Listen to the recent CSIS Bipartisan Alliance for Global Health Security book launch of Fair Doses: An Insider's Story of the Pandemic and the Global Fight for Vaccine Equity by Seth Berkley. As the gravity and magnitude of the Covid-19 pandemic became apparent in the first half of 2020, how did the vision for a mechanism to ensure equitable access to new vaccines come together and what role did organizations including Gavi, the Vaccine Alliance; the Center for Epidemic Preparedness Innovations (CEPI); and the World Health Organization (WHO) play in enabling the COVAX Facility to distribute nearly two billion doses of COVID-19 vaccines to people in 146 countries, the majority of which were low-income countries, by the end of  2022? This event featured conversation between Katherine Bliss, Senior Fellow and Director of Immunizations and Health Systems Resilience, CSIS Global Health Policy Center, and Dr. Seth Berkley, author of Fair Doses and CEO of Gavi, the Vaccine Alliance, from 2011 to 2023  regarding the challenges of vaccine equity and the lessons from the COVID-19 pandemic that can inform preparations for inevitable future health crises.  This event is made possible through the generous support of the Gates Foundation.

Tonix Pharmaceuticals Holdings CEO Dr. Seth Lederman joined Steve Darling from Proactive to announce that the first patient has been dosed in the investigator-initiated FOCUS study at Massachusetts General Hospital. The study is focused on adult patients with arginine-vasopressin deficiency (AVP-D)—a rare endocrine disorder formerly known as central diabetes insipidus—that is now understood to be linked to oxytocin deficiency and adverse mental health outcomes. Dr. Lederman explained that oxytocin, commonly known as the “bonding hormone,” plays a critical role in regulating mood, social behavior, and emotional wellbeing. Emerging evidence suggests that individuals with AVP-D suffer from low oxytocin levels, contributing to symptoms such as anxiety, depression, and socioemotional impairment. The FOCUS study is designed as a randomized, double-blind, placebo-controlled crossover pilot trial. It will assess Tonix's investigational intranasal potentiated oxytocin formulations—TNX-1900 (24 IU) and TNX-2900 (6 IU)—to determine their impact on mood and social functioning markers in patients with AVP-D. In addition to the single-dose evaluations, the study will include an exploratory two-week treatment phase to assess longer-term effects on mental health and quality-of-life outcomes. Dr. Lederman noted that Tonix is providing study support and that results from this research may inform the broader development of oxytocin replacement therapies for neuropsychiatric and endocrine-related disorders. The successful dosing of the first patient marks a significant milestone in Tonix's ongoing efforts to advance innovative, mechanism-driven therapies for complex disorders with unmet medical needs. #proactiveinvestors #tonixpharmaceuticalsholdingcorp #nasdaq #tnxp #Biotech #MPOXVaccine #Smallpox #TNX801 #VaccineDevelopment #ClinicalTrials #PharmaceuticalNews #MedicalResearch #WHO #GlobalHealth #InfectiousDiseases #Biopharma #ProactiveInvestors

We've been taught that “the dose makes the poison.” But what if that's no longer true? In this episode of Practical Nontoxic Living™, Sophia Ruan Gushée unpacks how low doses of toxic chemicals—like PFAS, endocrine disruptors, and heavy metals—can create powerful biological effects, especially during critical stages of life. You'll hear why the Endocrine Society warns that there may be no safe dose for certain hormone-disrupting chemicals, and how these exposures can interact like a “toxic cocktail” to amplify harm. Most importantly, Sophia offers simple, practical steps to reduce your exposure at home—because if small doses can harm, small changes can heal. Plus, stay tuned for next week's episode introducing Sophia's Household Repeat Offenders™ framework—a commonsense way to simplify detoxing your home and avoid dozens of toxic exposures at once.

Ozempic Breakthrough: New Study Reveals Significant Weight Loss Potential with Higher Doses and Holistic Approach

