S., Bethge, M., Berens, P., Franke, K., Ecker, A. S., Euler, T. Abstract: The retina transforms patterns of light into visual feature representations supporting behaviour. These representations are distributed across various types of retinal ganglion cells (RGCs), whose spatial and temporal tuning properties have been extensively studied in many model organisms, including the mouse. However, it has been difficult to link the potentially nonlinear retinal transformations of natural visual inputs to specific ethological purposes. Here, we discover a novel selectivity to chromatic contrast in an RGC type that allows the detection of transitions of the horizon across a retinal region. We trained a convolutional neural network (CNN) model on large-scale functional recordings of RGC responses to natural mouse movies, and then used this model to search in silico for stimuli that maximally excite distinct types of RGCs. This procedure predicted centre colour-opponency in transient Suppressed-by-Contrast RGCs (tSbC), a cell type whose function is being debated. We confirmed experimentally that these cells indeed responded very selectively to Green-OFF, UV-ON contrasts, which we found to be characteristic of transitions from ground to sky in the visual scene, as might be elicited by head- or eye-movements across the horizon. Because tSbCs reliably detected these transitions, we suggest a role for this RGC type in providing contextual information (i.e. sky or ground) necessary for the selection of appropriate behavioural responses to other stimuli, such as looming objects. Our work showcases how a combination of experiments with natural stimuli and computational modelling allows discovering novel types of stimulus selectivity and identifying their potential ethological relevance. Abstract: Neuroscience is pushing toward studying the brain during naturalistic behaviors with open-ended tasks. Grid cells are a classic example, where free behavior was key to observing their characteristic spatial representations in two-dimensional environments. In contrast, it has been difficult to identify grid cells and study their computations in more restrictive experiments, such as head-fixed wheel running. Here, we challenge this view by showing that shifting the focus from single neurons to the population level changes the minimal experimental complexity required to study grid cell representations. Specifically, we combine the manifold approximation in UMAP with persistent homology to study the topology of the population activity. With these methods, we show that the population activity of grid cells covers a similar two-dimensional toroidal state space during wheel running as in open field foraging, with and without a virtual reality setup. Trajectories on the torus correspond to single trial runs in virtual reality and changes in experimental conditions are reflected in the internal representation, while the toroidal representation undergoes occasional shifts in its alignment to the environment. These findings show that our method can uncover latent topologies that go beyond the complexity of the task, allowing us to investigate internal dynamics in simple experimental settings in which the analysis of grid cells has so far remained elusive. Der russische Krieg in der Ukraine sitzt in Gestalt einer Flüchtlingsfamilie auf dem Sofa. Und zugleich hat die russische Aggression eine ganze Reihe vermeintlich sicherer Grundlagen unserer gedanklichen Weltordnung abgeräumt, beginnend mit der Kraft des Völkerrechts und der Geltung längst geschlossener Verträge. Zur Jubiläumsfolge ein Gespräch mit einem der Lieblingsgäste aus den ersten hundert Folgen: Thomas Klindt, Jurist und Rechtsanwalt. Michael und Tom diskutieren, ob das Völkerrecht nur ein Recht für sonnige Tage ist. Haben wir uns einlullen lassen von der Vorstellung, einen stabilen Rahmen etabliert zu haben, obwohl die russische Regierung schon lange und sehr sichtbar krass dagegen verstößt? Georgien, Tschetschenien, die Krim waren alle schon da. Was aber sollen wir denn sonst tun, wenn nicht Verträge unterschreiben? Gilt das Völkerrecht als Grundlage friedlicher Zusammenarbeit global? Oder haben wir es vielmehr mit partikularen Interessen zu tun? Haben wir uns überhoben mit dem Anspruch darauf, dass diese Art zu Leben und zu Regieren die Allseligmachende ist? Auch wenn wir das für uns glauben, bleibt es doch unsere Perspektive. Sind die Menschenrechte, so unverhandelbar sie für uns sind, möglicherweise gar nicht weltweit in die kulturelle Genetik der Völker eingegangen. Auch wenn wir die Menschenrechte nur universell denken können, müssen wir hinnehmen, dass das Konzept der Menschenrechte nicht so universell anerkannt ist. Jill Klindt recalls that back in 2008, when she joined Workiva, the software start-up was somewhat removed from the career path that she had envisioned for herself. “It wasn't that obvious to me at first, and it wasn't what I was looking for,” explains Klindt, who says that at the time, the Ames, Iowa, company employed six to eight people. Once on board, Klindt found that her accounting skills and willingness to problem-solve quickly made her a go-to executive along the entrepreneurial byways that ruled Workiva's early years. “I was relied on, which felt really good, and people kept bringing me opportunities, so I never felt passed over,” explains Klindt, who notes that even when an outside CFO was recruited to lead Workiva's 2014 IPO, she never felt displaced. “I would not have been ready for that role at that point in my career,” comments Klindt, who adds that her predecessor in the CFO office became an outstanding mentor to her, as did other members of Workiva's C-suite. Meanwhile, a talent-focused culture that took root during Workiva's early days has continued to thrive during the company's post-IPO life, says Klindt, who characterizes the culture as one where employees can ask others for help. Having stepped into the CFO office earlier this year, Klindt says that talent is now a primary concern when it comes to unlocking future growth at Workiva, where employees now number nearly 2,000. Says Klindt: “We want to be a much bigger company, and in order to do this, we need to keep hiring.” –Jack Sweeney

Abstract: Visual processing in the retina has been studied in great detail at all levels such that a comprehensive picture of the retina's cell types and the many neural circuits they form is emerging. However, the currently best performing models of retinal function are black-box CNN models which are agnostic to such biological knowledge. In particular, these models typically neglect the role of the many inhibitory circuits involving amacrine cells and the biophysical mechanisms underlying synaptic release. Here, we present a computational model of temporal processing in the inner retina, including inhibitory feedback circuits and realistic synaptic release mechanisms. Fit to the responses of bipolar cells, the model generalized well to new stimuli including natural movie sequences, performing on par with or better than a benchmark black-box model. In pharmacology experiments, the model replicated in silico the effect of blocking specific amacrine cell populations with high fidelity, indicating that it had learned key circuit functions. Also, more in depth comparisons showed that connectivity patterns learned by the model were well matched to connectivity patterns extracted from connectomics data. Thus, our model provides a biologically interpretable data-driven account of temporal processing in the inner retina, filling the gap between purely black-box and detailed biophysical modeling.
