"Only Continued Silver Drawdowns From Above-Ground Stocks Can Balance Market In Next Few Years" We've talked with analysts, miners, and economists over the past few years to see if there's any relief on the horizon towards the current imbalance in the silver market. The Silver Institute and Metals Focus have reported the silver market being in a deficit for the past 5 years (5 years if the ETF flows are accounted for in the net balance, 3 years if they are not). Which at times has been funded by depletion of the above ground stockpiles. But with the silver mining industry still just starting to recover from a harsh 2 year bear market, while the junior side of the market in many cases still has yet to see significant investment inflows, Metals Focus just put out a report saying that only continued draw downs from the above-ground stockpile will be able to balance the market over the next few years. Gold Hits Another New All-Time High After Powell's Capitulation The gold market just set another new all-time record high this morning, and December futures are currently trading at $2,641. Of course this is coming on the heels of the Fed's aggressive decision on Wednesday to cut interest rates by 50 basis points rather than only 25. Following the announcement we saw both gold and silver spike higher, before finishing lower on the day. Yet as the news is being processed by the markets a few days later, gold has just broken out to a new record high, while silver is trading at $31,73. A intelixencia artificial ou IA, polas súas siglas, deunos moito do que falar e fíxonos aprender unha chea nos últimos tempos. Este concepto, que ás veces nos resulta algo ambiguo, xa é unha constante no noso día a día e asistentes como Siri e Alexa ou as recomendacións musicais de Spotify son un bo exemplo diso. "Aínda que poidamos facer un uso sinxelo de ChatGPT, esta ten moito potencial á hora de escribir os copies para as nosas publicacións.". "Na última edición, do IA Song Contest, podemos atopar a canción participante de Galicia no segundo posto". "DALL-E 2 defínese como un novo sistema de intelixencia artificial que pode crear imaxes realistas a partir dunha descrición de texto e mediante a posterior combinación de conceptos, atributos e estilos". Que é a intelixencia artificial? A intelixencia artificial defínese como unha "disciplina científica que se ocupa de crear programas informáticos que realizan operacións comparables ás que realiza a mente humana, como o aprendizaxe ou o razonamento lóxico". A IA xa está presente en sectores moi diversos. Na sanidade, a través de chatbots que nos preguntan polos nosos síntomas e realizan un diagnóstico; no transporte e loxística, para optimizar o tráfico; na agricultura, con análises predictivos que melloran o rendemento dos terreos; no ámbito comercial, mediante a súa previsión de vendas e recomendacións de produtos a usuarios; na educación, con ofertas personalizadas para optimizar a aprendizaxe; ou na banca, a través da predición de patróns do mercado para aconsellar a clientes e incluso detectar fraudes; e moitas outras posibilidades. Non obstante, tamén está a facer acto de presenza nun ámbito que, convencionalmente, críamos que estaba máis guiado polo corazón que pola razón: a arte. A IA que pinta: DALL-E 2 OpenAI é a responsable de que Chat GPT estea na boca de todos. Este é unha especie de chatbot de intelixencia artificial que responde ás nosas preguntas ou peticións máis disparatadas, desde a creación dun traballo académico de mil palabras sobre o cambio climático ata a resolución dunha ecuación. Esta mesma compañía, fundada por Elon Musk e outros investidores, e que afirma ter como obxectivo a promoción e desenvolvemento da IA amigable de maneira que beneficie á sociedade, tamén conta cunha vea máis artística: DALL-E. DALL-E 2 defínese como un novo sistema de intelixencia artificial que pode crear imaxes realistas a partir dunha descrición de texto e mediante a posterior combinación de conceptos, atributos e estilos. Esta IA, tería aprendido a relación entre imaxes e o texto usado para describilas. Utiliza un proceso chamado "difusión", que comeza cun patrón aleatorio de puntos e que gradualmente vai alterando cara a unha imaxe cando recoñece aspectos específicos dela. Está