Die Sauerstoffarmut stellt für einen Teil der Tumorzellen, die aus dem Wachstum solider Tumoren entstehen, im besonderen Maße für die Strahlentherapie und auch für die Chemotherapie ein Hindernis dar. Um dieses Hindernis, das zum Teil für die lokale Rezidivierung und die maligne Progression von Tumoren verantwortlich ist, zu einem neuen Ansatzpunkt für die Therapie zu machen, hat man Substanzen wie das Tirapazamine (TPZ) entwickelt, die unter Sauerstoffarmut ihre tumortoxische Wirkung entfalten. In dieser Untersuchung sollte in einem Tumor-Maus-Modell erforscht werden, wie sich die Kombination von Tirapazamine mit ionisierender Strahlung und cis-Platin auf das Tumorwachstum auswirkt. Als Vergleich diente die Standardtherapie mit ionisierender Strahlung und cis-Platin. An einem Tumor-Maus-Modell (C3H-Maus, murines Fibrosarkom Rif-1) wurden auf dem rechten Fußrücken durch subkutan-Injektion von Tumorzellsuspensionen Tumoren erzeugt. Nach einer initialen Wachstumsphase wurden die Tumoren mit ionisierender Strahlung, cis-Platin und TPZ behandelt. Die Wirkung der Therapeutika sollte in den oben genannten Kombinationen mit der Wachstumsverzögerung eines unbehandelten Tumors verglichen werden. Die Kombination von ionisierender Strahlung, TPZ und cis-Platin kann mit steigender TPZ-Dosis eine jeweils stärkere Wachstumsverzögerung des Tumors erreichen. Unter der Kombination von ionisierender Strahlung, TPZ und cis-Platin tritt zum Teil eine hohe Letalität auf. Diese Letalität ist unter der Behandlung mit ionisierender Strahlung und cis-Platin nicht zu beobachten. Vor dem Hintergrund dieser Ergebnisse kann die Anwendung von TPZ in der Tumortherapie derzeit nicht empfohlen werden. Weitere Untersuchungen sollten sich mit den Veränderungen, die TPZ im Organismus verursacht, beschäftigen.
