Welcome back to The Pursuit of Health Podcast. This week, we sit down with Stacy Cortechine, Rachel Moyer and Dr. Mitani from Japan to discuss the cultural differences between Japan and the US in approaching primary and secondary prevention of SCA/SCD in youth. As shared in our discussion, Stacy and Rachel both lost their sons to sudden cardiac arrest, and now dedicate their efforts to raising awareness, lobbying lawmakers and screening young people through their respective work for Who We Play For and Greg Moyer AED Fund.Their work is helping to save lives, and yet so many hurdles persist: slow legislation, reluctance to purchase AEDs, misinformation and denial about the necessity of measures - the list goes on.Dr. Mitani reveals the simplicity of Japan's approach - compulsory screening, AED programs, educating primary care physicians - and their numbers show that it is working. The blueprint exists. It's time for our healthcare system to adopt it in full. Follow me on Instagram and Facebook @ericfethkemd and checkout my website at www.EricFethkeMD.com. America celebrated Independence Day earlier this week, on the Fourth of July, so while the spotlight was on the US we thought it would be the perfect time to catch up with New York-based Adelaidean, Josh Pugh aka America Josh, to get a sense of the South Australian expat community in the Big Apple as well as reflect on some comparisons between life there and here. The SA Drink Of The Week is a gold medal wine, the Baroota Creek 2020 Cabernet Shiraz from Flinders Run. Watch this winery. I hear stories of people buying Penfolds Grange for quite low prices before it was famous, this has the potential to be one of those wines. And in the Musical Pilgrimage, Gerry Masi is back as part of Distractor Code. You can navigate episodes using chapter markers in your podcast app. Not a fan of one segment? Oh, and here's our index of all episode in one concise page Running Sheet: From Adelaide To The USA With Love And America Josh 00:00:00 Intro Introduction 00:03:53 SA Drink Of The Week We have two special wines this episode, the Flinders Run Baroota Creek 2020 Cabernet Shiraz and the 2021 Baroota Creek 2021. Flinders Run is a winery in the Southern Flinders area, run by Emanuel and Laura Skorpos. This wine has just been awarded a gold medal score of 95 by Wine Showcase Magazine. This special tasting was held at Enzos's Ristorante, with Emanuel and Laura (although Laura preferred to take photos and videos rather than be on microphone) and Damon Musha from Wine Australia USA. 00:17:01 Josh Pugh, America Josh In 2017, Josh Pugh left what he described as a "fantastic life" in Adelaide and headed to New York to start a new life there. He handed over control of his company, sold his house, sold his car, and even gave away his cat so his odyssey could begin. Luckily for him, things have fallen into place: He found a home, a wife, and a dog, all in a matter of a few short years. However, last month, another South Australian, Fairlie Delbridge, connected with Josh at the launch of the South Australia Club in NYC, and then connected Josh with The Adelaide Show. And, as fate would have it, we're having this chat with Josh while he still has South Australia's famous Mitani chicken salt in his system. America Josh How did you get Mitani chicken salt? Is there a list of items or places you miss? Food items, events like Fringe? Are there some things you are glad to have left behind? Tell us about the South Australia Club in NYC. The opening was attended by Stuart Nutting, who is Austrade's Regional Director South Australia Trade and Investment, along with Trade and Investment Minister, Nick Champion. If it is anything like the Adelaide Club, the only way you get in is if you're a pastoralist, an Anglican, or you share DNA with Alexander Downer? We have much to unpack but we need to start with your decision to leave Adelaide. If your life was truly "fantastic", why did you leave? You currently run open gatherings in Central Park. Why do you do that? Your website says you created America Josh to help people plan to move to or settle into New York. What is involved in getting a Green Card or residency? You mention you were attracted by the "buzz" of New York. Something we've long argued at The Adelaide Show is that you can have a miserable boring life here or in New York; most of it depends on how proactively you are engaging with life. Firstly, has the buzz faded like the initial burst of romance in a relationship? And, secondly, can you reflect on the extrinsic and intrinsic factors that have helped you feel settled? What was Fourth of July like? Fairlie Delbridge is a mover and shaker in SA but is spending a year studing at Harvard. How have your paths crossed? Cricket. Was watching the other day and remember you talking fondly of cricket. Commentators were talking about timers to keep games moving at pace and how baseball now has countdown times for pitchers. I must say, I watched some baseball. It is soooooo boring. Being there is okay but, just like golf, it's not made for tv. Your thoughts? Also, what do you think of the stumping in the Ashes? I get confused with cricket umpire Steve Davis and I notice there is an English comedian, Josh Pugh. Do you ever get mis-identified? Who will you vote for in the elections? The dangers of stalking guests is that I saw a picture of you as Mrs Doubtfire. Is that a regular habit? Final thoughts for people thinking of moving? And are you encouraging Americans to move here? 01:24:15 Musical Pilgrimage Our featured song this episode is This Town by Gerry Masi under the banner of Distractor Code; his collaboration with Paul White. It's all played on 80s synths with Gerry singing and, unlike the songs under his non de plume, Mad Dog Malcolm, there are no inappropriate lyrics!!! Given the US flavour if this episode, I should point out that This Town is not the classic Frank Sinatra song, nor is it Adelaide's version of New York New York, however, it puts Gerry's operatic power to good use and it broods with darkness and passion. For Josh, he'll note The Arts Theatre in the video cover image, below.Support the show: https://theadelaideshow.com.au/listen-or-download-the-podcast/adelaide-in-crowd/See omnystudio.com/listener for privacy information.