claro que as imitacións sairían pronto e xa contamos con outras propostas como IMAXEN de Google ou DALL-E Mini. Ademais, as redes sociais ou ferramentas que empregamos para preparar o noso contido para redes, xa conta con integracións de IA: por exemplo, en Photoshop podemos completar -e facilitar- a nosa edición. A plataforma ten unha ferramenta que, mediante unha descrición nosa, é capaz de crear formas, imaxes e todo o que nos poidamos imaxinar. Isto tamén o podemos ver noutras ferramentas de creación de contido como pode ser Canva. Chat GPT: algúns consellos se nos falta inspiración E isto, por suposto, ten a súa aplicación en redes sociais. Non só a través da publicación, por exemplo, das imaxes de DALL-E ou das creatividades xeradas con Photoshop ou Canva, senón co Chat GPT. Aínda que poidamos facer un uso sinxelo da plataforma, esta ten moito potencial á hora de escribir os copies para as nosas publicacións. ✔️ Estilo. É así que podemos aproveitarnos de extensións como Tweet GPT, que crea unha biografía e imaxe automática en función aos tuits que publicamos; YouTube Summary, que permite resumir vídeos de YouTube e artigos da rede; ChatGPT Writer, destinado a escribir emails e mensaxes, solventar eses erros de gramática que poidamos ter, cambiar o ton de escritura ou resumir, entre outras opcións; Fireflies, é un asistente de voz dixital que se une como participante ás conversas en Zoom e que axuda a transcribir, tomar notas e realizar accións durante as reunións; ou Engage AI, destinada a mellorar o alcance en LinkedIn a través da automatización e mellora de comentarios. ✔️MusicLM: do papel ao pentagram Google presentou un modelo que xera música de alta fidelidade a partir de descricións de texto. Que MusicLM sexa capaz de facer isto débese a un exhaustivo adestramento de arredor de 280.000 horas de música, o que lle permite discernir diferentes conceptos. Aínda que na súa web xa podemos escoitar varios exemplos diso, o usuario de a pé aínda non o pode probar xa que, por agora, Google non se plantexa lanzar esta ferramenta ao público. ✔️Unha cita para os eurofans: AI Song Contest Se es dos que agarda a que chegue Eurovisión como auga de maio, quizais che soe a cita eurovisiva con IA que se celebra dende 2020 nos Países Baixos. AI Song Contest é unha competición internacional que explora o uso de intelixencia artificial no proceso de composición musical na que poden participar tanto músicos como investigadores ou desenvolvedores. Na última edición, celebrada en xuño de 2022, participaron 46 equipos creadores de todo o mundo. To the outside world, cryptocurrency is that part of the internet where you can lose money to scams and rugpulls. It's where some YouTuber tells you about a token called PAMP or SAFEMOON that is about to PUMP 1000x and YOU NEED TO BUY RIGHT NOW TO GET ON THE ROCKETSHIP. Isn't that what crypto is? Bankless readers are aware that this is not the case. Bankless sees cryptocurrency as a land of opportunity, a new frontier, a digital world built on free and open-source software that allows us to break free from the Wall Street and Silicon Valley institutions that have enslaved us. So, why do most people believe cryptocurrency is full of scams? And why do people keep coming to learn about it if it's full of scams? What piques their interest? Or is cryptocurrency just a big con? Today's article introduces my "Concentric Circles of the Crypto Industry" model, which explains why so many people outside of the crypto industry believe it is a scam. This mental model can help crypto-newcomers navigate the Dark Forest of crypto with a little more ease, and with a little more understanding of the types of people you find in crypto — which ones to pay attention to, and which ones to avoid. There are four concentric circles. A set of four concentric circles is my model for who makes up the crypto industry. The Core Devs are in the centre, with the rest of the world on the outside and the Crypto Believers and Grifters in the centre. * The Core Developers. * Crypto enthusiasts. * The swindlers. * The remainder of the world. Core Developers Let's begin in the middle. The Core Devs are the architects and philosophers who are constructing this industry from the ground up. They have a thorough