Abstract: Comparative "omics" studies have revealed unique aspects of human neurobiology, yet an evolutionary perspective of the brain N-glycome is lacking. Here, we performed multi-regional characterization of rat, macaque, chimpanzee, and human brain N-glycomes using chromatography and mass spectrometry, then integrated these data with complementary glycotranscriptomic data. We found that in primates the brain N-glycome has evolved more rapidly than the underlying transcriptomic framework, providing a mechanism for generating additional diversity. We show that brain N-glycome evolution in hominids has been characterized by an increase in complexity and (2-6)-linked N-acetylneuraminic acid along with human-specific cell type expression of key glycogenes. Finally, by comparing the prenatal and adult human brain N-glycome, we identify region-specific neurodevelopmental pathways that lead to distinct spatial N-glycosylation profiles in the mature brain. Rob shares many mindset changes he went through after starting his business, Fusion Painting. Like many of us, he learned painting from his father, but unlike many of us, his dad had him working from the age of 12 so that he would be persuaded to never paint, but to work hard in school and avoid manual labor and get an office job. Rob’s father taught him how to work hard and he excelled. Rob became a mechanical engineer only to hate it and want nothing more than to be out painting. Rob then worked for his dad until his dad recommended he start his own company, which Rob willingly and excitedly did. Feeling like he couldn’t fail, he took on what he thought was the best job and best market for him on millionaires lane—only to find it was nightmare lane. After losing $27,000 on that job, receiving no referrals, and not even being remembered by the home owner, he went to work for many GC’s. Having an awful time with them, his amazing paint rep help him and told him he should break into the residential market (avoiding the Big House on the Hill clients from then on!). This was one of many mindset changes or mind-shifts Rob went through and he shares them with us today. That was really the shift, the transition from a craftsman painter into a businessman." 12:05 "I think it was Richard Branson who said, "Look after your staff and they'll look after your customers." And really, really the team is everything to our business and it should be to everyone else's business. They are really the engine room of the business. You know, I go out and sell a job and do all that and generate leads and stuff, but really, at the end of the day, if you can't produce a job, you're not gonna make any money. So these guys are out there, they're at the coalface, really producing the job. They are the ones who are interacting with our customers day to day, they are the face of your business. They're the guys that have got the branding on the tee shirts, branding on the vans. They are really everything in a business, you know, without them, we don't have a business really. ... Really that is so important to me that's where the company culture has to come into it. So it's strong, it's a very strong thing. And I've always, always, always valued that." 12:58 "(Following Up) This is something you read about in books, back when I started, many years ago: follow up, follow up, follow up. It was something that I was resistant to do. I could have given you 20 reasons why. I didn't want to do it. It was only when I had people come over and bid to do things around my own home. And the guys that followed up, it really, really left an impression on me. And that was it. That was my mind shift. The way I felt when I got followed up, because I almost (used to feel) it was a bit cheesy or a bit almost like a used car salesman thing, but really, it's the way you do it. And these people will follow me up because they were genuinely interested in the job. It really left a great impression on me. And I tell you from that moment, I'm following up." Het heeft even geduurd voordat we Atlas MMA featherweight kampioen gesproken hadden maar we hebben het eindelijk geregeld. We spreken met Dennis Vonk (18:57) over Atlas MMA en zijn laatste partij daar waar hij de featherweight titel wist binnen te slepen. OOk spreken we over de toekomst en wat hij wel en wat hij niet wil bereiken in de sport en dat is net niet het antwoord wat je verwacht. Hij heeft een duidelijk pad voor ogen en stapt binnenkort in het huwelijksbootje. Dat vergt natuurlijk een hoop planning en is hij bezig met het grote plaatje en niet alleen vechten. Experts estimate that computers gobble up as much as 10% of global electricity. They predict that that share will only grow as data centers expand and the internet of things brings scads of new computer-controlled devices to the world. Jelena Vuckovic is an electrical engineer who sees a light on the horizon — quite literally. She is building computers that calculate and communicate more with photons than electrons. These “photonic” devices could cut energy consumption in half and empower exciting new technologies, like quantum computing, in the process. Before that day can come, however, Vuckovic and other proponents of photonics — the science of using light for practical purposes — will have to design smaller devices and improve manufacturing techniques to compete with today’s ultra-small electronics. To hasten that arrival, Vuckovic is turning to artificial intelligence to develop new device designs and new materials that could usher in the age of photonics. In this episode, Casey and Megan spoke to Dr. Dian Ginsberg and Sheila Vuckovic about Hormones and PCOS. We also discussed their new book, The PCOS Environmental Roadmap, now available! Dr. Ginsberg completed her residency in Obstetrics and Gynecology at the Bowman-Gray School of Medicine in North Carolina before Houston's sunny climate brought her to Texas. Dr. Ginsberg has years of experience providing comprehensive obstetrics and gynecology treatment and as her career advanced she developed a special interest in functional medicine and the manner in which it bridges the gap of a patients illness while minimizing severe symptoms and optimizing health. This passion now encompasses special focus on the understanding of genetics and methylation, optimal hormone balance and overall healthy aging. Being a black belt in Tae Kwan Do and an accomplished marathon runner and triathlete has led her to explore the latest medical advances in wellness for athletes along with anti-aging therapies for both men and women. She is motivated to continue her research in nutritional therapies for many reasons, most significantly for the benefits it has brought to her sons with specific learning needs. Dr. Ginsberg has used her extensive experience to help men and women with hormone imbalance regain the vitality and fitness they thought was lost forever. She lectures for Functional Medicine University on the topic of PCOS, its natural reversal, and pathways to fertility. She is the author of The PCOS Environmental Roadmap. Rectangularization of aging is her personal passion, and she is faculty at the American Academy of Anti-Aging where she has lectured about hormones and telomeres (DNA caps) and how instrumental their preservation is for ongoing health. Sheila received her undergraduate degree in Exercise Science from Texas A&M University. Passionate about health and wellness she began working as a personal trainer at the corporate wellness center at BP. Her interest in nutrition grew and combined with her Exercise Science background, she received her Masters in Health and Nutrition Education from the University of Houston. After much research and completely overhauling the whole family's diet and vitamin supplementation, she saw such a dramatic change in her son that it fueled her passion for helping others with similar inflammatory conditions. Sheila is married to her college sweetheart, Gordan, and they have 3 children. She is an avid cross-fitter and takes various dance classes in her spare time. Stefan Vuckovic is the first man ever to win an Olympic silver medal in Triathlon. The sport made its debut at the Sydney Games in 2000 and in one of the best moments of that wonderful event, Stefan grabbed a German flag just before the triathlon finish line and danced the last 50 metres of the race, waving the flag and celebrating. On today's show we reflect on that incredible moment, the journey that led Stefan to it, and what happened next.

I would like to say Leslie Hanti's photographic work is serendipitous, but that would imply his work comes by chance or luck. While there is alway an element of luck and chance, it is also Leslie's vision, determination and persistence which allows him to capture his images. As you will hear, Leslie can spend hours, days, weeks and years waiting for all of the elements in an image to come together. Leslie's card reads "Stop-by-Stop Images", because he is almost always with camera in-hand and ready to record the world as he sees it. Like an onion, you have to peel back the many layers to get to know Leslie Hanti. Over the last few years and I think I've only seen a small fraction of his work. Hopefully soon we will all have an opportunity to see more of his work.
