Clinical Geneticist & Professor Dr Nicholas (Nick) Pachter chats with Stephanie about Lynch syndrome, prevalence in the community, ongoing surveillance as well as treatment options. Lynch syndrome is caused by a change in a gene that normally functions to protect a person from getting cancer. A mutation carriers lifetime risk for bowel or other syndrome cancers of around 70-90% and each of their children has a 50% chance of inheriting the mutation.'If you have Lynch syndrome don't be discharged (from your medical team) . . . as you need ongoing follow-up well into your 80's as you are still at risk of getting another bowel cancer in your lifetime', Dr Pachter explains.

Video game giants Call of Duty and World of Warcraft have a new home: Microsoft. On Tuesday, Microsoft announced it will acquire Activision Blizzard, the publisher behind Call of Duty, one of the top-selling video games in the U.S., along with several other titles including Overwatch, World of Warcraft and Candy Crush. Microsoft CEO Satya Nadella said the deal will help the company's expansion into the metaverse. For consumers, the deal could mean one of gaming's most successful franchises will only be available on a PC or Xbox. "Gaming is the most dynamic and exciting category in entertainment across all platforms today and will play a key role in the development of metaverse platforms,” Nadella said in a statement. "We're investing deeply in world-class content, community and the cloud to usher in a new era of gaming that puts players and creators first and makes gaming safe, inclusive and accessible to all." Microsoft will acquire Activision Blizzard in an all-cash deal valued at $68.7 billion. The announcement follows reports in November that Microsoft was evaluating its relationship with the video game publisher amid allegations Activision Blizzard CEO Bobby Kotick had known about sexual misconduct claims at the company for years. The state of California sued Activision Blizzard last year, claiming the publisher fostered a sexist culture and paid women less than men, despite doing more work. Employees at Activision Blizzard staged walkouts urging the company to remove Kotick from the top executive position and make changes to the company's culture. In its statement, Microsoft said Kotick will stay on as CEO of the publisher, reporting to Phil Spencer, head of Microsoft Gaming. Wedbush analyst Michael Pachter said the deal hurts Sony, which makes the competing PlayStation 5 video game console. "You're very likely going to lose Call of Duty on PlayStation one of these days," said Pachter. According to NPD Group, Call of Duty has been the best-selling video game franchise in tracked dollar sales for 13 straight years. Microsoft has joined other companies including Meta and Fortnite creators Epic Games in carving out their space in the metaverse, a virtual universe that supporters believe will take place of the modern internet. Microsoft has dabbled in elements of this universe through its popular world-building game Minecraft, as well as in augmented reality and virtual reality tools. GlobalData principal analyst Rupantar Guha said this deal will help Microsoft jump forward as a leader in the metaverse. Charlie Pachter is a Canadian artist. Today, along with his famous depictions of the maple leaf flag, they are regarded affectionately as Canadian pop art icons and the artist is broadly recognized as a northern Warhol. One of Canada's leading contemporary artists, Mr. Pachter is a painter, printmaker, sculptor, designer, lecturer and all-around “witty intellectual, historian, entrepreneur and lovable bon vivant,” according to the Globe and Mail. His paintings hang in the Toronto Stock Exchange and in the Canadian Embassy in Washington. His muralHockey Knights in Canada highlights a Toronto subway station. Retrospective Pachter exhibitions have toured France and Germany, and an illustrated biography on Pachter and his work has been published by McClelland & Stewart.

The story so far: some people in Cordoba did a randomized controlled trial of Vitamin D for coronavirus. The people who got the Vitamin D seemed to do much better than those who didn’t. But there was some controversy over the randomization, which looked like this Remember, we want to randomly create two groups of similar people, then give Vitamin D to one group and see what happens. If the groups are different to start with, then we won't be able to tell if the Vitamin D did anything or if it was just the pre-existing difference. In this case, they checked for fifteen important ways that the groups could be different, and found they were only significantly different on one - blood pressure. Jungreis and Kellis, two scientists who support this study, say that shouldn't bother us too much. They point out that because of multiple testing (we checked fifteen hypotheses), we need a higher significance threshold before we care about significance in any of them, and once we apply this correction, the blood pressure result stops being significant. Pachter challenges their math - but even aside from that, come on! We found that there was actually a big difference between these groups! You can play around with statistics and show that ignoring this difference meets certain formal criteria for statistical good practice. But the difference is still there and it's real. For all we know it could be driving the Vitamin D results.