understanding of the world's current problems, how cryptography and blockchain technology can help to solve them, and the radical implications for the future. They understand the philosophy, code, and moral good that this technology has the potential to bring to the world. This is what we mean by "crypto values." In this circle, you'll find Satoshi Nakomoto, the Bitcoin's anonymous creator, who birthed the Bitcoin blockchain before slowly disappearing into the internet because he knew Bitcoin would be better without a leader. Vitalik Buterin, the creator of Ethereum, is also present. Vitalik is a digital monk, a master of cryptography, philosophy, math, and, of course, crypto-economics. Vitalik aspires to create public goods and social systems that benefit the entire world. Even though he is extremely wealthy, he has been known to sleep in hostels and live out of a single backpack in order to avoid consuming more resources than he requires. He is a crypto-monk for the modern era. Working as a Core Dev can be a thankless task. Building open-source software is frequently under-appreciated and compensated in proportion to the value it adds to the world. Every day, Bitcoin Core developers work to improve Bitcoin. Every week, Ethereum core developers meet in public to discuss what to work on next and what society needs from the foundations they're laying. These people are here because they believe in the mission, see a brighter future ahead, and want to help build it. The Core Devs are the purists at the heart of the cryptocurrency universe, and their leadership creates the ultimate gravitational pull. These are the revolutionaries with good hearts and sound minds, and they are fighting for a cryptographic-power future... and they're all huge nerds in the best way possible. We are here to help them... and they are here to help us. Believers in cryptocurrency The Crypto Believers are the next to speak up. These are the users of crypto power. These people believe in the same future that the Core Devs do, and they live on the foundations that the Core Devs have laid. However, this circle is not limited to users! Application developers, DAO members, and businesses are all creating interwoven layers of products and services based on these new protocols. Ethereum is a protocol around which we all organise. Apps like Uniswap, Aave, and Maker are built with smart contracts on the Ethereum app layer. Ethereum is used to organise DAOs such as BanklessDAO, PleasrDAO, and even Constitution DAO. Companies such as venture funds, news agencies, and media companies, such as Bankless, also organise around Ethereum. This circle is populated by locals rather than tourists. These are crypto citizens who are constructing structures in this new frontier, testing products in their early stages, gradually becoming bankless by managing their money and property on crypto rails, and creating new digital identities for themselves. These settlers have come to live in an open and free metaverse that has been built with open-source software and the open-source ethos. These are people like Rune Christensen, who saw the need for a decentralised dollar and developed the MakerDAO vision... Before DAOs even existed! Now, billions of DAI exist to escape the mismanagement of their government's currency. Or Hayden Adams, who built Uniswap, a public and free asset exchange system, entirely on his own after learning to code and with the help of a $10,000 grant from the Ethereum Foundation. The values built into the base level protocols by the Core Devs are expressed in the applications built on top of them at this layer of cryptography. Then there are people like Cami Russo, who lived in Argentina and saw firsthand how the Argentine Peso's hyperinflation created demand for crypto-related products. DAI, the crypto-native stablecoin, has seen widespread adoption in Argentina because it is the only tool available to Argentines to avoid 50-100% inflation rates. Cami studied journalism and founded the media publication The Defiant after becoming crypto-pilled. Another of my favourite crypto believers is Anthony Sassano. He creates a video in which he informs the world about what has occurred in the Ethereum ecosystem in the last 24 hours. Every single