The rabies virus kills at least 60,000 people a year, worldwide. Because it is far more prevalent in lower economic status countries, it doesn't receive adequate funding for better vaccines or treatment. Researchers like Yves Gaudin are trying to change this. He works on understanding the mechanisms of rhabdovirus transmission. Listen and learn How rhabdoviruses enter cells and install their viral factories, which are protected by liquid phase separation,  Why the polymerase is key to making the viral particle infectious, and How their efforts to destabilize the liquid phase separation that contains the viral factory is one main avenue to a possible treatment. Yves Gaudin is a researcher with CNRS and the Institut de Biologie Intégrative de la Cellule. CNRS is France's National Center for Scientific Research. He's a virologist studying the rhabdovirus family, which includes the rabies virus, Chandipura virus (CHAV), and vesicular stomatitis virus (VSV). He describes two central mechanisms that characterize this family. First the rhabdovirus causes a particular entry process and requires a polymerase for infection: the glycoprotein, which makes a spike around the virion, recognizes the cell receptor and attaches. This triggers endocytoses and an eventual fusion event allows the viral genome and polymerase is to be released into the cytoplasm. The polymerase immediately copies that genome into an mRNA, which then hijacks the cellular machinery. The polymerase presence is vital to infection—without it, the viral particle would be inert, so targeting that polymerase offers one attack possiblity.  Second, his lab has characterized the chemical nature of viral factory itself. It is distinguished by liquid phase separation—it is housed inside a compartment that separates it, like oil in water, from the cell cytoplasm. The interaction between this factory and our innate immune system spreads the infection, using the pathway to the synthesis of interferon, and so on. This liquid phase separation helps the virus keep its identity in the cell and provides another focus for fighting these viruses. We talk with Dr. Zhuoming Liu, a postdoctoral researcher, about his work using chimeric VSV to identify potential therapeutics monoclonal antibodies and drugs that prevent SARS-CoV-2 infection.

In this episode, Dr. Robert Bailer, Senior Director for Vaccines at Q2 Solutions, describes the processes for testing vaccines against the novel coronavirus, SARS-CoV-2. He covers both ELISA and virus neutralization assays. Q2 Solutions has developed an ELISA assay to evaluate multiple different antigens at the same time. Looking primarily at the spike protein, it is dissected into different parts: S1, S2, S1 plus S2, and  the receptor binding domain. The nucleocapsid protein is being evaluated as well. We are taking a 2 pronged approach to the viral neutralization assays, working in collaborations with academic institutions to deliver a cutting edge approach. The first, in collaboration with the laboratory Dr. Pei-Yong Shi at the University of Texas Medical Branch in Galveston, involves viral-like particles (VLPs) where the structural genes of wild type SARS-CoV-2 virus are removed and replaced with a reporter gene. In this particular case, it's a GFP or green fluorescent protein gene along with a neomycin resistance gene. The second approach is a SARS Co-V 2 surrogate neutralization assay used under a license agreement with the Albert Einstein College of Medicine. This is the vesicular stomatitis pseudovirion process, commonly used for viral neutralization assays. It’s a different way of expressing an artificial virus. It involves taking the vesicular stomatitis virus genome, replacing some of the genes for VSV by inserting the SARS-CoV-2 spike genes, plus a reporter gene. Expressing that produces a virus-like particle or a pseudovirion that can be utilized. This approach has been used over the years in a wide variety of different applications at Q2 Solutions for example in Ebola. The vaccine operation at Q2 Solutions has the infrastructure, the capabilities and support to have a three shift operation. We anticipate doing this 24/7, which is exactly what’s needed in the context of a pandemic. Further, our center for vaccine excellence located in Beijing, China will evaluate clinical trial samples coming to us there with the same repertoire of assays that are available in San Juan Capistrano, California. Aber auch seine Arbeit als Präsident des VSV von 1984 bis 2003 war erfolgreich. In seiner Zeit hat sich die Mitgliederzahl vom 3000 auf 13 000 erhöht.  Als gebürtiger Oberarther ist er der innerschweizer Ländlermusik am meisten verbunden, allen voran dem Klarinettler Kasi Geisser, der 1899 in Arth geboren wurde. Peter Fässler hat mit dafür gesorgt, dass der musikalische Nachlass von Kasi Geisser heute im Haus der Volksmusik in Altdorf archiviert und für alle Interessierten freigegeben ist. Von 1992 bis 2016 hat Peter Fässler in seinem Heimatdorf Oberarth den Ländlermusik-Grossanlass «z' Oberarth isch Ländler Trumpf» organisiert. Dieses Jahr hat er bei bester Gesundheit seinen 70. Geburtstag gefeiert.

