they know they are going to pass. Here is why it ties into Trump's victory last week: Trump won because he is the repository for grievance. If you don't like the left you vote for what's not the left, because you only have a choice of two. Under MMP you have more choices. We have taken a lot of time to work that out, but as MMP matures we are reaching a place where more parties will establish themselves as ongoing contributors to the system, as opposed to being seen as fringe and on the verge of survival. This Government will be especially helpful if it performs well as a group, gets re-elected and possibly goes for a third term, because it will show three parties can coalesce and agree to disagree, while remaining separate and independent. The Greens have already arrived at this place. They are a permanent fixture on the landscape, and they don't dabble with 5%. The point here is, as a result, the big parties will shed support. The days of National being 45%-ish are gone because parties like ACT and New Zealand First look to head towards 10%, if not more. The Treaty bill might well be ACT's ride to permanence. The same way Trump hoovered up blacks and Hispanics who were sick of being treated like a block and not individuals, ACT could hoover up New Zealanders sick of race-based policy. If National don't, or won't deal to it, ACT can. In an MMP environment Trump would not have stood a chance. But their system is less sophisticated than ours. Choice is good. It gets over-represented in jurisdictions where the threshold is too low because you end up with single issue nutters. But at 5% it looks like we have picked it right because you can be small without being too fringe. Hopefully ACT and New Zealand First break the MMP hoodoo where small parties vanish in Government because there are enough specific issues and enough electoral confidence for parties to stake clams and build support. Big parties no longer have to be everything to everyone. If this is an emerging trend, MMP will have properly arrived, and we will all be better off for it.