day. This is the layer of cryptocurrency that broadcasts the industry's progress. Because they are focused on building the future, protocol core developers and application builders are not very good at marketing themselves. To accomplish this, the crypto industry relies on the surrounding community. And this surrounding community exists only because we are all here for the same reasons: the belief that cryptocurrency is here to help build a better and more free world... and that it is a necessary step for humanity's future progress. This is the world of cryptocurrency that many people do not see or understand. And one of the main reasons for this is that the Crypto Grifters keep it separate from the rest of the world. Crypto swindlers There is an obstacle course between crypto believers and the rest of the world... an asteroid belt of Crypto Grifters who make it difficult to hear the signal coming from the crypto industry's true spearhead Grifters are loud, self-aggrandizing, and arrogant. They use polarising tactics and styles that have proven to be effective in politics. Crypto swindlers aren't stupid; they know there's a lot of money to be made on the crypto frontier... and they're here to take advantage of it. To save money. To take shortcuts and the easy way out. They will sell you fool's gold while stealing your money. Crypto swindlers are the reason crypto has a bad reputation. They're louder and more bombastic than regular cryptocurrency users. They promote themselves more than they develop technology. They frequently don't care about the technology they are developing; all they care about is making money from it, regardless of how sustainable or ethical it is. Crypto Grifters purposefully create malicious products in order to catch the next inexperienced crypto noob. They set up paid Telegram channels to share "alpha," but instead just dump on you. They create complicated DeFi projects that, if you fall into their traps, merely transfer money from your pocket to theirs. Most Grifters appear in bull markets and imitate what's popular at the time. In 2017, they created fake ICOs, and in 2021, they created low-effort NFT projects with Fiver. Wherever there's a profit to be made, the Grifters smell it and rush in, ready to grift. Crypto swindlers share a few characteristics: An egotistical personality... someone who is large, bombastic, and can be a bully at times. A small, but highly engaging and manic community forms around these people, a kind of personality cult. They recite the messages, say the lines, and shill a token. They are the classic mix of humans and bots that we've come to recognise in the world of modern social media. A product or system that is fundamentally unsustainable... even if it does not appear to be so at the time. A gleaming new crypto product that simply does not make sense under the hood... and eventually has a date with fate. Mashinsky, Alex Alex Mashinsky founded and ran Celsius, a custodial borrowing and lending service. A centralised company that accepted customers' crypto-assets and paid them high interest rates for their deposits. This is a typical business, and it is known as a bank. And there are many legitimate crypto products and services that do this. Mashinsky, on the other hand, carried the DeFi banner and boasted about how DeFi will bring down the Banks. But he did build a bank! Celsius accepted customer deposits, used completely reckless and leveraged risky trading strategies, and gambled away other people's money. Sesta, Daniel The Wonderland ecosystem was built by Daniele Sesta. An army of Pepe Frog accounts trailed Sesta around the internet, swarming him wherever he went. You name it: cryptocurrency Twitter, YouTube comments, livestream chat boxes... They invaded and infested the digital spaces where crypto-people spend their time, as well as our minds, because they were so goddamn loud. This strategy has been used before, both inside and outside of crypto, but when money is at stake, these internet armies can become deafening. Anyway, Wonderland collapsed when people realised that an unbacked stablecoin is just an obfuscated ponzi scheme, and also when one of the project's anonymous co-founders was revealed to be Michael Patryn, a convicted money