Psychedelic drugs have received attention recently for their potential use as treatments for psychiatric disorders. Single, high doses of LSD have shown promise for treating depressive disorders. But there's another way in which people have been using LSD, and it's what's known as micro-dosing, taking LSD at below noticeable levels, where it doesn't seem to have a psychedelic impact—but users say it does in fact have an impact on their overall sense of well-being. This is just what Harriet de Wit, University of Chicago professor in the department of psychiatry and behavioral neuroscience, wanted to study. Abstract: Psychedelics have emerged as promising candidate treatments for various psychiatric conditions, and given their clinical potential, there is a need to identify biomarkers that underlie their effects. Here, we investigate the neural mechanisms of lysergic acid diethylamide (LSD) using regression dynamic causal modelling (rDCM), a novel technique that assesses whole-brain effective connectivity (EC) during resting-state functional magnetic resonance imaging (fMRI). We modelled data from two randomized, placebo-controlled, double-blind, cross-over trials, in which 45 participants were administered 100 g LSD and placebo in two resting-state fMRI sessions. We compared EC against whole-brain functional connectivity (FC) using classical statistics and machine learning methods. Multivariate analyses of EC parameters revealed widespread increases in interregional connectivity and reduced self-inhibition under LSD compared to placebo, with the notable exception of primarily decreased interregional connectivity and increased self-inhibition in occipital brain regions. This finding suggests that LSD perturbs the Excitation/Inhibition balance of the brain. Moreover, random forests classified LSD vs. placebo states based on FC and EC with comparably high accuracy (FC: 85.56%, EC: 91.11%) suggesting that both EC and FC are promising candidates for clinically-relevant biomarkers of LSD effects. BetterLife Pharma CEO Ahmad Doroudian joined Steve Darling to share details the company has obtained the first set of neurological receptor binding data on its lead compound, 2-bromo- LSD which was generated by Eurofins Discovery. Doroudian saying the TD-0148A which is a second-generation Lysergic Acid Diethylamide or LSD derivative molecule, and the company feels will mimic the projected therapeutic potential of LSD without causing the undesirable psychoactive dissociative side effects, such as hallucinations. Doroudian sad this initial data shows significant TD-0148A binding to receptors while there is none to limited binding to receptors such as 5-HT3, GABA-A1, and NMDA receptors.

Interest is growing in the use of the psychedelic drug LSD for psychiatric research and even potentially for treatment. But placebo-controlled studies conducted to date have used just one dose of the drug—none have investigated the impacts of a variety of dosages within the same subjects. In addition, past studies did not use pharmaceutically-defined dosages of LSD, which has made verifying the effects of a particular dose difficult. To address this gap, Matthias Liechti, professor in the department of clinical pharmacology and toxicology at the University of Basel in Switzerland, and his colleagues conducted a study. Listen NowPsychedelic agents or drugs, for example Lysergic Acid Diethylamide or LSD (a cereal fungus and first synthesized in 1938), were widely researched for their therapeutic potential through the 1950s. Among other noted users of these agents was the actor, Cary Grant. However, when these drugs escaped the research lab in the 1960s, moral panic set in. America's youth would, it was feared, "turn on, tune in and drop out" (a phrase popularized the Berkeley/Harvard psychologist Timothy Leary) and social upheaval would ensue. By 1970 LSD, psilocybin and other psychedelic agents were banned for both research and recreational use purposes. Beginning in the 1990s the FDA began giving limited approval to conduct psychedelic-related efficacy studies. Two decades later it appears the US has now, finally, turned the corner on researching the potential wide spread therapeutic benefits of psychedelics. This was again made evident last month when by Johns Hopkins announced opening its Center for Psychedelic and Consciousness Research. This announcement was preceded in April by the Imperial College in London announcing the opening of its Centre for Psychedelic Research. During this 26 minute interview, Dr. Doblin begins by describing what effect these agents have on the brain or what parts of the brain are stimulated to what effect. He discusses moreover his organization's MDMA (methylenedioxy-methamphetamine) research, currently a Phase 3 trial to study the effects on patients suffering from PTSD (oddly not supported by the DoD or VA), and a wide range of other current research efforts concerning treating, among other diagnoses, addiction, depression, eating disorders, phobias, OCD, schizophrenia and terminal illnesses. He also weighs in on the future legal status of these drugs, again currently illegal or banned as Schedule I controlled substances. Rick Doblin, Ph.D., is the founder and executive director of the Multidisciplinary Association for Psychedelic Studies (MAPS). He received his doctorate in Public Policy from Harvard's Kennedy School of Government, where he wrote his dissertation on the regulation of the medical uses of psychedelics and marijuana and his Master's thesis on a survey of oncologists about smoked marijuana vs. the oral THC pill in nausea control for cancer patients. His undergraduate thesis at New College of Florida was a 25-year follow-up to the classic Good Friday Experiment, which evaluated the potential of psychedelic drugs to catalyze religious experiences. He also conducted a thirty-four year follow-up study to Timothy Leary's Concord Prison Experiment. Rick studied with Dr. Stanislav Grof and was among the first to be certified as a Holotropic Breathwork practitioner. His professional goal is to help develop legal contexts for the beneficial uses of psychedelics and marijuana, primarily as prescription medicines but also for personal growth for otherwise healthy people, and eventually to become a legally licensed psychedelic therapist.
