BUFFALO, NY- August 15, 2024 – A new #research paper was #published in Oncotarget's Volume 15 on August 14, 2024, entitled, “A nanobody against the V-ATPase c subunit inhibits metastasis of 4T1-12B breast tumor cells to lung in mice.” The vacuolar H+-ATPase (V-ATPase) is an ATP-dependent proton pump that functions to control the pH of intracellular compartments as well as to transport protons across the plasma membrane of various cell types, including cancer cells. Researchers Zhen Li, Mohammed A. Alshagawi, Rebecca A. Oot, Mariam K. Alamoudi, Kevin Su, Wenhui Li, Michael P. Collins, Stephan Wilkens, and Michael Forgac from Tufts University School of Medicine; Tufts University; Dana Farber Cancer Institute, Harvard Medical School; University of Minnesota School of Medicine; Prince Sattam Bin Abdulaziz University; Korro Bio; SUNY Upstate Medical University; and Foghorn Therapeutics, have previously shown that selective inhibition of plasma membrane V-ATPases in breast tumor cells inhibits the invasion of these cells in vitro. BUFFALO, NY- March 19, 2024 – A new #research paper was #published in Aging (listed by MEDLINE/PubMed as "Aging (Albany NY)" and "Aging-US" by Web of Science) Volume 16, Issue 5, entitled, “PR55α-controlled protein phosphatase 2A inhibits p16 expression and blocks cellular senescence induction by γ-irradiation.” Cellular senescence is a permanent cell cycle arrest that can be triggered by both internal and external genotoxic stressors, such as telomere dysfunction and DNA damage. The execution of senescence is mainly by two pathways, p16/RB and p53/p21, which lead to CDK4/6 inhibition and RB activation to block cell cycle progression. While the regulation of p53/p21 signaling in response to DNA damage and other insults is well-defined, the regulation of the p16/RB pathway in response to various stressors remains poorly understood. In this new study, researchers Chitra Palanivel, Lepakshe S. V. Madduri, Ashley L. Hein, Christopher B. Jenkins, Brendan T. Graff, Alison L. Camero, Sumin Zhou, Charles A. Enke, Michel M. Ouellette, and Ying Yan from the University of Nebraska Medical Center report a novel function of PR55α, a regulatory subunit of PP2A Ser/Thr phosphatase, as a potent inhibitor of p16 expression and senescence induction by ionizing radiation (IR), such as γ-rays. “During natural aging, there is a gradual accumulation of p16-expressing senescent cells in tissues [76]. To investigate the significance of PR55α in this up-regulation of p16, we compared levels of the p16 and PR55α proteins in a panel of normal tissue specimens derived from young (≤43 y/o) and old (≥68 y/o) donors.” The results show that ectopic PR55α expression in normal pancreatic cells inhibits p16 transcription, increases RB phosphorylation, and blocks IR-induced senescence. Conversely, PR55α-knockdown by shRNA in pancreatic cancer cells elevates p16 transcription, reduces RB phosphorylation, and triggers senescence induction after IR. Furthermore, this PR55α function in the regulation of p16 and senescence is p53-independent because it was unaffected by the mutational status of p53. Moreover, PR55α only affects p16 expression but not p14 (ARF) expression, which is also transcribed from the same CDKN2A locus but from an alternative promoter. In normal human tissues, levels of p16 and PR55α proteins were inversely correlated and mutually exclusive. “Collectively, these results describe a novel function of PR55α/PP2A in blocking p16/RB signaling and IR-induced cellular senescence.” DOI - https://doi.org/10.18632/aging.205619 Corresponding authors - Michel M. BUFFALO, NY- December 27, 2023 – A new #research paper was #published in Aging (listed by MEDLINE/PubMed as "Aging (Albany NY)" and "Aging-US" by Web of Science) Volume 15, Issue 23, entitled, “Angelica gigas extract inhibits acetylation of eNOS via IRE1α sulfonation/RIDD-SIRT1-mediated posttranslational modification in vascular dysfunction.” Angelica gigas NAKAI (AG) is a popular traditional medicinal herb widely used to treat dyslipidemia owing to its antioxidant activity. Vascular disease is intimately linked to obesity-induced metabolic syndrome, and AG extract (AGE) shows beneficial effects on obesity-associated vascular dysfunction. However, the effectiveness of AGE against obesity and its underlying mechanisms have not yet been extensively investigated. In this new study, researchers Geum-Hwa Lee, Hwa-Young Lee, Young-Je Lim, Ji-Hyun Kim, Su-Jin Jung, Eun-Soo Jung, Soo-Wan Chae, Juwon Lee, Junghyun Lim, Mohammad Mamun Ur Rashid, Kyung Hyun Min, and Han-Jung Chae from Jeonbuk National University and Jeonbuk National University Hospital supplemented 40 high fat diet (HFD) rats with 100–300 mg/kg/day of AGE to determine its efficacy in regulating vascular dysfunction. “[...] the primary aim of this study is to examine the inhibitory effects of AGE on dyslipidemia-associated vascular dysfunction, with a focus on its potential mechanisms of action.” The vascular relaxation responses to acetylcholine were impaired in HFD rats, while the administration of AGE restored the diminished relaxation pattern. Endothelial dysfunction, including increased plaque area, accumulated reactive oxygen species, and decreased nitric oxide (NO) and endothelial nitric oxide synthase (eNOS) Ser1177 phosphorylation, were observed in HFD rats, whereas AGE reversed endothelial dysfunction and its associated biochemical signaling. Furthermore, AGE regulated endoplasmic reticulum (ER) stress and