launderer and co-founder of the QuadringaCX exchange, the one with the mysterious death of the founder, after the exchange became insolvent. Kwon Do Then there was Do Kwon, Terra's eccentric and aggressive founder. The largest capital destruction event in crypto history, with $50 billion in capital destroyed. The Terra ecosystem abruptly lost all of its incoming money flows, revealing itself to be an unsustainable structure... ...which, in retrospect, appears to be a massive Ponzi scheme. Do Kwon, like Sesta, had a massive army of self-described "Lunatics." Ryan and I were extremely sceptical of the Terra Luna project, and when we voiced our concerns and dissatisfaction, we were harassed on Twitter by these raving lunatics. Do Kwon's actions made this possible. As Terra pumped, Do Kwon became louder and more aggressive on Twitter, rallying his followers. Do we have to put up with the swindlers? Both yes and no. We can't directly stop the swindlers. That's the deal we make when we prioritise permission-lessness above all else. Access for all is a core value that cannot be compromised. Crypto, like the internet, does not require permission to use... It's a money and finance public utility that anyone with an internet connection can use. And as the financial world of DeFi grows and improves organically as more people use it, these public utilities become more useful over time. Unfortunately, the same property of permission lessness makes it extremely difficult to stop grifters from grifting. Permission-less financial innovation enables us — you and me — to break free from the financial prisons erected by banks and Wall Street, but it also makes it difficult to prevent others from making unethical money. We can't stop them, but we don't have to live with them. We combat them through education. As an industry, we must improve our ability to reach the masses before the swindlers do. Massive marketing campaigns that use grandiose and bombastic tactics are difficult to compete with. Understanding what makes this industry tick is more difficult than simply listening to some charismatic individual telling you to buy their token. The good news is that most people today have a basic instinct to avoid spam emails and viruses on the internet. Crypto will eventually be no different. Bankless is here to confront the swindlers and ensure that the good being built here is communicated to the rest of the world, so that the rest of the world knows that Crypto is here to set you free... and that we despise swindlers just as much as you do. Stāsta folkloras kopas "Tarkšķi" un mandolīnistu ansambļa "Tarkšķu mandolīnisti" vadītāja Kristīne Karele Mūsdienās vārds "mandolīna" kādam saistās ar Itāliju, kādam ar Lūisa de Bernjēra grāmatu un lielisko filmu "Kapteiņa Korelli mandolīna" ar Penelopi Krusu un Nikolasu Keidžu galvenajās lomās. Kaut arī pēdējā piecgadē mandolīnu – šo interesanto strinkšķināmo instrumentu - iemīļojuši un sākuši spēlēt krietns pulciņš Latvijas iedzīvotāju, nevarētu teikt, ka tas ir plaši atpazīstams un iecienīts mūzikas instruments. Pirms gadiem desmit situācija ar mandolīnu bija pavisam pamirusi – to nespēlēja folkloras kopas, to nespēlēja tautas mūzikas kapelas, nedz arī citi muzikanti. Kad kāds etnomuzikologs tās spēli mēģināja citiem demonstrēt, tad lielākoties klausītāji mandolīnu neatpazina vai sauca par slāvu instrumenta balalaikas māsu. Tomēr, pārcilājot vecus preses materiālus un senas fotogrāfijas, nu jau droši varam apgalvot, ka mandolīna ir pilntiesīgs latviešu tautas mūzikas instruments – pagājušā gadsimta sākumā un līdz pat 50. gadiem tas bijis tautā iemīļots. Gan tāpēc, ka viegli apgūstams, gan tāpēc, ka salīdzinoši lēts un arī – ērti pārnēsājams, jo neliels izmērā. Par to, ka droši mandolīnas varam pieskaitīt mūsu tautas mūzikas instrumentu pulkam, liecina vairākas pazīmes. Kā raksta etnomuzikologs Ilmārs Pumpurs biedrības "Skaņumāja" izdevumā "Tradicionālās mandolīnas spēle", Latvijā mandolīnas plaši sāka parādīties 19. gadsmita beigās. Sākotnēji vairāk pilsētas smalkajā sabiedrībā, bet pavisam drīz arī tradicionālo mūzikas ansambļu jeb kapelu sastāvos. Piemēram, 20. gadsimta 30. gados Rīgas krievu prese rakstīja par "vectēvu laiku" kadriļu dejošanu pie "pašķidras mandolīnas strinkšķināšanas". 