Abstract: Human cognitive abilities, and particularly hippocampus-dependent memory performance typically decline with increasing age. Immunosenescence, the age-related disintegration of the immune system are increasingly coming into the focus of research as a considerable factor contributing to cognitive decline. In the present study, we investigated potential associations between plasma levels of pro- and anti-inflammatory cytokines and learning and memory performance as well as hippocampal anatomy in young and older adults. Plasma concentrations of the inflammation marker CRP as well as the pro-inflammatory cytokines IL-6 and TNF- and the anti-inflammatory cytokine TGF-{beta}1 were measured in 142 healthy adults (57 young, 24.47 +/- 4.48 years; 85 older, 63.66 +/- 7.32 years) who performed tests of explicit memory (Verbal Learning and Memory Test, VLMT; Wechsler Memory Scale, Logical Memory, WMS) with an additional delayed recall test after 24 hours. Hippocampal volumetry and hippocampal subfield segmentation were performed using FreeSurfer, based on T1-weighted and high-resolution T2-weighted MR images. When investigating the relationship between memory performance, hippocampal structure, and plasma cytokine levels, we found that TGF-{beta}1 concentrations were positively correlated with the volumes of the hippocampal CA4-dentate gyrus region in older adults. These volumes were in turn positively associated with better performance in the WMS, particularly in the delayed memory test. Our results support the notion that endogenous anti-inflammatory mechanisms may act as protective factor in neurocognitive aging. Copy rights belong to original authors. Eine Beeinträchtigung kognitiver Parameter lässt sich u.a. durch die Durchführung von Sakkadenaufgaben nachweisen, wo beispielsweise Schizophreniepatienten hier spezifische Defizite zeigen. Die Ausführung von Aufgaben des Antigap Paradigmas erfordert die zeitlich und räumlich korrekte Ausführung von Antisakkaden. Dazu müssen sowohl die reflexgesteuerten Reaktionen inhibiert als auch gleichzeitig eine willentliche Handlung ermöglicht werden. Hier wurden bei Schizophrenie verlängerte Latenzen sowie erhöhte Fehlerraten beschrieben. Mit dem memory- Paradigmus wurden die Fähigkeiten zur Unterdrückung von reflexiven Sakkaden unter overlap- Bedingungen ermittelt, sowie gedächtnisgetriggerte und –gesteuerte memory- Sakkaden aufgezeichnet. Beim Memory Test wurden einerseits die Fähigkeit untersucht, auf einen Punkt zu schauen und sich gleichzeitig die Position eines weiteren zu merken (Ultrakurzzeitgedächtnis). Die Fähigkeit der Perzeption und sensomotorischen Integration (Abgleichen visueller Information mit vorgespeicherten Bildern im Gehirn), die Repräsentation des Gedächtnisstimulus im Arbeitsgedächtnis sowie die Fähigkeit der Regression (rückführende Augenbewegung auf bereits bekannte visuelle Objekte), ermöglichen die Positionserinnerung und Ausführung des Memory Test. Es wird davon ausgegangen, dass das glutamaterge System in die Pathophysiologie der Schizophrenie ursächlich involviert ist. So weisen u.a. post mortem Studien auf eine Beteiligung der AMPA- Rezeptoren als Teil des glutamatergen Systems hin. Das GRIA1 Gen, welches den AMPA1- Rezeptor kodiert, ist zusätzlich in einer mit Schizophrenie gekoppelten Region lokalisiert. Dazu wurden bei 60 Patienten und 108 Kontrollprobanden die Augenbewegungen während der Durchführung des Antigap- und Memory- Tests mit Hilfe einer VOG- Brille aufgezeichnet und ausgewertet und eine Assoziation zur Genotyp- und Allelfrequenz der SNPs rs578772 und rs548294 im GRIA1-Gen überprüft. Die Ergebnisse zeigen für den Polymorphismus rs578772 im Antigap Test eine Reduktion der zeitlichen Flexibilität assoziiert mit den A-Allelträgern bzw. AA-Genotyp sowohl in Form einer verzögerten Reaktion als auch einer erhöhten Fehlerrate durch verfrüht ausgeführte Antisakkaden. Das Gleiche läßt sich auch im Memoryparadigma nachweisen. Für den Polymorphismus rs548294 wurden im Antigap Test sowohl zeitliche als auch räumliche Abweichungen registriert. Eine erhöhte Latenz durch verzögerte Reaktionszeit bei gleichzeitig räumlich nicht korrekt ausgeführter Antisakkade assoziiert mit A-Allelträgern und AA-Genotyp. G-Allelträger fallen weniger durch zeitliche, dafür aber durch räumliche Abweichungen im Antigap Test auf. Beim Memoryparadigma war eine zeitlich inkorrekte Ausführung der Sakkaden mit AG-Allelträgern und Genotyp AG assoziiert. Räumlich und zeitlich inkorrekte Antisakkaden assoziieren mit AA-Allelträgern und AA-Genotyp. Die Studie konnte den Nachweis eines Zusammenhangs des GRIA1 Gens mit den Sakkadenleistungen erbringen. Eine Erweiterung dieser Studie in Hinblick auf eine erhöhte Stichprobenzahl sowie den Einschluss weiterer genetischer Variationen ist im Anschluss an diese Pilotstudie möglich, um die Validität der Ergebnisse zu festigen.
