La Dra. Nidia Paulina Zapata Canto, hematóloga adscrita al Instituto Nacional de Cancerología en la Ciudad de México, México, tiene como invitada especial en este episodio de “Preguntémosle al experto…” a la Dra. Gabriela Sanchez-Petitto, hematóloga en The Ohio State University Comprehensive Cancer Center en Ohio, EE.UU, para abordar un caso clínico sobre trasplante haploidéntico de médula ósea. Las expertas responderán con base en evidencia científica y en su experiencia a varias interrogantes. El caso trata de un hombre de 33 años con tabaquismo actualmente suspendido y consumo de marihuana ocasional. En octubre de 2021 inició con un cuadro de dos meses de evolución, con astenia, adinamia, petequias y equimosis. La biopsia aspirado de médula ósea reportó una leucemia aguda mieloblástica; se realizaron diferentes perfiles, los cuales se reportaron negativos y el cariotipo sin crecimiento. Se inició tratamiento con un esquema de inducción (7+3), logrando al final del tratamiento una remisión completa y como complicación una infección de tejidos blandos. Se dieron cuatro consolidaciones con citarabina, se envió a trasplante para evaluación y en el seguimiento el paciente presentó una recaída. Debido a ello, el paciente tuvo una reinducción con un régimen FLAG-VIDA, logrando una segunda remisión completa y así iniciando un esquema de mantenimiento con azacitidina. En noviembre de 2022 se realizó el trasplante alogénico de médula ósea, donde tuvo un síndrome de liberación de citocinas. Al egresar de la unidad de trasplante, presentó dolor abdominal, por lo que se le realizó una colecistectomía por colecistitis aguda. En febrero de 2023 inició mantenimiento con azacitidina y en mayo del mismo año se realizó el último aspirado. Se reportó un quimerismo al 100% con una enfermedad mínima residual no detectable. Con base en este caso, las preguntas para la experta son las siguientes: ¿Cómo se consideran las hipodiploidías para clasificar el pronóstico de los pacientes con leucemia aguda mieloblástica? ¿Tiempo promedio entre el envío del paciente a la unidad de trasplante y la realización del mismo? ¿Cuál sería la mejor opción terapéutica para este paciente? Computomics: Discussions On Machine Learning Algorithms For Plant Breeding Challenges
In this episode Thomas Lübberstedt a professor of agronomy from Iowa State University shares his research goals focusing on doubled haploid technology in maize breeding. What are the challenges, what did they achieve already and what are the further research plans? How can doubled haploid speed up the process of getting homozygous lines? A second focus area is on the organic farming industry. Within 2 organic projects, Thomas is working with a sweet corn breeder and a popcorn breeder on different research goals ranging from the use of natural traits instead of GMO to inbred performance under weedy conditions. Thomas Lübberstedt is Professor and K.J. Frey Chair at the Department of Agronomy, Iowa State University (ISU). Thomas serves as Director of the R.F. Baker Center for Plant Breeding, and the Distance M.S. in Plant Breeding program. He is founder of the Doubled Haploid (DH) Facility, and Faculty Scholar of Plant Sciences Institute at ISU. Thomas served as chair of the Plant Breeding Coordinating Committee (PBCC) in 2016/17. He continued to contribute to the PBCC education objective and was co-host of the 2022 NAPB meeting at ISU.

Today on the Mushroom Hour Podcast we are joined by Dr. Michael Perlin, Professor in Biology at the University of Louisville. Dr. Perlin's interests drew him to study host/pathogen interactions at the University of Chicago and his PhD work centered on the evolution of bacterial resistance to certain classes of antibiotics/antimicrobials. As he was near to completing his degree, a friend of his stopped by the lab and introduced him to a different microbial system: a fungal pathogen that infects flowering species in the Carnation family and replaces the pollen of the flower with fungal spores. From that point he was hooked. Over the years he has continued to work on bacterial resistance, but in the last 10 years, he has shifted the focus of his lab exclusively to that on fungal plant pathogens, with projects encompassing three different types of pathogens on different hosts and the use of the yeast, Saccharomyces cerevisiae, as a model tool in the exploration of some questions more easily investigated in this well-developed system. Dr. Perlin has roughly 70 peer-reviewed scientific articles in reputable journals, including Nature; mBio; BMC Genomics; Genes, Genomes, Genetics (GGG); Fungal Genetics and Biology; Journal of Bacteriology; Eukaryotic Cell; and Molecular Microbiology. As a member of a number of scientific organizations and a prolific teacher and mentor, Dr. Perlin has a far-reaching influence on his field and has directly played a role in the careers of many PhD students. I'm excited to learn more about how fungal phytopathogens that have a massive impact on human culture. Abstract: Motivation: With the availability of new sequencing technologies, the generation of haplotype-resolved genome assemblies up to chromosome scale has become feasible. These assemblies capture the complete genetic information of both parental haplotypes, increase structural variant (SV) calling sensitivity and enable direct genotyping and phasing of SVs. Yet, existing SV callers are designed for haploid genome assemblies only, do not support genotyping or detect only a limited set of SV classes. Results: We introduce our method SVIM-asm for the detection and genotyping of six common classes of SVs from haploid and diploid genome assemblies. Compared against the only other existing SV caller for diploid assemblies, DipCall, SVIM-asm detects more SV classes and reached higher F1 scores on two recently published assemblies of the HG002 individual.