20. gadsimta sākumā mandolīnu cienītāju skaits jau bija pieaudzis tiktāl, ka tie sāka apvienoties mandolīnu ansambļos un pat orķestros. Ar brīnišķīgu bildi nesen Facebook dalījās Saldus vēstures un mākslas muzejs: 1928. gada attēlā redzams Pampāļu pamatskolas mandolīnu orķestris patiešām iespaidīgā sastāvā – vairāk nekā 50 jauniešu! Arī Iecavā darbojušies vismaz divi mandolīnu orķestri – ir gan fotogrāfijas, gan arī stāsti no cilvēkiem, kas tajos muzicējuši. Šķiet, vai ikviens Latvijas iedzīvotājs arī var lepoties ar kādu senci, kas reiz bērnībā spēlējis mandolīnu vai atceras, ka viņa radagabals vai kaimiņš labprāt strinkšķinājis to. Kaut arī Latvijā daudz spēlētas Vācijā, Itālijā un citur gatavotās mandolīnas, arī šeit, uz vietas, kā raksta Ilmārs Pumpurs, darbojušies vairāki instrumentu meistari – A. Ieviņš, V. Saulītis, O. Zutis, kā arī salīdzinoši daudzi lauku amatnieki. Tam kā pierādījums ir visā Latvijā atrastie instrumenti. Tas ir vēl viens apliecinājums tam, ka mandolīnas bija ļoti iecienīts tautas mūzikas instruments vēl pirms 100 gadiem. Līdz ar Ilmāra Pumpura un biedrības "Skaņumāja" aktivitātēm mandolīnas kā tautas instrumenta izplatība šobrīd Latvijā ir atdzimusi – tās spēli apgūst bērni tautas mūzikas nometnēs, folkloras kopās, arī pieaugušie tautas mūzikas instrumentu kursos. Mandolīna atkal atradusi vietu tautas mūzikas kapelās, kur tiek spēlēta kopā ar vijolēm, basi, ermoņikām, kā arī ar lielu mīlestību un aizrautību skan manis vadītajā mandolīnistu ansamblī "Tarkšķu mandolīnisti", kur darbojas ap 15 bērnu un jauniešu, kurus vieno mīlestība uz šo savdabīgi un interesanti skanošo tautas mūzikas instrumentu - MANDOLĪNA.

Abstract: Pathogen-associated molecular patterns (PAMP)-triggered immunity (PTI) and effector-triggered immunity (ETI) are designated as a two-branched system of innate immunity. Although PTI/ETI share a series of downstream molecular events, systematic analysis of convergent and divergent signaling in PTI/ETI is currently lacking. The phytohormones salicylic acid (SA) and jasmonic acid (JA) are considered to constitute the hormonal backbone of plant immunity, are functionally antagonistic, and play essential roles in defending against biotrophic and necrotrophic pathogens, respectively. However, the distinct performance of two phytohormones in PTI/ETI remains unclear. Here, we systemically investigate and validate the reprogramming of molecular networks during PTI and ETI. Using publicly available Arabidopsis RNA sequence data from 560 samples, we construct a co-expression network under Mock conditions and then explore the differential expression/co-expression changes during PTI, ETI, and Pto DC3000 infection. During PTI and ETI, one-third of genes in the Arabidopsis genome exhibit the same directional differential expression in a manner independent of JA/ethylene/PAD4/SA signaling but show differential co-expression patterns. However, the defense phytohormone network is required for defense against Pto DC3000 infection. Kvinnors musik på väg! vill undersöka hur det står till med kvinnor och konstmusik tio år efter det att P2 2006 sände den legendariska programserien Konsten att humla. Idag möter vi tre musikkvinnor som, istället för att sitta i sitt elfenbenstorn och komponera verk för evigheten, valt att samarbeta med andra konstnärer och att vara omgivna av fler personer i den kreativa processen. Multikonstnären Paula af Malmborg Ward - tonsättare, arrangör, dirigent, sångerska, pianist, sitter visserligen i ett torn och komponerar, men det är ett renoverat kokstorn vid inloppet till Göteborgs hamn där fiskmåsar, trutar och Stena Lines hornstötar delar på det akustiska utrymmet utanför fönstret. De har inte alls fått samma uppmärksamhet, de har inte blivit spelade och lyssnade till. Det är ju först när musik får gå igenom andra som den kan växa och utvecklas och bli nåt större än tonsättaren själv och de noter hen skrivit på papperet. Musiken blir då en del av universum, säger Mika i programmet. Mika är frustrerad över hur få timmar som ägnades åt kvinnliga tonsättare under alla hennes utbildningar i Göteborg, Prag och Köpenhamn. Hon har spelat viola i orkestrar sedan hon var tio år. - Det finns tydliga och plågsamma maktsystem och hierarkier inom