Send us Fan MailHow far can pathologists take visual biomarker scoring before human vision becomes the bottleneck?In this episode, I talk with Doug Bowman. PhD, VP Precision Medicine at Indica Labs, about what happens when companion diagnostics move from traditional visual scoring into the era of AI-powered image analysis. Doug comes from a biomedical and electrical engineering background, with experience in microscopy, digital image analysis, pharma workflows, and now precision medicine at Indica Labs. That combination makes him a great person to talk to about how image analysis actually fits into real companion diagnostic development.We start with a very practical question: what is a companion diagnostic, and why is it becoming so important in precision medicine? Doug explains that companion diagnostics are developed alongside therapeutics to help identify which patients are most likely to benefit from a specific treatment, especially in more complex therapies like antibody-drug conjugates (ADCs). We use HER2 as an example, and from there we get into the real challenge: once a biomarker cutoff matters clinically, visual estimation around that cutoff becomes much harder than many people want to admit.That is where this conversation gets especially useful for pathologists and digital pathology trailblazers. We talk about the limits of human vision, why low or ultra-low biomarker expression is difficult to score consistently, and how AI helps at multiple levels of the workflow: slide QC, tissue classification, cell segmentation, membrane and cytoplasmic measurement, and spatial analysis. Doug makes the case that AI is not only a convenience here. In some cases, it is the only realistic way to capture the kind of quantitative information modern therapies need.We also get into one of the more interesting examples from the episode: the Trop2 story, where a ratio of cytoplasmic to membrane expression appears to predict therapeutic efficacy better than looking at one compartment alone. That kind of compartment-level quantitation is exactly where computational pathology becomes more than a digital version of what the eye already does. It starts uncovering measurements and signatures the eye cannot reliably extract on its own.Another important part of the discussion is workflow and regulation. Doug walks through how AI-powered companion diagnostics are developed from preclinical work, to human feasibility studies, to RUO or clinical trial assays, and eventually toward analytical and clinical validation with regulatory engagement happening early. We also talk about the Indica Labs and Leica Biosystems partnership, and why end-to-end capability matters when you are trying to build something clinically deployable rather than just analytically interesting.What I liked about this conversation is that it stayed grounded. We did not talk about AI as magic. Jack Kelly has been making waves in the pool since graduating from Brown University last year. The 3x NCAA finalist in the breaststroke events decided he would give pro swimming a go and moved to Austin to train with Bob Bowman. Since making the move, Kelly has progressed immensely, not only going best times in all 3 breaststroke distances, but showing a ton of consistency throughout the last year. He broke the 1:00 barrier for the first time in December, but has since dipped under it on 6 different occasions. He's also been under the 27-second barrier in the 50 breast twice and the 2:10 barrier once in the 200. He's qualified to swim all 3 events at the European championships in August. Kelly discusses what it's been like training with Bowman, racing in the Ivy League, and being a lifelong New York Knicks fan.

Ed Komoszewski, and Faith Has Its Reasons: Integrative Approaches to Defending the Christian Faith, co-authored with Kenneth D. Boa. Dr. Bowman holds the M.A. and Ph.D. degrees in biblical studies from Fuller Theological Seminary and South African Theological Seminary. After a 27-year tenure at Madison Memorial, Coach Collins is shifting his focus toward digital mentorship and building the next generation of leaders. We sat down with the veteran instructor and coach to discuss the "muck and grind" of a long career, the overlap between the classroom and the court, and what's next on his whiteboard. Interviewer: Coach, 27 years at one program is a rarity in today's coaching climate. When you look back at that first season in Madison compared to your final whistle last spring, what is the most profound change you've noticed? Coach Collins: The speed—not just of the players, but of the information. When I started, we were trading physical VHS tapes and drawing plays on napkins. Now, players have access to every NBA highlight and breakdown on their phones before they even hit the locker room. But while the technology changed, the "Human Element" remained exactly the same. You still have to look a kid in the eye and make them believe they are capable of more than they thought. The 27 years taught me that players don't care how much you know until they know how much you care. Interviewer: You've spent a significant portion of your career teaching Advanced Placement Statistics. How does a deep understanding of probability and data affect your late-game decision-making? Coach Collins: It's a double-edged sword. In the classroom, we talk about the Law of Large Numbers—the idea that as a sample size grows, the observed mean will get closer to the expected value. On the court, I know that a high-volume shooter is "due" for a make, or that our Effective Field Goal Percentage ($eFG%$) is higher when we touch the paint. But coaching is where the "Statistically Significant" meets the "Humanly Unpredictable." You can have a $95%$ confidence interval that a certain play will work, but if a teenager is having a bad day or loses focus for a split second, that $5%$ "error" happens. My background in stats helps me stay calm; it reminds me to focus on the Process rather than the outcome of a single possession. Interviewer: You've transitioned into a major role with platforms like TeachHoops.com, essentially coaching the coaches. What prompted the shift into the digital space? Coach Collins: It was about scale. At Memorial, I could impact 12 to 15 players a year. Through digital communities and podcasts, I can help a coach in Ireland or a youth director in San Francisco solve a problem in real-time. Coaching can be a very lonely profession—that "Alone in the Crowd" feeling is real. I wanted to build a "Digital Truth Room" where coaches could find the resources, sets like the Princeton or Shuffle Offense, and the community support they need to avoid burnout. Interviewer: We hear you're a man of many interests outside the gym—from high-end sports trading cards to planning trips to the Orlando theme parks. How do you "unplug" after a long season? Coach Collins: You have to find your "Magic" somewhere. For me, the focus required to analyze a Topps or Bowman release or the logistics of navigating a family trip to Disney provides a different kind of mental challenge. It's about balance.