‘When I arrived at Ngogo, it didn't take me long to figure out this place was unusual.' For the second installment of our Making of Chimp Empire Series, we were privileged to have the esteemed Professor John Mitani as a guest on Talking Apes. John shared intimate stories about his extraordinary lifetime of work studying the Ngogo chimpanzees and his latest adventure serving as a scientific adviser to the team of filmmakers working on the  hit Netflix docuseries. ‘The Ngogo chimps have been filmed many times by many people - but nothing like this.'During our chat, John spoke passionately about his deep connection with the Ngogo chimps owed to his decades-spanning research efforts. His extensive knowledge and expertise in the field of primatology added a profound layer of understanding to the documentary series. As the scientific adviser to the filmmakers, he played a pivotal role in ensuring accuracy and authenticity in the portrayal of chimpanzee behavior and social dynamics. One remarkable aspect of Professor Mitani's relationship with the director, James Reed, was their blossoming friendship. Through their shared passion for chimpanzees, a bond was formed that transcended the boundaries of their respective disciplines. James recognized the importance of including the scientific community in the filmmaking process and, in doing so, granted Professor Mitani unprecedented levels of creative access. This collaboration between a scientist and a filmmaker brought a unique perspective to Chimp Empire, combining scientific rigor with compelling storytelling. ‘The chimps accept our presence. I have a hard time wrapping my head around why, but I'm grateful for it.' By granting Professor Mitani a significant role in the making of Chimp Empire, James elevated the documentary series beyond a mere visual spectacle. The integration of scientific expertise added depth and credibility to the project, ensuring that the behaviors and social dynamics of the Ngogo chimpanzees were accurately portrayed. John told us that this level of collaboration between scientists and filmmakers at Ngogo is not commonplace, making Chimp Empire a truly exceptional exploration of chimp life. John will be joining us again on Season 3 of Talking Apes Podcast to talk about his astounding career studying chimpanzee society with the Ngogo Chimpanzee Project. Coming Autumn 2023. Click HERE to read more about how the chimps are doing NOW, which was put together with John's help. Click HERE to watch a short behind-the-scenes video of Chimp Empire featuring John. This podcast contains spoilers and we recommend that listeners watch the series on Netflix before listening. Netflix theme, and ChimpSupport the showTalking Apes is an initiative of the nonprofit GLOBIO.

Anti-nucleolin aptamer, iSN04, inhibits the inflammatory responses in myoblasts by modulating the β-catenin/NF-κB signaling pathway

Link to bioRxiv paper: http://biorxiv.org/cgi/content/short/2023.04.01.535227v1?rss=1 Authors: Yamamoto, M., Miyoshi, M., Morioka, K., Mitani, T., Takaya, T. Abstract: A myogenetic oligodeoxynucleotide, iSN04, is the 18-base single-stranded DNA that acts as an anti-nucleolin aptamer. iSN04 has been reported to restore myogenic differentiation by suppressing inflammatory responses in myoblasts isolated from patients with diabetes or healthy myoblasts exposed to cancer-releasing factors. Thus, iSN04 is expected to be a nucleic acid drug for the muscle wasting associated with chronic diseases. The present study investigated the anti-inflammatory mechanism of iSN04 in the murine myoblast cell line C2C12. Tumor necrosis factor- (TNF-) or Toll-like receptor (TLR) ligands (Pam3CSK4 and FSL-1) induced nuclear translocation and transcriptional activity of nuclear factor-{kappa}B (NF-{kappa}B), resulting in upregulated expression of TNF- and interleukin-6. Pre-treatment with iSN04 significantly suppressed these inflammatory responses by inhibiting the nuclear accumulation of {beta}-catenin induced by TNF- or TLR ligands. These results demonstrate that antagonizing nucleolin with iSN04 downregulates the inflammatory effect mediated by the {beta}-catenin/NF-{kappa}B signaling pathway in myoblasts. In addition, the anti-inflammatory effects of iSN04 were also observed in smooth muscle cells and pre-adipocytes, suggesting that iSN04 may be useful in preventing inflammation induced by metabolic disorders.