symfoniorkesterkulturen. Det handlar om vem som skall bestämma över repertoaren. Oftast väljs musik av män. Den som dirigerar musiken och bestämmer hur den skall spelas är nästan alltid en man. Det finns även hierakiska system i stämmorna. Vem bestämmer hur stämman skall spelas? Ja, det finns tydliga maktroller i hela orkestern, berättar Mika Persdotter Svensson i programmet. Norska tonsättaren Cecilie Ore studerade piano vid Norges Musikhögskola och i Paris och därefter komposition i Utrecht och hos Ton de Leeuw Amsterdam. Hon vann International Rostrum for Electro-Acoustic Music och är intresserad av tidsaspekter i musik. På 2000-talet har hon mer och mer arbetat med textbaserad och samhällskritisk musik. När tonsättaren Xenakis avled 2001 stod den norske poeten Paal-Helge Haugen utan tonsättare till sin skuggopera A. Cecile Ore fick ta över uppgiften efter Xenakis. Hennes verk har framförts av BBC Symphony Orchestra, Ensemble Modern, BBC Singers samt en rad norska och nordiska ensembler. Så här sa Cecilie Ore i Konsten att humla 2006: -Det är mycket svårare att vara medelmåttig kvinnlig tonsättare än att vara medelmåttig manlig komponist. Och det går inte alls an att vara en dålig kvinnlig tonsättare. De kvinnliga tonsättare som godkänns har en mycket hög konstnärlig nivå. Girighet - män äger 99% av världens rikedomar. Kvinnor endast 1 %. Frosseri - män frossar i sexuella övergrepp och våldtäkter. Avund - män är svartsjuka och täcker kvinnor i burka, niqab och liknande. Lättja - prostitution och trafficking, där män låter kvinnor arbeta för sig och tar profiten. De spelar fem kompositioner från 1100-talet fram till idag: av Hildergard av Bingen, Wilhelmina av Preussen, Elfrida Andrée, Frangis Ali Zade och Ellen Lindquist. - Det är så sorgligt att tänka på hur litet kärlek kvinnliga tonsättare har fått genom historien. De har inte alls fått samma uppmärksamhet, de har inte blivit spelade och lyssnade till. Det är ju först när musik får gå igenom andra som den kan växa och utvecklas och bli nåt större än tonsättaren själv och de noter hen skrivit på papperet. Musiken blir då en del av universum, säger Mika i programmet. Mika är frustrerad över hur få timmar som ägnades åt kvinnliga tonsättare under alla hennes utbildningar i Göteborg, Prag och Köpenhamn. Hon har spelat viola i orkestrar sedan hon var tio år. - Det finns tydliga och plågsamma maktsystem och hierarkier inom symfoniorkesterkulturen. Det handlar om vem som skall bestämma över repertoaren. Oftast väljs musik av män. Den som dirigerar musiken och bestämmer hur den skall spelas är nästan alltid en man. Det finns även hierakiska system i stämmorna. Vem bestämmer hur stämman skall spelas? Ja, det finns tydliga maktroller i hela orkestern, berättar Mika Persdotter Svensson i programmet. Norska tonsättaren Cecilie Ore studerade piano vid Norges Musikhögskola och i Paris och därefter komposition i Utrecht och hos Ton de Leeuw Amsterdam. Hon vann International Rostrum for Electro-Acoustic Music och är intresserad av tidsaspekter i musik. På 2000-talet har hon mer och mer arbetat med textbaserad och samhällskritisk musik. När tonsättaren Xenakis avled 2001 stod den norske poeten Paal-Helge Haugen utan tonsättare till sin skuggopera A. Cecile Ore fick ta över uppgiften efter Xenakis. Hennes verk har framförts av BBC Symphony Orchestra, Ensemble Modern, BBC Singers samt en rad norska och nordiska ensembler. Så här sa Cecilie Ore i Konsten att humla 2006: -Det är mycket svårare att vara medelmåttig kvinnlig tonsättare än att vara medelmåttig manlig komponist. Och det går inte alls an att vara en dålig kvinnlig tonsättare. De kvinnliga tonsättare som godkänns har en mycket hög konstnärlig nivå. Avund - män är svartsjuka och täcker kvinnor i burka, niqab och liknande. Lättja - prostitution och trafficking, där män låter kvinnor arbeta för sig och tar profiten. Hace tiempo que Iago de Radio Bertoldo anunció su intención de realizar un especial cómics y arquitectura en su podcast sobre banda diseñada, iniciativa muy celebrada por varios de los que formamos parte de LMEY. Del contacto entre ambos, y también con el Sr. B de Metrópoles Delirantes, surgió este crossover que nos ha hecho especial ilusión y al que como uno más de La Morsa se unió José Ramón Hernández Correa