Anxiety, depression, and other mental distress can be caused by pyrrole disorder (aka pyroluria). How can this loss of zinc, Vitamin B6, and other nutrients be identified and treated? “He suddenly developed anxiety, intrusive thoughts and images…[of] brutal things happening to his parents,” says Dr. Jingduan Yang MD, describing his then 9-year-old patient, who also suffered insomnia, restlessness, and low attention span. A simple urine test of the boy confirmed much higher than normal kryptopyrrole in his system – a problem Dr. Yang sees in 50 to 70 percent of his patients; often severely impacting their mental health. Dr. Yang is a psychiatrist and integrative medicine doctor who combines western medicine and holistic medicine, including acupuncture and advanced nutrient therapy in his practice. “People come in with all kinds of diagnoses–bipolar disorder, major depression, ADHD, borderline personality disorder, schizophrenia, schizoaffective [disorder], and all kinds of things,” he says. “But nobody…[asked if] there was an underlying biochemical imbalance contributing to a critical nutritional deficiency, he adds.” Dr. Yang joins Vital Signs with Brendon Fallon to highlight pyrrole disorder as an overarching cause of mental disorder.   Anxiety, anger, and stress are a normal part of the human experience. But if you find yourself unable to tolerate even the smallest amount of stress without feeling lots of anxiety and anger, you may have a little-known stress disorder known as Pyrrole disorder. Pyrrole disorder (also called Pyroluria) occurs when a biochemical abnormality results in the overproduction of pyrrole molecules. Pyrrole molecules are excreted continuously in the urine and are normal by-products of hemoglobin synthesis and other bodily processes. For the average healthy person, this is not a problem. However, for someone with Pyrrole disorder, nutrient deficiencies occur because these molecules have an affinity for zinc and vitamin B6, causing them to latch onto and excrete them in the urine before the body can adequately absorb them, leading to severe oxidative stress. Pyrrole disorder is connected to a plethora of symptoms and disorders such as extreme anger and rage, rapid mood swings, night owl tendencies, high anxiety, depression, morning nausea, alcoholism, and bipolar disorder. Pyrrole disorder is something I regularly test for in my clinic. It is typically seen in people of all ages and is often genetically inherited in Northern European people groups such as the Irish and Scandinavian populations. Abstract: Regulating desired loci in the genome with sequence specific DNA binding molecules is a major goal for the development of precision medicine. Pyrrole imidazole (Py Im) polyamides are synthetic molecules that can be rationally designed to target specific DNA sequences to both disrupt and recruit transcriptional machinery. While in vitro binding has been extensively studied, in vivo effects are often difficult to predict using current models of DNA binding. Determining the impact of genomic architecture and the local chromatin landscape on polyamide DNA sequence specificity remains an unresolved question that impedes their effective deployment in vivo. In this report we identified polyamide DNA interaction sites across the entire genome, by covalently crosslinking and capturing these events in the nuclei of human LNCaP cells. This method, termed COSMIC seq, confirms the ability of hairpin polyamides, with similar architectures but differing at a single ring position, to retain in vitro specificities and display distinct genome-wide binding profiles. These results underpin the development of Py Im polyamides as DNA targeting molecules that mediate their regulatory or remedial functions at desired genomic loci. The past few months I’ve been hearing more and more about people doing psychedelic mushrooms and having positive mental/spiritual effects. I have always been very sceptic to everything that effects my brain and that is out of my control, but it peaked my interest. I know they have been commonly used in rituals in many ancient cultures, especially in South America. I just wanted to get your thoughts on the matter. Pyrrole-imidazole alkaloids represent a unique family of natural products which are exclusively found in marine sponges, mainly the Agelasidae, Axinellidae, and Halichondridae families. The pyrrole-imidazole alkaloids are fascinating due to thier biogenetic relationship leading to a large, strucurally diverse family based on a common key metabolite, oroidin. Some of the pyrrole-imidazole alkaloids have interesting biological activities, e. g., potent antitumor or antibacterial activities. One of these chellenging alkaloids is dibromophakellstatin which was first isolated from Phakellia mauritiana. In this work an efficient five step synthesis of dibromophakellstatin is described providing the natural product in an overall yield of 10%. Based on a broad investigation of the reactivity of dipyrrolopyrazinone-type enamides, a novel three component imidazolidinone anellation was exploited as the key step of the total synthesis. With this findings the overall yield of the synthesis could increased up to 18%. Based on these results it was possible to develope the first enantioselective total synthesis of (-)-dibromophakellstatin in a chiral pool approach (10 steps, 3.5% overall yield). In a collaboration effort the antitumor activity of dibromophakellstatin was confirmed. It was found that only the natural enantiomer shows biological activity by an unknown mechanism. Encouraged by the biological activity of (-)-dibromophakellstatin, different cyclopropane variations of the tetracyclic phakellin- and isophakellin scaffolds were obtained by reaction of dichlorocarbene with tri- and bicyclic precursors. Some of these compounds werde found to have interesting new anti proliferative activities.
