Abstract: One of the defining pathological features of Alzheimer's Disease (AD) is the deposition of neurofibrillary tangles (NFTs) composed of hyperphosphorylated tau in the brain. Aberrant activation of kinases in AD has been suggested to enhance phosphorylation and toxicity of tau, making the responsible tau-directed kinases attractive therapeutic targets. The full complement of tau interacting kinases in AD brain and their activity in disease remains incompletely defined. Here, immunoaffinity enrichment coupled with mass spectrometry (MS) identified TANK-binding kinase 1 (TBK1) as a tau-interacting partner in human AD cortical brain tissues. We validated this interaction in both human AD and familial frontotemporal dementia and parkinsonism linked to chromosome 17 (FTDP-17) caused by mutations in MAPT (R406W) postmortem brain tissues as well as human cell lines. Further, we document increased TBK1 activity in both AD and FTDP-17 and map the predominant TBK1 phosphorylation sites on tau based on in vitro kinase assays coupled to MS. Lastly, in a Drosophila tauopathy model, activating expression of a conserved TBK1 ortholog triggers tau hyperphosphorylation and enhanced neurodegeneration, whereas knockdown had the reciprocal effect, suppressing tau toxicity. Neuere Studien zeigen, dass Fehlregulationen alternativer Spleißprozesse erheblichen Einfluss auf die Entstehung von Krankheiten besitzen können. Exon 10 des Tau-Gens wird alternativ gespleißt und je nachdem, ob das Exon ein- oder ausgeschlossen wird, werden Isoformen mit drei (Tau -Exon 10) bzw. vier (Tau +Exon 10) Mikrotubuli-Bindungsmotiven generiert. Bei der Frontotemporalen Demenz mit Parkinsonismus gekoppelt an Chromosom 17 (FTDP-17) führen bestimmte Spleißmutationen zur Retention von Exon 10. Wie die FTDP-17 gehört auch die Alzheimer-Erkrankung zu den sog. Tauopathien, einer Gruppe von Krankheiten, die durch übermäßige Ablagerungen des Tau-Proteins im Gehirn charakterisiert sind. Im Rahmen dieser Arbeit wurde an humanem post mortem Gewebe untersucht, ob der Alzheimer-Erkrankung ein Fehler im Regulationsmechanismus des alternativen Spleißens zugrunde liegt. Mittels RT-PCR und Western Blot wurde das Spleißmuster von Tau Exon 10, dem humanen Transformer2-beta (Htra2-ß) und der CDC-ähnlichen Kinase2 (Clk2) untersucht. Ebenso wie Tau Exon 10 werden auch Htra2-ß und Clk2 alternativ gespleißt, wodurch Isoformen mit unterschiedlichen Funktionen entstehen können. Auf mRNA-Ebene konnte gezeigt werden, dass es im präfrontalen und temporalen Kortex der Alzheimer-Gehirne im Vergleich zu Kontrollhirnen zu einer signifikant stärkeren Expression der Isoform tau +Exon10 kommt. Ein möglicher Mechanismus könnte die Regulation des in Zellkulturexperimenten beobachteten Einflusses von hTRA2-ß auf das alternative Spleißen von Tau Exon 10 sein. Die im Temporalkortex der Alzheimer-Patienten verstärkt exprimierte Isoform htra2-ß1 unterstützt diese Befunde. Ebenfalls aus Zellkulturexperimenten ist bekannt, dass CLK2 sowohl das alternative Spleißen von Htra2-ß wie auch wahrscheinlich indirekt das von Tau Exon 10 beeinflusst. Untersuchungen zur RNA-Expression von clk2 ergaben, dass die phosphorylierungsaktive Isoform clk2 +Exon 4 in allen untersuchten Regionen bei Alzheimer-Patienten im Vergleich zu Kontrollen signifikant vermindert exprimiert wird. Dies deutet auf ein Zusammenspiel zwischen Clk2, Htra2-ß und dem alternativen Spleißen von Tau Exon 10 hin, sodass man sowohl von Htra2-ß wie auch Clk2 einen Einfluss auf die Regulation des alternativen Spleißens von Tau Exon 10 annehmen könnte. Basierend auf Forschungsergebnissen zum Einfluss von Phosphatase-Inhibitoren auf alternatives Spleißen in der spinalen Muskelatrophie (SMA), ist es denkbar, mittels spezifischer Phosphatase-Inhibitoren Einfluss auf die Fehlregulation des alternativen Spleißens von Tau Exon 10 zu nehmen. So könnten beispielsweise Phosphatase-Inhibitoren gegen htra2-ß1 eine verstärkte Phosphorylierung dieses Proteins bewirken und somit den Ausschluss von Tau Exon 10 verstärken. Dieser Mechanismus könnte für die Etablierung eines neuen Therapieansatzes herangezogen werden.

Tau ist ein Mikrotubuli- assoziiertes Protein, dessen Expression im Nervensystem des Menschen der Regulation durch alternatives Spleißen unterliegt. Das Exon 10 dieses Gens, welches für einen Teil der Mikrotutuli- bindenden Domäne kodiert, ist ein für Erwachsene spezifisches Kassettenexon. Mutationen, die den Einschluss von Exon 10 verstärken, resultieren in der Produktion von Tau- Protein, das vier Mikrotubuli- bindende Aminosäuresequenzwiederholungen enthält. Diese Mutationen scheinen in ursächlichem Zusammenhang mit der Frontotemporalen Demenz mit Parkinsonismus gekoppelt an Chromosom 17 (FTDP- 17) zu stehen. In dieser Arbeit konnte mittels Transfektionsexperimenten gezeigt werden, dass die Verwendung von Exon 10 durch ein komplexes Zusammenspiel der CDC- ähnlichen Kinase Clk2, einer SR- Proteinkinase, und des SR- ähnlichen Proteins humaner Transformer 2- b (Htra2- b) reguliert wird. Kotransfektionsexperimente legen den Schluss nahe, dass diese Regulation über mehrere ineinandergreifende Prozesse abläuft. Die Kinase Clk2 scheint dabei sowohl direkt durch Posphorylierung bzw. Hyperphosophorylierung von Htra2- b als auch indirekt durch Einfluss auf die alternative Expression der Htra2- b Isoformen in den Regulationsmechanismus einzugreifen. Phosphorylierung von SR- Proteinen führt zu deren Freisetzung aus den nukleären Speicherkomponenten, den speckles, und damit zur Aktivierung der Spleißreaktion, während sowohl eine Hyper- als auch eine Hypophosphorylierung in der Regel einen hemmenden Einfluss auf Spleißen ausüben. Kontrollierte Phosphorylierung scheint demnach zu einer regulierbaren Veränderung von prä- mRNA- Prozessierungswegen zu führen. Eine Interpretation dieser Resultate könnte als Grundlage für die Entwicklung neuer therapeutischer Konzepte verwendet werden.