de Arquitectamos locos?. Bacterial pathogens causing systemic disease commonly evolve from organisms associated with localized infections but differ from their close relatives in their ability to overcome mucosal barriers by mechanisms that remain incompletely understood. Here we investigated whether acquisition of a regulatory gene, tviA, contributed to the ability of Salmonella enterica serotype Typhi to disseminate from the intestine to systemic sites of infection during typhoid fever. To study the consequences of acquiring a new regulator by horizontal gene transfer, tviA was introduced into the chromosome of S. enterica serotype Typhimurium, a closely related pathogen causing a localized gastrointestinal infection in immunocompetent individuals. TviA repressed expression of flagellin, a pathogen associated molecular pattern (PAMP), when bacteria were grown at osmotic conditions encountered in tissue, but not at higher osmolarity present in the intestinal lumen. TviA-mediated flagellin repression enabled bacteria to evade sentinel functions of human model epithelia and resulted in increased bacterial dissemination to the spleen in a chicken model. Collectively, our data point to PAMP repression as a novel pathogenic mechanism to overcome the mucosal barrier through innate immune evasion.

Ein Ziel der vorliegenden Arbeit war es, die biologische Aktivität des kürzlich von Schneider et al. klonierten, rekombinanten Hühner IL-6 (rChIL-6), zu charakterisieren. In vitro wurde der Nachweis der biologischen Aktivität von rChIL-6 durch die Induktion der Proliferation der streng IL-6 abhängigen murinen Zelllinie 7TD1 erbracht. Diese Tatsache wurde für die Etablierung eines quantitativen rChIL-6 Nachweissystems genutzt. Für weiterführende Studien wurde ein neutralisierendes, polyklonales ChIL-6 Antiserum entwickelt. Durch wiederholte Vakzinierung von Kaninchen mit rChIL-6 (E.coli) war es möglich, ein ChIL-6 Antiserum zu gewinnen, welches auch die biologische Aktivität von natürlichem ChIL-6 neutralisiert. In vivo induzierte die intravenöse Applikation von rChIL-6 (E.coli) einen deutlichen Anstieg der Kortikosteronkonzentration im Serum von Hühnern. Somit standen mit biologisch aktivem rChIL-6, einem ChIL-6 Nachweistest und einem neutralisierenden Antiserum geeignete Werkzeuge zur Verfügung, die für erste Untersuchungen zur funktionellen Charakterisierung des aviären Toll-like Rezeptor (TLR)-Systems genutzt wurden. Diese, unter anderem auf Makrophagen exprimierten Rezeptoren, sind für eine adäquate Reaktion des Immunsystems von essentieller Bedeutung, denn sie erkennen die Anwesenheit pathogener Mikroorgansimen anhand so genannter „pathogen-associated molecular pattern“ (PAMP). Eine Konsequenz der Rezeptoraktivierung ist die Transkription von Genen pro-inflammatorischer Cytokine, unter anderem von IL-6. Primäre Hühnermakrophagen wurden durch Inkubation mit verschiedenen, aus dem Säuger-System bekannten TLR-Agonisten stimuliert; als Nachweissystem der erfolgreichen Aktivierung wurde die quantitative Analyse der ChIL-6 Sekretion genutzt. Zunächst wurden primäre Hühnermakrophagen mit LPS stimuliert worauf sie mit einer starken ChIL-6 Sekretion reagierten. Durch die gleichzeitige Inkubation der Makrophagen mit LPS und IFN- ließ sich das Ausmaß der ChIL-6 Sekretion deutlich steigern. Auch die Inkubation mit bakterieller DNA induzierte die Sekretion von ChIL-6. Dieser Makrophagen-aktivierende Effekt war DNA-spezifisch, denn zum einen beendete der DNA Verdau mit DNase I diesen Effekt. Zum anderen induzierten CpG-ODNs nicht aber GpC-ODNs eine starke ChIL-6 Sekretion. Weiterhin führte die Stimulation der Makrophagen mit synthetischem bakteriellem Lipopeptid und synthetischer dsRNA zu einer deutlichen Sekretion von ChIL-6. Bei in vivo Studien wurde eine deutliche Induktion von ChIL-6 durch LPS (TLR 4) beobachtet. Im Rahmen dieser Arbeit konnte also gezeigt werden, dass rekombinantes ChIL-6 sowohl in vitro als auch in vivo biologisch aktiv ist. Auf der Basis der dabei etablierten Methoden konnte bei ersten Untersuchungen zur funktionellen Charakterisierung des TLR-Systems des Huhnes Hinweise dafür gewonnen werden, dass beim Huhn funktionelle Homologe zu den, aus dem Säuger bekannten TLR 2, 3, 4, und 9 vorhanden sind