Abstract: Intrinsic and extrinsic noise sources in gene expression, originating respectively from transcriptional stochasticity and from differences between cells, complicate the determination of transcriptional models. In particularly degenerate cases, the two noise sources are altogether impossible to distinguish. However, the incorporation of downstream processing, such as the mRNA splicing and export implicated in gene expression buffering, recovers the ability to identify the relevant source of noise. We report analytical copy-number distributions, discuss the noise sources' qualitative effects on lower moments, and provide simulation routines for both models. Veel coureurs doen nu mee aan digitale races in simracinggames. Maar wat maakt die games nou zo anders? En hoe begin je er aan? De ins en outs van games als iRacing en RFactor 2 worden besproken in deze podcast en ook werpen we een blik op wat je zoal nodig hebt, wil je echt een simracingwaardige setup neerzetten bij jou thuis. Daaruit blijkt dat Jelle en Wout redelijk voorzien zijn, maar er is zeker ruimte voor verbetering. Verbetering is ook wat Wout op een andere manier zoekt, namelijk in zijn fitheid. Mede geïnspireerd door de verhalen over Ring Fit Adventure besloot hij zich aan een vr-fitnessgame te wagen, die hem als een bokser laat slaan, ontwijken en squatten. Arnoud bespreekt de nieuwe Magic Keyboards van Apple, waarover hij van mening verschilt met Jelle. Wel lijken beide heren het er over eens dat de invloed van Jony Ive wellicht steeds minder goed zichtbaar is de ontwerpen van Appleproducten. Jelle heeft ondertussen flink zitten gamen. Eigenlijk wil hij meer vertellen over de nieuwe uitbreidingspakketten voor Civilization IV - en dat doet hij stiekem ook - maar toch orakelt hij liever over Mindustry, een tower defense game waarin het draait om het vergaren en managen van resources. Leuk detail: Jelle kocht het spel voor vijf euro op Steam, maar het is ook gewoon gratis te downloaden via de site van de ontwikkelaar. We blijven nog even hangen bij gaming, want Jurian haalt wat uitspraken aan die analyst Michael Pachter en voormalig Microsoft-topman Peter Moore onlangs deden in een podcast van gamepromoter Geoff Keighley. Volgens Pachter zo Microsoft wachten met het aankondigen van de prijs van de Xbox Series X tot Sony met een prijs voor de PlayStation 5 zou komen. Pachter verwacht dat de Series X honderd dollar goedkoper zal zijn dan de PlayStation 5 en schat de verkoopprijzen op respectievelijk 400 en 500 dollar. Today's guest painted Hockey Knights in Canada, the famous murals that are opposite each other at the College Street Subway station.  His name is Charles Pachter, and his work not only adorns subway halls in downtown Toronto, but also the Canadian Embassy in Washington, Canada House in London, the Parliament Buildings, the Prime Minister’s residence, and the Embassy of France in Ottawa.  There’s also a short video that goes with this episode that I’ve posted to Youtube. We are just one weekend removed from the incredible finale to the League of Legends World Championship. No spoilers here just in case some of our listeners haven't yet seen it, but expect more on Worlds next week. After a layoff spree in February of this year, things finally seem to be looking up at Sony Santa Monica, which has been hiring many new professionals. What could the studio behind God of War be up to? There are also rumors about some of the things in store for Destiny. "The Dark Below," its first expansion pack, is expected for release in December, and there are some rather specific reports about new sub-classes and raids! This past week has also been controversial because of one Michael Pachter, the video game industry analyst infamous for his predictions. He surprised gamers everywhere by announcing his prediction that Xbox One would outsell PlayStation 4 for September. Spoiler alert: he was completely wrong. We know this because September's NPD results are in, and we're happy to walk you through them! There are some interesting surprises in them, especially the sales of Destiny for Xbox One versus those of the PS4. The crew discusses why gamers should take that particular bit of information with reservations. This week, the intro music is "Warriors," a fantastic song by Imagine Dragons that has been League of Legends' theme for the World Championships.
