N., Monroe, D., Khosla, S. Abstract: Senescence drives organismal aging, yet the deep characterization of senescent cells in vivo remains incomplete. Here, we applied mass cytometry by time-of-flight (CyTOF) using carefully validated antibodies to analyze senescent cells at single-cell resolution. We used multiple criteria to identify senescent mesenchymal cells that were growth arrested and resistant to apoptosis (p16+/Ki67-/BCL-2+; p16KB cells). These cells were highly enriched for senescence-associated secretory phenotype (SASP) and DNA damage markers and were strongly associated with age. p16KB cell percentages were also increased in CD24+ osteolineage cells, which exhibited an inflammatory SASP in aged mice and were robustly cleared by both genetic and pharmacologic senolytic therapies. Following isolation, CD24+ skeletal cells exhibited growth arrest, SA-beta gal positivity, and impaired osteogenesis in vitro. These studies thus provide a new approach using multiplexed protein profiling by CyTOF to define senescent mesenchymal cells in vivo and identify a highly inflammatory, senescent CD24+ osteolineage population cleared by senolytics. Is the Mississippi River drying up? That's unlikely, but for 2022 this stalwart part of a vibrant transportation infrastructure has been challenged by the continuing Midwest drought. Recently, barge traffic was halted on the river, and when it resumed there were limits imposed on how barges. Ginger Rowsey, https://www.deltafarmpress.com (Delta Farm Press), has been following this ongoing river issue and she discusses what she's learned so far. And she shares what she learned talking with Tug Captain, Eric Badeaux, a 40-plus year river veteran. He discussed what's been happening to him on the river. Rowsey also points out that while grain flows are limited which is weakening basis at river terminals, there is another challenge – moving fertilizer for the 2023 crop.

J. James Rowsey, M.D. is a board-certified ophthalmologist, specializing in refractive surgery and corneal diseases. Dr. Rowsey was a primary investigator in the use of Laser for treating myopia in 1988. He expanded his expertise to become the VISX principal certifying surgeons for other physicians in Florida. Dr. Rowsey has trained numerous surgeons to perform refractive surgery in Florida, the United States, and throughout the world. Dr. Rowsey came to Florida in 1991 to assume the position of the James P. and Heather Gills Professor of Ophthalmology and to chair the Ophthalmology Department at the University of South Florida. In this position, he built the 28,000 square foot USF Eye Institute. His current practice includes LASIK refractive surgery, corneal transplantation, cataract surgery, and infectious and immunological diseases of the eye. Dr. Rowsey completed his undergraduate degree and medical school at The George Washington University, Washington, D.C. His internship and residency were at the Henry Ford Hospital in Detroit, Michigan. In 1976 he initiated the corneal research program at the Dean A. McGee Eye Institute, University of Oklahoma, where he continued until his 1991 relocation to Florida. Dr. Rowsey is married to Judith Carole and has four daughters Kimberly Vargas (Husband Michael); Dr. Shannon Mann (Husband Jesse and son Joshua); Jheri Dee Scott (Husband David, daughters Jordan and Mollie, sons Luke, Evan, Ethan) and Jennifer Whittington (Husband Kevin and sons John James, Landon, and daughter Lauren). He is blessed by his sons-in-love and his nine grandchildren, who bring new joys daily to the family.