Vesicular Stomatitis I had been hearing news about an equine disease vesicular stomatitis that was quarantining horses in Texas, Colorado, New Mexico, and Wyoming.  Looking for more about the disease, I contacted my vet, Dr. John Tolley of Bakersfield Large Animal Veterinary Hospital.  I recorded this interview on a Thursday, and I write this the following Tuesday.  The number of quarantined premises have doubled. If you look up the words "vesicular" and "stomatitis" on Google, you'll find vesicular means blister.  Stomatitis is an inflammation of the mouth.  Vesicular stomatitis usually presents in horses with blisters in the mouth, lips, and coronary bands.  The disease spreads several ways, but most commonly through flies and mosquitos.  Some of those flies can move 150 miles in a day.  I wasn't able to find a good answer about a vaccine for VSV.  It is a summer disease and spreads up from the gulf in Texas. Dr. Tolley gives us the details on vesicular stomatitis.  He talks about suspected origins and why the Federal government keeps a close eye on the number and location of animals affected.  Good horse-keeping practices and staying away from horses afflicted with the disease are currently the main preventative measures. The elimination of virus-infected cells to block viral spread is substantially conveyed by effector CD8+ T cells. Trafficking of naïve T cells into the lymph node is a crucial step for their activation to effector T cells. The entry into the lymph nodes is mediated by several cell adhesion molecules that are highly specific for given lymph nodes. For the ingress into the gut-associated lymphoid tissue (GALT) such as mesenteric lymph nodes and Peyer’s patches, the integrin α4β7 is the major homing receptor that interacts with its main ligand MAdCAM-1. On naïve T cells, α4β7 is expressed at low level, however upon activation in the GALT its expression is significantly upregulated on effector T cells. Consequently these effector T cells migrate into the gastrointestinal tract, where MAdCAM-1 is also expressed. The migration patterns of virus-specific CD8+ T cells upon viral infection have been studied in detail. In contrast, the migration of the major population of virus-unspecific CD8+ T cells, i.e. bystander-activated T cells is poorly understood. In our study, we aimed to determine how trafficking of these non-cognate, bystander-activated T cells is affected upon viral infection. We discovered that α4β7 expression was negatively regulated on bystander-activated CD8+ T cells upon injection of poly (I:C), which imitates innate immune activation upon viral infection. This effect was also observed for viral infections such as Sendai, EMCV and the mutant form of VSV virus. Furthermore, we scrutinized the direct role of IFN-α on T cells to exert α4β7 modulation. In the case of EMCV infection, IL-6 played a dominant role in the alteration of α4β7 expression. Finally, using an adoptive transfer model we could prove that the downregulation severely impacted the trafficking of T cells into the Peyer’s patches and to a lesser extent into the mesenteric lymph nodes. These findings demonstrate that a mechanism to regulate trafficking of bystander-activated T cells during viral infection exists and that this is controlled by the induction of cytokines such as IFN-α and IL-6. We hypothesize that the downregulation of α4β7 on naïve T cells functions 1) to allow space for virus-specific effector T cells to expand in the GALT and 2) to exclude bystander-activated T cells from the GALT in order to prevent mistrafficking, which could cause autoimmune diseases.

The intrinsic oncolytic specificity of vesicular stomatitis virus (VSV) is currently being exploited to develop alternative therapeutic strategies for hepatocellular carcinoma (HCC). Identifying key regulators in diverse transduction pathways that define VSV oncolysis in cancer cells represents a fundamental prerequisite to engineering more effective oncolytic viral vectors and adjusting combination therapies. After having identified defects in the signalling cascade of type I interferon induction, responsible for attenuated antiviral responses in human HCC cell lines, we have now investigated the role of cell proliferation and translation initiation. Cell cycle progression and translation initiation factors eIF4E and eIF2Bepsilon have been recently identified as key regulators of VSV permissiveness in T-lymphocytes and immortalized mouse embryonic fibroblasts, respectively. Here, we show that in HCC, decrease of cell proliferation by cell cycle inhibitors or siRNA-mediated reduction of G(1) cyclin-dependent kinase activities (CDK4) or cyclin D1 protein expression, do not significantly alter viral growth. Additionally, we demonstrate that translation initiation factors eIF4E and eIF2Bepsilon are negligible in sustaining VSV replication in HCC. Taken together, these results indicate that cellular proliferation and the initiation phase of cellular protein synthesis are not essential for successful VSV oncolysis of HCC. Moreover, our observations indicate the importance of cell-type specificity for VSV oncolysis, an important aspect to be considered in virotherapy applications in the future.

