We are headed west on day two of our 12 Nightmares Before Christmas. It was pitch black on a gorgeous night in the middle of the Arizona desert in October 2003. There was not a cloud in the sky, but billions of stars were observable by the naked eye because Bumble Bee Road is in the middle of nowhere. In every direction, there was nothing but 90,000 acres of rolling hills dotted with Saguaro cactus spikes. Brandon Rumbaugh, age 20, a personal fitness instructor, and his girlfriend, Lisa Marie Gurrieri, a nineteen-year-old secretary for the Salt River Project, were celebrating their one-year anniversary. They had initially wanted to go to Disneyland to commemorate the occasion, but it was too expensive. The couple had been discussing marriage, so they didn't want to spend much money.  So, instead, they thought it would be romantic to go back to the Arizona desert and do some stargazing as they did on their first date. This time, though, they decided to camp out under the stars. Lisa had never been camping before, and the couple didn't really have camping equipment either, so they asked Lisa's mother, Paula, if they could borrow her brand-new white F-150 truck, and they would just sleep in the back of the vehicle with some sleeping bags. Paula wasn't thrilled about them taking the truck since it needed an oil change, but in the end, she gave them permission. Lisa also borrowed her mother's video camera. Then, Lisa made another request; she asked her mother not to tell her Uncle Mike where they were going because she knew that he'd be upset and worried about them. The area had become known as a “party destination. Republicans in Georgia, during a recent segment broadcasted by MSNBC, enthusiastically voiced their steeled support for former president Donald Trump in the face of numerous ongoing legal confrontations. An intriguing poll displayed a convincing lead for Trump over Biden in Georgia statewide by 9%, despite his tussles with the local and out-of-state law enforcement agencies. This heightened resolve towards Trump is a direct response to what appears to be, from the perspective of these voters, an undeserved witch hunt against the former president. Antonio Jones, an unremitting Trump supporter, during an interview with NBC News correspondent Blayne Alexander, refuted the possibility of his support waning due to Trump's trials. Jones expressed his skepticism at the multiple trials taking place in different locations, fueling his and others' conviction that this was far from a coincidence. This trial frenzy stoked questions of why such tribulations surfaced at this particular moment in time, bolstering support for Trump rather than diminishing it. It's worthwhile to note that Trump carried Georgia in 2016 with a slim margin, however, fell short to President Joe Biden in the 2020 election. The sentiment among the voters still echoes a strong belief in Trump and a zealous anticipation for his potential running in future elections. First-time voter Thaden Rumbaugh fervently articulated his admiration for the combative spirit of Trump. According to Rumbaugh, facing harsh criticism from numerous news outlets has not diminished Trump's resolve. Abstract: Gene expression intersects with neural activity to produce cortical circuits during brain development. However, the cell biological mechanisms linking gene expression to activity-dependent cortical circuit assembly remain unclear. Here, we demonstrate in mice that a newly discovered function of the neurodevelopmental disorder gene, Syngap1, is to cell-autonomously control intrinsic membrane excitability (IME) in developing cortical glutamatergic neurons. Syngap1 regulation of IME was mechanistically linked to wiring of a cortical circuit motif required for sensory processing and behavioral action. Restoring depressed IME in Syngap1 deficient neurons through genetic targeting of hyper-functional potassium currents unleashed deficient dendritic morphogenesis in upper lamina sensory cortex pyramidal neurons. Furthermore, enhancing dendritic morphogenesis was sufficient to stimulate assembly of translaminar feed-forward excitatory circuit motifs. Thus, Syngap1 promotes excitatory circuit assembly during cortical development by maintaining IME in a range that enables trophic neuronal activity to maximize pyramidal cell somatodendritic maturation and subsequent synapse formation. But I also noticed that you would either play in a sport or activity six days a week or you weren't doing anything. You will maybe home on electronics. And so I said, something's happening. So I started researching it, and I discovered that since the 1980s, children have lost eight to 10 hours of free play time a week. And because of that, I said, I need to do something about it.”Why balance is important.  “I hope that people don't jump back into signing up for an adult-led activity every day because frankly, I don't think you need it. I think you need time to be able to just totally relax and have your choice of play, and that's a very wide range. That could be reading a book that you enjoy or a comic book, or it could be writing in your journal. At the same time, you and I both really understand and get what it does for us when we physically move, and that's why coming from a physical educators background, I say to people, “You need to think about your body, you're gonna feel so much better if you regularly get physical activity.” I don't like to call it exercise or going to the gym, I like to call it an activity that you enjoy.”Why team play is a great option.“Being on a team is so wonderful because you can help your teammates in so many ways. One of the biggest ways you can help them is by giving positive feedback and really just being supportive. Abstract: SYNGAP1 is a major genetic risk factor for global developmental delay, autism spectrum disorder, and epileptic encephalopathy. De novo loss-of-function variants in this gene cause a neurodevelopmental disorder defined by cognitive impairment, social-communication disorder, and early-onset seizures. Cell biological studies in mouse and rat neurons have shown that Syngap1 regulates developing excitatory synapse structure and function, with loss-of-function variants driving formation of larger dendritic spines and stronger glutamatergic transmission. However, studies to date have been limited to mouse and rat neurons. Therefore, it remains unknown how SYNGAP1 loss-of-function impacts the development and function of human neurons. To address this, we employed CRISPR/Cas9 technology to ablate SYNGAP1 protein expression in neurons derived from a human induced pluripotent stem cell line (hiPSC). Reducing SynGAP protein expression in developing hiPSC-derived neurons enhanced dendritic morphogenesis, leading to larger neurons compared to those derived from isogenic controls. Consistent with larger dendritic fields, we also observed a greater number of morphologically defined excitatory synapses in cultures containing these neurons. Moreover, neurons with reduced SynGAP protein had stronger excitatory synapses and expressed synaptic activity earlier in development. Finally, distributed network spiking activity appeared earlier, was substantially elevated, and exhibited greater bursting behavior in SYNGAP1 null neurons. We conclude that SYNGAP1 regulates the postmitotic maturation of human neurons made from hiPSCs, which influences how activity develops within nascent neural networks. Xander is wise beyond his years. At just 24 years old, Xander has an incredible perspective on life. He was born and raised in West Virginia and joined the Army shortly after graduating high school. While serving as a Military Police soldier, he suffered a devastating knee injury and was subsequently forced to transition out of the military due to a medical discharge. I'm tired of the stale, depressing exercise and yoga advice for the "older set." It too often characterizes people over 50 as weak, unfit, and fragile. Desiree Rumbaugh and Michelle Marchildon confirm that I am right! (And who doesn't like to be right?) Upon completion of boot camp in February 2008, he went on to the Infantry Training Battalion, after which he was assigned to the 1st Battalion 8th Marines as an 81mm mortar gunner. In March 2009, he was deployed to Iraq for a seven- month tour. Shortly after his return home from his first tour in September 2009, Corporal Rumbaugh was deployed to Afghanistan, where he was squad leader. Having already suffered multiple injuries from IED explosions during this second tour, Corporal Rumbaugh's life dramatically changed one day during a routine security patrol. As he attempted to aid a fellow Marine who was wounded from stepping on an IED, Corporal Rumbaugh triggered a secondary IED within just 20 feet of his comrade, resulting in the loss of both of his legs. After spending nearly two years undergoing extensive medical therapy at Walter Reed Army Medical Center in Washington D.C., he returned to his hometown of Uniontown, PA. In September 2012, at the age of 24, Corporal Rumbaugh retired medically from military service, after five years of active duty. Having dedicated his life to his country, Corporal Rumbaugh is now committed to serving his community through charitable works and inspiring others with his motivational story of perseverance, loyalty, and faith.
