In deze podcast is Team Lokaal raadslid Marco van den Berg te gast. Hij gaat in op het indienen van een motie van wantrouwen, die Forum voor Democratie op de raadsvergadering in mei gaat indienen tegen wethouder Bouke van Vught. Er was ophef rond de verkoop van een zonnepark aan de A73. Volgens Marco van den Berg: "en dat is mijn eigen persoonlijke mening, is een rode kaart natuurlijk niet aan de orde. Een gele kaart is hier meer op zijn plaats, dus een motie van treurnis." Maar ik wacht af wat de wethouder te zeggen heeft. Verder gaat hij in het gesprek in op zijn aanstaande vertrek inzake een verhuizing naar Nijmegen en blikt hij terug op zijn politieke carrière. Hij was ook van mening dat de middelbare school in Mill had moeten blijven, "maar dat hadden ze een aantal jaren terug al moeten regelen, door meer onderwijs aan te bieden. Dat is nu te laat. Helaas."

Als das Auto endlich anhält, versucht der Fahrer zu Fuß zu flüchten - das gelingt ihm aber nicht. Als die Polizist:innen den Kofferraum öffnen, machen sie eine schreckliche Entdeckung. Mit Alexander Brock spricht Franziska Wagenknecht über das schreckliches Verbrechen, dass die Anwohner:innen der Reindelstraße im Winter 2018 tief erschütterte.

Burgemeester Bruls heeft woensdagavond 39 nieuwe raadsleden geïnstalleerd. Door het afleggen van de eed of de verklaring hebben deze raadsleden de start van hun raadsperiode ingeluid. De raad is iets jonger dan vier jaar geleden, wist Bruls. Het gemiddelde raadslid is nu 40,2 jaar oud. Bruls gaf de nieuwe raadsleden direct een serieuze boodschap mee. Er ligt een zware taak voor hen klaar, onder meer vanwege de bezuinigingen die vanuit het Rijk op de gemeentes afkomen. Een dag eerder werd afscheid genomen van de oude gemeenteraadsleden, van wie er 21 niet meer terugkeren. Daarover praten Rob Jaspers en Raymond Janssen deze week in deze aflevering van Ien, de podcast. En uiteraard kwam ook de formatie te sprake. Het ligt voor de hand dat de drie grootste partijen als eerste met elkaar praten, maar volgens Jaspers is dat niet zonder meer zo. “D66 zal echt een andere rol spelen, nu ze zo veel kleiner zijn geworden dan ze aanvankelijk hoopten”, zeg hij. Een coalitie zonder D66 behoort dan ook tot de mogelijkheden, zegt Janssen. “Bijvoorbeeld met de PvdA die een kleine winst boekte”, aldus Janssen. Ongedeelde stad Onlangs kwamen op verzoek van woningcorporatie Talis verschillende Nederlandse en Britse woningcorporaties naar Nijmegen om te kijken hoe de stad met woningbouw en het woningprobleem omgaat. Centraal bij dit bezoek stonden de ‘ongedeelde stad', waarin gewerkt wordt aan het verkleinen van de scheiding hebben tussen rijk en arm en de ‘groeistad', die Nijmegen al weer enkele jaren is. ProXp-ala prevents proteome-wide Pro-to-Ala mutations by hydrolyzing misacylated Ala-tRNAPro, which is synthesized by prolyl-tRNA synthetase (ProRS). Bacterial ProXp-ala was previously shown to combine a size-based exclusion mechanism with conformational and chemical selection for the recognition of the alanyl moiety, while tRNAPro is selected via recognition of tRNA acceptor stem elements G72 and A73. The identity of these critical bases changed during evolution with eukaryotic cytosolic tRNAPro possessing a cytosine at the corresponding positions. The mechanism by which eukaryotic ProXp-ala adapted to these changes remains unknown. In this work, recognition of the aminoacyl moiety and tRNA acceptor stem by human (Hs) ProXp-ala was examined. Enzymatic assays revealed that Hs ProXp-ala requires C72 and C73 in the context of Hs cytosolic tRNAPro for efficient deacylation of mischarged Ala-tRNAPro. The strong dependence on these bases prevents cross-species deacylation of bacterial Ala-tRNAPro or of Hs mitochondrial Ala-tRNAPro by the human enzyme. Similar to the bacterial enzyme, Hs ProXp-ala showed strong tRNA acceptor-stem recognition but differed in its amino acid specificity profile relative to bacterial ProXp-ala. Changes at conserved residues in both the Hs and bacterial ProXp-ala substrate binding pockets modulated this specificity. These results illustrate how the mechanism of substrate selection diverged during the evolution of the ProXp-ala family and provides the first example of a trans-editing domain whose specificity evolved to adapt to changes in its tRNA substrate.