Recombinant parapoxvirus vaccine against classical swine fever: comparative characterization of immune reactions in swine Despite good efforts in eradication of classical swine fever (CSFV) within the EU massive disease outbreaks cannot be excluded. Therefore an “intervention vaccination strategy” is one option in legal EU combat strategies against CSF. At the moment an E2 subunit vaccine is licensed in the EU but possibly can be replaced by an improved vaccine to achieve optimal stimulation of the humoral as well as the cellular immune responses. The aim of the present work was the generation of a new Parapoxvirus ovis vector expressing the E2 glycoprotein of CSFV which was named ORFV D1701VrVE2. It could be successfully demonstrated that his vector vaccine efficiently expresses the foreign gene even in cells of swine as a non permissive host. It was shown to be completely avirulent for pigs. The vector is a potent stimulator of IFN-α and VSV antiviral activity in porcine PBMC supernatants. In several animal immunisation experiments with swine ORFV D1701VrVE2 induced CSFV-neutralizing serum antibodies already after a single application. Immunized piglets were protected against lethal CSFV challenge and did not develop fever. Vaccinated and challenged animals recovered from the characteristic CSFV-induced B-cell reduction. The distribution of the vaccine dose over four intra-muscular injection sites (multi-site application) led to a rapid induction of neutralizing antibodies and to solid protection from CSF after a single vaccination. In contrast to single site vaccinated animals, multi-site vaccinated piglets did not transmit the challenge virus to a naive sentinel. Two successive vector virus applications in a homologous prime-boost regimen did not provoke IFN-γ producing cells among PBMC. However, a heterologous prime-boost regimen as a combination of prime with baculovirus expressed glycoprotein E2 followed by boost with the parapoxvirus vector induced high numbers of IFN-γ producing cells. A similar beneficial effect became evident when the challenge infection mimicked the booster vaccination after a single vector prime. In contrast when the challenge CSFV was applied after a homologous prime with modified live CSFV vaccine the immunised piglets responded with lower numbers of IFN-γ producing cells as well as lower titres of CSFV-neutralizing serum antibodies. In additional experiments the adjuvant effect of lipoprotein L-OprI of Pseudomonas aeruginosa was tested in combined administration with baculovirus expressed E2 in piglets using sub-optimal doses of the E2 protein. The adjuvant showed a beneficial effect on the formation of CSFV neutralizing serum antibodies but had no influence on the number of IFN-γ producing cells in piglets before challenge. As another important CSFV subunit the immunogenic properties of the E. coli produced non structural protein NS3 have been tested. Although NS3 induced high amounts of IFN-γ producing cells in PBMC of vaccinated piglets their serum antibodies did not neutralize CSFV and even after two booster vaccinations the animals were not protected against lethal CSFV challenge.

Eine neue Strategie wäre eine Kombination von Chondrozytentransplantation mit gentherapeutischen Methoden, z. B. die stabile Ex-vivo-Transduktion von primären Chondrozyten mit retroviralen Vektoren, die gewebeaufbauende Zytokine exprimieren. Ziel der vorliegenden Arbeit war deshalb Etablierung und Optimierung des retoviralen Gentransfers in primäre Kaninchenchondrozyten und die Beobachtung von Verbleib, Vitalität und Genexpression der Zelltransplantate in vivo. Transduzierte Zellen sollten ohne weitere Selektionsverfahren für eine spätere Transplantation verwendet werden können. Da regulierbare Genexpression in einem solchen Modell von Vorteil ist, war ein weiteres Ziel die Entwicklung retroviraler Tet-On-Vektoren. Es wurden konstitutiv exprimierende retrovirale Vektoren und Tet-On-Vektoren kloniert und Retroviren mit unterschiedlichen Infektionsspektren generiert. Effizienz und Stabilität des Gentransfers wurden ohne weitere Selektionsverfahren mit Hilfe konstitutiv nlslacZ-exprimierender retroviraler Vektoren in vitro und in vivo beurteilt. Zusätzlich wurde stellvertretend für ein therapeutisches Gen der Wachstumsfaktor hbmp-2 transferiert. Retrovirale Ein-Vektor-Systeme, die den reversen Transaktivator rtTA2s-M2 und den Tet-responsive Promotor enthielten, wurden in vitro durch die Expression der Reportergene nlslacZ oder egfp in verschiedenen Zelllinien getestet. Mit VSV.G-pseudotypisierten Retroviren konnte eine Transduktionseffizienz von bis zu 99 % erzielt werden, amphotrope Viren waren deutlich weniger effizient. Transduzierte Chondrozyten zeigten in vitro über mindestens 12 Wochen eine stabile Transgenexpression, die auch in 3D-Kultur auf Kollagenschwämmen fortbestand. In vivo war für mindestens drei Wochen Transgenexpression nachweisbar. hbmp-2 transduzierte Zellen exprimierten zudem dieses Transgen in vitro. Die neu entwickelten Tetrazyklin-induzierbaren Retroviren zeigten eine starke Basalexpression bei nur geringer Steigerung nach Induktion mit Doxyzyklin. Die Transduktionseffizienz dieser Retroviren war wesentlich geringer als bei der Verwendung konstitutiv exprimierender Vektoren. VSV.G-pseudotypisierte Retroviren sind eine optimale Methode für den retroviralen Gentransfer in primäre Kaninchenchondrozyten ohne weitere Selektionsverfahren. Neben dem Transfer von Markergenen ist dies die Grundlage für den Transfer von therapeutischen Genen, um deren Effekt in vitro und in vivo zu untersuchen. Zudem wurde ein neuartiger universal einsetzbarer Vektor entwickelt, der die Klonierung in die retroviralen U3 Region und somit die Konstruktion retroviraler Double-copy-Vektoren erlaubt. Die Entwicklung zuverlässiger retroviraler Tet-On-Vektoren bleibt weiterhin eine Herausforderung für die Zukunft.
