A., Olesen, J. B., Smaldino, P. J., Rubenstein, E. M. Abstract: The relationship between lipid homeostasis and protein homeostasis (proteostasis) is complex and remains incompletely understood. We conducted a screen for genes required for efficient degradation of Deg1-Sec62, a model aberrant translocon-associated substrate of the endoplasmic reticulum (ER) ubiquitin ligase Hrd1, in Saccharomyces cerevisiae. This screen revealed that INO4 is required for efficient Deg1-Sec62 degradation. INO4 encodes one subunit of the Ino2/Ino4 heterodimeric transcription factor, which regulates expression of genes required for lipid biosynthesis. Deg1-Sec62 degradation was also impaired by mutation of genes encoding several enzymes mediating phospholipid and sterol biosynthesis. The degradation defect in ino4{Delta} yeast was rescued by supplementation with metabolites whose synthesis and uptake are mediated by Ino2/Ino4 targets. Stabilization of a panel of substrates of the Hrd1 and Doa10 ER ubiquitin ligases by INO4 deletion indicates ER protein quality control is generally sensitive to perturbed lipid homeostasis. Further, loss of INO4 sensitized yeast to proteotoxic stress, suggesting a broad requirement for lipid homeostasis in maintaining proteostasis. Abundance of the ER ubiquitin-conjugating enzyme Ubc7 was reduced in the absence of INO4, consistent with a model whereby perturbed lipid biosynthesis alters the abundance of critical protein quality control mediators, with broad consequences for ER proteostasis. A better understanding of the dynamic relationship between lipid homeostasis and proteostasis may lead to improved understanding and treatment of several human diseases associated with altered lipid biosynthesis. Andy Niekamp, or more commonly known as Captain Blue, joins Jester this week to chat about his hiking adventures, volunteering, and his company, Outdoor Adventure Connection. Andy has over 30,000 miles of hiking experience, with more than 11,000 of those miles being on the Appalachian Trail. Andy has completed the AT five times as a section hiker with his most recent completion in 2021. Andy was the first solo thru hiker to complete the 1444 mile Buckeye Trail in 2011. Andy published a book about his hike titled: Captain Blue on the Blue Blazes, The First Solo Thru-Hike of Ohio's 1,444 Mile Buckeye Trail. Tyler Niekamp is set to begin his 10th season as the head basketball coach at Van Buren High School in 2020-2021. The northwest Ohio native grew up and attended high school in Ft. Recovery, Ohio, He attended college and played basketball at the University of Findlay. Upon his graduation from Findlay, Tyler spent two seasons as a graduate assistant coach in the Oiler program before beginning his teaching and coaching career at Upper Scioto Valley High School. After two years as the junior varsity coach at USV, Coach Niekamp was positioned to become the head coach. During the summer, Coach Niekamp and I actually spent time planning for me to join his staff as an assistant coach. However, when an opportunity arose for Tyler to take a teaching job with Van Buren schools, just an 8-minute drive from his house, he opted to change jobs. The change in jobs also meant walking away from a head coaching position to be the freshman coach at Van Buren. Ultimately, Coach Niekamp became the head coach at Van Buren and today he shares the story of his journey in coaching as well as his vision for Van Buren Basketball. This week on Inside the Headset we speak with Travis Niekamp, Defensive Coordinator for Illinois State University. We discuss the transformation of the Illinois State football program, his development as a coach on both sides of the ball, and how he strives to prioritize his family while also being a coach. Coach Niekamp's career began at his Illinois State, his alma mater, as an outside linebackers and safeties coach (1998-1999). In 2000, he joined the staff at Army working with the tight ends (2000-2003). Niekamp served as the linebackers coach and special teams coordinator at Eastern Washington (2004-2007) before moving to Washington State to coach linebackers (2008-2011). He later joined Louisiana-Monroe to initially coach the tight ends, but crossed back over to the defensive side to coach linebackers and special teams after his first season, and he eventually served as the co-defensive coordinator (2012-2015). Niekamp coached linebackers and special teams at Montana (2016-2017) prior to being named the defensive coordinator at Illinois State in 2018.