IRE1α sulfonation and its subsequent sirt1 RNA decay through controlling regulated IRE1α-dependent decay (RIDD) signaling, ultimately promoting NO bioavailability via the SIRT1-eNOS axis in aorta and endothelial cells. Independently, AGE enhanced AMPK phosphorylation, additionally stimulating SIRT1 and eNOS deacetylation and its associated NO bioavailability. Decursin, a prominent constituent of AGE, exhibited a similar effect in alleviating endothelial dysfunctions. The king umbrella is Effective Ventures Foundation, which hosts CEA, 80k, Longview, EA Funds, Giving What We Can, Asterisk magazine, Centre for Governance of AI, Forethought Foundation, Non-Trivial, and BlueDot Impact. Posts on the castle also describe it as an EVF project, although it's not listed on the website. Rethink Priorities has a program specifically to provide sponsorship to groups that need it. LessWrong/Lightcone is hosted by CFAR, and have sponsored at least one project themselves (source: me. It was my project). Fiscal sponsorship has a number of advantages. It gets you the privileges of being a registered non-profit (501c3 in the US) without the time-consuming and expensive paperwork. That's a big deal if the project is small, time-limited (like mine was) or is an experiment you might abandon if you don't see results in four months. Even for large projects/~orgs, sharing a formal legal structure makes it easier to share resources like HR departments and accountants. In the short term, forming a legally independent organization seems like a lot of money and effort for the privilege of doing more paperwork. The downsides of fiscal sponsorship …are numerous, and grow as the projects involved do. The public is rightly suspicious about projects that share a legal entity claiming to be independent, so bad PR for one risks splash damage for all. The government is very confident in its belief that you are the same legal entity, so legal risks are shared almost equally (iamnotalawyer). So sharing a legal structure automatically shares risk. That may be fixable, but the fix comes at its own cost. The easiest thing to do is just take fewer risks. Don't buy retreat centers that could be described as lavish. And absolutely, 100%, don't voluntarily share any information about your interactions with FTX, especially if the benefits to doing so are intangible. So some amount of value is lost because the risk was worth it for an individual or small org, but not to the collective. [it is killing me that I couldn't follow the rule of three with that list, but it turns out there aren't that many legible, publicly visible examples of decisions to not share information] And then there are the coordination costs. Even if everyone in the legal org is okay with a particular risk, you now have an obligation to check with them.The answer is often "it's complicated", which leads to negotiations eating a lot of attention over things no one cares that much about. Even if there is some action everyone is comfortable with, you may not find it because it's too much work to negotiate between that many people (if you know anyone who lived in a group house during covid: remember how fun it was to negotiate safety rules between 6 people with different value functions and risk tolerances?). A new research paper was published in Oncotarget's Volume 14 on February 2, 2023, entitled, “Everolimus downregulates STAT3/HIF-1α/VEGF pathway to inhibit angiogenesis and lymphangiogenesis in TP53 mutant head and neck squamous cell carcinoma (HNSCC).” TP53 mutant head and neck squamous cell carcinoma (HNSCC) patients exhibit poor clinical outcomes with 50–60% recurrence rates in advanced stage patients. In a recent phase II clinical trial, adjuvant therapy with everolimus (mTOR inhibitor) significantly increased 2-year progression-free survival in p53 mutated patients. TP53-driven mTOR activation in solid malignancies causes upregulation of HIF-1α and its target, downstream effector VEGF, by activating STAT3 cell signaling pathway. In this recent study, researchers Md Maksudul Alam, Janmaris Marin Fermin, Mark Knackstedt, Mackenzie J. Noonan, Taylor Powell, Landon Goodreau, Emily K. Daniel, Xiaohua Rong, Tara Moore-Medlin, Alok R. Khandelwal, and Cherie-Ann O. Nathan from LSU-Health Sciences Center investigated the effects of everolimus on the STAT3/HIF-1α/VEGF pathway in TP53 mutant cell lines and xenograft models. “The role of mTOR inhibitors (mTORi) as potent growth inhibitory and antiangiogenic/anti-lymphangiogenic agents in HNSCC is well established [18]. Moreover, mTORi significantly suppressed baseline invasiveness of endothelial and HNSCC tumor cells [19]. However, the underlying molecular mechanisms for mutant p53 protein-mediated activation of the mTOR pathway which drive the oncologic processes in HNSCC are yet to be elucidated.” Treatment with everolimus significantly inhibited cell growth in vitro and effectively reduced the growth of TP53 mutant xenografts in a minimal residual disease (MRD) model in nude mice. Everolimus treatment was associated with significant downregulation of STAT3/HIF-1α/VEGF pathway in both models. Further, treatment with everolimus was associated with attenuation in tumor angiogenesis and lymphangiogenesis as indicated by decreased microvessel density of vascular and lymphatic vessels in HN31 and FaDu xenografts. Everolimus downregulated the STAT3/HIF-1α/VEGF pathway to inhibit growth and in vitro tube formation of HMEC-1 (endothelial) and HMEC-1A (lymphatic endothelial) cell lines. “Our studies demonstrated that everolimus inhibits the growth of TP53 mutant tumors by inhibiting angiogenesis and lymphangiogenesis through the downregulation of STAT3/HIF-1α/VEGF signaling.” DOI: https://doi.org/10.18632/oncotarget.28355 Correspondence to: Cherie-Ann O. Check out today's questions:    Sierra: Hello Dr Cabral, And thank you so much for everything. You have been such an amazing mentor to my husband and I and we are loving the IHP course, can't wait for level 2! I had my first child two years ago (2019) with no complications during pregnancy. The only “complication” that I can say is that for a few hours after giving birth I couldn't urinate on my own, which led to a catheter insertion to empty my bladder. I'm not sure if this can be related but I've been having this excruciating pain every month for two years, about 14days or so after my period (which made me believe it could be Mittelschmerz) that basically Inhibits me from living a normal life for 2-3 days. I can't walk and if I do I'm hunched over holding my left lower abdomen (where my ovary is) due to a constant dull inflamed pain. Any how, I have been to the ER countless amount of times, have had blood work, seen multiple specialist, and I have been left with the same depressing answers “everything's normal” or “you're probably stressed because you are a new mom”… It's been two years and the only options they are giving me is birth control (which I tried for 6 months and it didn't help) or exploratory surgery. What would you recommend or what could be the reasons to why I'm having this on going pain? Thanks again   Alicja (Ah-leets-yah): Hi Dr.Cabral, Thank you for all your amazing work! I hope you can help me with my gut question: I've been dealing with gut issues for a while. I took probiotics daily and it wasn't until I found your podcast and learned that with people like me, probiotics may make things worse. I stopped taking probiotics and I noticed less bloating, however my mood dropped down significantly. I understand the gut-brain connection and I know I need to heal my gut to feel mentally happy too. Taking probiotics daily helped me stay happy and positive, but I noticed big shift in mood when I don't take them. I feel depressed. I'm currently on Para Support protocol (next will be CBO) Any advice for me during my protocols. Should I suck it up for a bit and still avoid probiotics for a while. Or should I start taking probiotics again and along with Para Support? Maybe just s.boulardii? Any advice would be appreciated. Thank you again for all you do.   Darren: Good day Dr Cabral. I don't know if a trigger warning is needed prior to this question but is semen retention something that's actually beneficial for men? That is going days, weeks, even months with intentionally minimising ejaculating usually done by either abstention, stopping (intercourse or self pleasure) before the point of ejaculation, or learning how to climax without ejaculating. While I'm seeing many anecdotal benefits I understand some in ayurveda say that's Prajnyaparadha (unhealthy suppression). I know you usually have a more balanced approach. Will appreciate your take on this.   Malissa: Day two very Nauseous. Took the day off, Just did a dry brush, Epsom bath and drank water mixed with sea salt and lemons (Supposed to be a natural Gatorade). I did sweat so that's good. Unfortunately. Suggestions for Nausea?   Francesca: Hi Dr. Cabral. I am a type 1 diabetic, female and 24 y/o. When I was 20 y/o, I started restrictive eating and my weight dropped significantly over the course of a year, as did my estrogen levels (below the charts). At that point, I was put on HRT and given doses of estradiol, and progesterone. I also began ED recovery. 3 years later, I have put on 50 pounds while still needing to eat very restrictive, my metabolism is very low, and most concerningly, I am experiencing blood sugars above 500 every day (very low carb diet!). My question is, What EXACTLY breaks a fast? I believe the HRT/ ED recovery has caused insulin resistance, and causes me to take insulin even when in a fasted state. is the insulin itself breaking a fast? There are a lot of studies that interchange the action of eating w/ the action of insulin secretion, but because I am a diabetic, one does not automatically follow the other. I just began taking Triphala, and noticed it has lowered blood glucose. But again, is this a fix to lowering insulin needs, or is this essentially processing the same as insulin, and furthering fat deposits? Your past podcasts on fasting suggests a lot of things that cause a glucose spike will break a fast, but does not truly define if its the insulin causing the fast break. UN Special Rapporteur Alena Douhan Report on Human Rights on the Impact on Unilateral Coercive Measures on Venezuela following her 2/1- 2/12/21 visit is reviewed. It called for an immediate end to the US and Allied sanctions on Venezuela. The report found that sanctions have resulted in economic and human calamities for the entire population especially the poor and most vulnerable. While billions in Venezuelan assets that could be used for medicine and vaccines remain frozen at foreign banks the report claimed some 2.5 million Venezuelans now face severe food security. With the hardening of the sanctions over the years since 2005, state revenues to Caracas have been slashed by a staggering 99% leaving the country to live on just 1% of its presanction income.
