The Mechanics of Genetic Drift Plants are natural genetic mosaics. As they grow and divide over time, typos naturally accumulate in their DNA code, which are known as somatic mutations. Traditional vegetative cuttings and field-grown plants accumulate these mutations at a very low, linear baseline rate over decades. However, the extreme genomic degradation highlighted in the paper is driven by rapid generational cell cycling in vitro. Forcing an unorganized mass of cells to constantly divide and regenerate bypasses the plant's normal cellular quality control checkpoints. Somatic Embryos vs. Meristem Cultures Somatic embryogenesis involves stripping a plant down to a single cell or callus phase to regenerate an embryo. This is considered high risk for mutations because if the founder cell contains a mutation, that mutation becomes permanently fixed in every single cell of the thousands of plants propagated from it. This differs from meristem tip culture, which is the low risk, predominant form of tissue culture used in cannabis for clearing Hop Latent Viroid. Because meristem tip culture preserves the existing, highly organized architecture of the shoot apical meristem rather than forcing single cells to reprogram, the genetic risk is fundamentally lower. DNA Typos vs. Chromosome Catastrophes The high-stress cell cycling environment caused macroscopic genomic errors, such as whole-chromosome duplications called trisomies. In the walnut clones, these large-scale errors made it incredibly difficult to grow the tissue back out into healthy adult plants. The stress of rapid lab cell cycling also desilenced transposable elements, or jumping genes, allowing pieces of DNA to insert themselves randomly throughout the genome and disrupt normal gene expression. It is worth noting that not all mutations are bad. Small point mutations, or single base changes, happen constantly in nature as life being life. Most occur in non-coding regions and have zero impact, while a rare few can actually create desirable new traits as seen historically in citrus and wine grapes. Genetics vs. Epigenetics The guests distinguish between true genetic mutations, which are permanent changes to the A, C, T, G code, and epigenetic modifications, which act like chemical bookmarks such as methylation that change how a gene is read without altering the underlying sequence. Some instances of a mother plant losing vigor may actually be epigenetic shifts caused by environmental stress or endophyte and pathogen buildup over time, rather than a permanent genetic mutation. Actionable Advice for Cannabis Nurseries Commercial operations scaling up clones to the tens of thousands should utilize genetic sequencing early in the pipeline. Testing the early founder material can easily catch large-scale catastrophic mutations like chromosome duplications or deletions, protecting the nursery from mass-producing defective production plants. Whole-genome sequencing costs have plummeted significantly, and the guests encourage cultivators and nursery operators with unique, degenerating, or shifting clonal lines to collaborate with academic labs to sequence the data and map exactly what is changing. Guest Information and Contact Details J. Grey Monroe is an Associate Professor in the Department of Plant Sciences and the Genome Center at UC Davis. Before we dive into the grim reality of the Russian collapse, a quick note on the channel art. I've been listening to your feedback regarding our AI thumbnails. The community poll was an absolute dead heat at 52% vs 48%, so we had to find a middle ground. We are shifting the visual baseline to a highly stylized, character-free 2D Anime aesthetic. It removes the uncanny valley, avoids the public figure issues, and gives us a distinct, unified look. It might not be a perfect solution for everyone, but it's a solid half-step compromise to keep the show looking sharp while we focus on the actual journalism. Let me know what you think in the comments. Now, onto the nightmare. In Episode 2.34, we document a state legally canceling its own existence. While the Western media panics over fake Excel spreadsheets, the Russian reality is terminal. We break down the 10% macro-drop in petroleum products and the glorious return of Soviet-style 20-liter gas ration cards in occupied Crimea. The Ukrainian Armed Forces aren't just holding the line; they have turned the R280 logistics highway into a mechanical graveyard using advanced drone swarms. But the real fatal damage is happening in the rear. We cover Finance Minister Siluanov's desperate 16-trillion-ruble sequester of the civilian economy, the mafia-style raid on Wildberries, and the absolute absurdity of Putin's own regional proxy having a daughter who is actively funding the Ukrainian military from Georgia. And the ultimate punchline? Russian oligarchs are now buying heavy anti-aircraft weapons to build their own PMCs. They aren't preparing for NATO; they are arming up for the post-Putin Civil War.The Empire of Clones is burning. What if you're not burned out or unproductive—what if you're simply working against your natural instincts? In this episode of the Leaders of Transformation, Nicole Jansen sits down with Amy Bruske, President and Integrator of Kolbe Corp, to explore the powerful principles behind her new bestselling book, Do More, More Naturally, co-authored with David Kolbe. Amy explains why success isn't about forcing yourself to improve your weaknesses or manage your time more efficiently. Instead, it's about understanding how you're naturally wired to take action, solve problems, and make decisions. Together, Nicole and Amy discuss the often-overlooked conative part of the mind, the role of energy management, and how leaders can create thriving teams by leveraging natural strengths rather than fighting against them. Whether you're a leader, entrepreneur, or high achiever feeling stretched thin, this conversation will help you discover how to work in alignment with your instincts and create sustainable success. What We Discuss in this Episode The three parts of the mind: cognitive, affective, and conative What the Kolbe Index measures and why it differs from personality assessments Why capability and desire aren't enough for sustainable performance How natural instincts influence decision-making, problem-solving, and execution The four Action Modes and how they show up in everyday work Why procrastination may be a clue - not a character flaw How to identify the tasks that energize you versus those that drain you The connection between burnout and working against your natural strengths Why time management is often less important than energy management How leaders can build stronger teams by understanding individual strengths The difference between hiring clones and creating complementary teams Why diverse thinking styles improve innovation, adaptability, and execution How to give people freedom while maintaining accountability for results Practical ways to create a more resilient, high-performing workforce Highlights and YouTube Chapter Titles 00:00 – What If You're Not Burned Out? 02:00 – Understanding the Conative Part of the Mind 05:00 – Why Natural Instincts Matter More Than You Think 08:00 – The Four Action Modes Explained 12:00 – The Hidden Reason Behind Procrastination 15:00 – Working in Your Zone of Strength 18:00 – The Kolbe Exercise That Reveals How People Take Action 22:00 – Building Teams Like a Chess Master, Not a Checkers Player 26:00 – Why Great Leaders Focus on Strengths, Not Weaknesses 29:00 – Hiring Complements Instead of Clones 32:00 – The Power of Diverse Thinking Styles 35:00 – Time Management vs. Energy Management 39:00 – Why Certain Tasks Drain You Faster Than Others 43:00 – Doing More by Working More Naturally 46:00 – Creating Sustainable Success and Freedom to Be Yourself Episode Show Notes and Links: https://leadersoftransformation.com/podcast/leadership/553-do-more-more-naturally-with-amy-bruske/ Check out our complete library of episodes and other leadership resources here: https://leadersoftransformation.com ________
