In this episode Kieran and I pick back up with our coverage of Michael Apted's Seven Up documentary series, with a conversation about '28 Up' from 1984. One note on this, we recorded this episode ages ago- back in September 2020 (in the period between the first and the second lockdown). Sorry this took so long to go live but it's a great chat. Oh, one more thing- this was the first 1980s episode we recorded (so this is Kieran's first time hearing this series' sign-off at the end). Gene Hackman e Mary Elizabeth Mastrantonio sono padre e figlia, ma anche due brillanti avvocati, che si vedranno su fronti opposti in aula di tribunale, di fronte ad una causa che potrebbe mettere in serio imbarazzo una importante casa automobilistica, rea di avere messo in commercio auto difettose, causando incidenti mortali. Per i due sarà l'occasione di mettere alcuni punti sul loro mai facile rapporto, specialmente dopo la morte della moglie/madre, che aveva sempre fatto da cardine di unione per la famiglia. Padre e figlia saranno costretti ad affrontare reciproche colpe, accuse personali e mancanze, in un lungo e difficile percorso per ritrovare, forse, equilibrio e stima. Due protagonisti straordinari, che fanno a gara di bravura, in quest'ottimo thriller di Apted, che tiene incollati allo schermo fino alla fine, prendendo la strada parallela del rapporto padre e figlia e narrandolo in maniera mai banale o scontata. Ci si commuove anche, promesso e non saprete per chi tifare. Curioso il tema trattato in aula, che esce dai soliti cliché in cui il reato è omicidio o stupro. Ben fatto, così come la colonna sonora di James Horner.

Abstract: SCN2A encodes Nav1.2, an excitatory neuron voltage-gated sodium channel and major monogenic cause of neurodevelopmental disorders, including developmental and epileptic encephalopathies (DEE) and autism. Clinical presentation and pharmocosensitivity vary with nature of SCN2A variant dysfunction with gain-of-function (GoF) cases presenting with pre- or peri-natal seizures and loss-of-function (LoF) patients typically having infantile spasms after 6 months of age. Here, we established and assessed patient induced pluripotent stem cell (iPSC) - derived neuronal models for two recurrent SCN2A DEE variants with GoF R1882Q and LoF R853Q associated with early- and late-onset DEE, respectively. Patient-derived iPSC lines were differentiated using a Neurogenin-2 overexpression yielding populations of cortical-like glutamatergic neurons. Electrophysiological and transcriptomic profiles were assessed after 2-4 weeks in culture. Increased neuronal activity at both cellular and network level was observed for R1882Q iPSC-derived neurons at three weeks of differentiation. In contrast, R853Q neurons showed only subtle changes in excitability after four weeks in vitro. In alignment with the reported efficacy in some GoF SCN2A patients, phenytoin (sodium channel blocker) reduced excitability of neurons to the control levels in R1882Q neuronal cultures. Transcriptomic alterations in neurons were detected for each variant and convergent pathways pointed at the shared mechanisms underlying SCN2A DEE. Copy rights belong to original authors. According to the documentary’s director, accomplished filmmaker Michael Apted (Coal Miner’s Daughter, Thunderheart, The World Is Not Enough), Robert Redford ruined “Incident at Oglala” out of fear of upsetting the powers that be. Although Redford was scarcely involved in the production of the documentary, he became heavily involved in its post-production, exercising his authority as executive producer to censor the final cut. When Mr. Apted objected to the omission of a vital section of commentary from two circuit court judges criticizing the handling of Leonard’s trial, he was overruled and banned from the film’s Sundance premiere.

For this Geek Challenge, Paul urges Arlo to watch Michael Apted's 1992 conspiracy thriller Thunderheart, starring Val Kilmer as an FBI agent who grows to embrace his Sioux heritage. In turn, Arlo makes Paul watch Jim Jarmusch's 1995 psychedelic Western Dead Man, wherein Johnny Depp's iteration of William Blake takes an offbeat journey to the next life. The boys address the major caveat of both films starring white men, as well as their own lily whiteness; determine that Graham Greene and Gary Farmer walk away with their respective movies; and discuss how both films explore spiritual death and rebirth.