Oprah's Weight Loss Dilemma: The Ozempic

Ozempic and similar medications have remained at the center of the weight loss conversation this week, with new studies and high-profile voices like Oprah Winfrey bringing both excitement and nuanced caution. Major clinical research just published in a leading medical journal demonstrates that higher, triple doses of semaglutide, sold under names like Ozempic and Wegovy, are driving even more significant weight loss in people with obesity. According to researchers led by Dr Sean Wharton at a leading Canadian weight management clinic, adults without diabetes who took the highest 7 point 2 milligram dose weekly saw an average weight loss approaching nineteen percent of their body weight, notably higher than what's been seen with typical doses. Nearly half of those patients lost twenty percent or more of their body weight in these trials, suggesting very real potential for those struggling with severe obesity. The same trials found that people with type two diabetes saw weight drops of thirteen percent using the highest dose. Importantly, the higher dose still appeared safe and generally well tolerated, with common side effects including nausea and diarrhea that mostly resolved over time. The study did not find a higher risk of dangerous drops in blood sugar or other serious complications. These results are meaningful because they expand the promise of these drugs for patients not reaching their goals on standard doses. However, experts stress that further research is essential to fully understand the long term effects of such a powerful regimen and whether the benefits continue to outweigh any risks as time goes on.Beyond the numbers, more people are sharing their personal stories about these new drugs, and Oprah Winfrey continues to be among the most influential. In new interviews and her recent podcast episode, Oprah has confirmed that she uses a prescription GLP dash 1 agonist, the same class of medication as Ozempic, as part of her ongoing wellness journey. She has not named the specific brand, but her openness has made it clear that for her, this is not just a quick fix or shortcut. Instead, Oprah describes the decision as a mental shift—acknowledging obesity as a chronic disease that is best managed with a partnership between lifestyle changes and medical treatment. She has been explicit that the medication alone is not magic. Oprah credits her progress to a holistic routine that includes daily exercise, often hiking three to five miles, planful eating with her last meal in the late afternoon, and close attention to water intake and overall health habits. She has left behind the self blame that characterized her earlier struggles and now uses her platform to challenge the idea that using medication means failure. Instead, she frames it as using every available tool responsibly—paired with self compassion and ongoing commitment. As these drugs become more widely used, big questions remain. A new RAND Corporation report finds that nearly one in eight Americans have tried Ozempic or a similar medication. Use is particularly high among women aged fifty to sixty four. Yet significant numbers report troublesome side effects—most commonly nausea and diarrhea—though these are usually manageable. There are persistent concerns about access and continuity, too. Research just presented at the European Association for the Study of Diabetes finds that about half of patients stop using these drugs within a year, mostly due to high cost, with prices remaining out of reach for many, especially younger people and those with lower incomes. Muscle loss, especially among older adults and women, is another emerging potential downside discussed in the latest studies, reminding users to stay mindful of their overall health—not just what the scale shows. And despite the massive popularity, there are still new legal warnings and lawsuits over rare but serious complications, such as gastroparesis, which can slow digestion to a dangerous degree. The US Food and Drug Administration has continued to update warning labels as new risks are identified. For listeners considering this medication, the message from both doctors and people like Oprah is clear. Ozempic and its cousins can be life changing, but only as part of a larger strategy, with realistic understanding of the risks and the need for sustained healthy habits. Science is still catching up to the real life experience of millions as both hope and caution shape the next chapter of the weight loss revolution. Thanks for listening, please subscribe, and remember—this episode was brought to you by Quiet Please podcast networks. Epinephrine auto-injectors, first developed in the 1970s, are the most common emergency treatments for anaphylaxis, often deadly allergic reactions. The limitations of liquid epinephrine and the device's decades-old technology have kept these life-saving devices out of patients' hands. Austin-based startup Windgap Medical, Inc. plans to break these barriers with a more convenient, shelf-stable alternative. In Episode 41 of the MedTech Speed to Data podcast Key Tech's Andy Rogers and Thomas James sit down with Windgap's co-founder and Chief Business Officer, Brent Buchine, to discuss the data-driven development of the company's life-saving technology.Need to know·       Epinephrine auto-injectors are big business — A $1.3 billion market at Windgap's founding, sales of epinephrine auto-injectors exceeded $3.1 billion in 2024.·       Few eligible patients get these life-saving devices — Only 52% of American food allergy patients ever receive prescriptions, and epinephrine auto-injectors are only available in 32% of the world's 195 countries.·       Traditional auto-injectors are relatively large and inconvenient — Only 55% of patients with prescriptions report having immediate access to their auto-injectors.·       Liquid epinephrine is thermally unstable — Doses lose their effectiveness with prolonged heat exposure, forcing patients to refill their prescriptions more frequently. The nitty-grittyBuchine and his co-founders saw an opportunity to make epinephrine delivery more convenient and accessible. “We developed a freeze-dried version of epinephrine to make it more stable and double, if not triple, the shelf life,” Buchine explains. However, lyophilization introduces an extra step in the treatment process. The dried epinephrine must be rehydrated and mixed in a solution before injection.“If you have a rescue product for emergency use, you have to get it very quickly,” Buchine says. “With modest training, you need to make sure people know how to use the product because their life is at risk if it doesn't work.”Windgap's ANDI® platform is a small, highly portable single-dose auto-injector, Buchine explains. “Simply twisting the cap automatically rehydrates that dose in a couple of seconds — no shaking, no swirling required. It's ready to inject by pressing the device next to the injection site.”Windgap and its pharmaceutical clients are still in the commercialization phase, but the company is already looking at the future of complex injectables.“We see that, fundamentally, formulation pipelines are getting more and more challenging,” Buchine says. “You have multiple injections, you have mixing, you have high viscosity/high volume. The conventional options out there are not as suitable anymore. We're solving those problems specifically because we think there's an opportunity to be best in class in that area.”Data that made the difference:Developing combination products is a multi-stakeholder problem. “It's drug, it's device, it's patient, it's prescriber, it's payer. You've got to think about all of those stakeholders along your development.”Get in front of stakeholders to understand the problem. “We did a lot of surveys and uncovered the opportunity. Patients weren't getting prescriptions filled or weren't even going to the doctor to get prescriptions. It was that segment that we spent a lot of time talking to.”Listen to your customers. “We'll talk to pharmaceutical companies [and ask] what are some of the biggest challenges you're facing in your pipeline? And then you just listen. Over time, you look for that recurring theme. That's what really drove our product strategy.”Use data to convince investors. “There was a vastly underserved market. Our ability to communicate that to investors and help them understand the opportunity of taking [at the time] a $1.3 billion business to something substantially above that.”

But Ayurveda and yogic wisdom remind us that it's not about how long you practice — it's about how often you return to presence. They explore the challenges of libido loss, the role of therapy, and the transformative effects of testosterone therapy. The conversation highlights the importance of understanding hormonal health in relationships and the awakening of self-awareness that comes with it. I n this conversation, Marcella Hill and Dr. Kelly Casperson explore the complexities of libido, sexual shame, and the transformative power of hormones in midlife relationships. They discuss the importance of self-discovery, emotional honesty, and the need for patience in navigating personal and relational growth. The dialogue emphasizes that pleasure and desire are not solely for others but are essential for individual fulfillment and connection. Takeaways The relationship was initially strong but faced challenges due to hormonal changes. Libido loss can lead to feelings of anxiety and disconnection in relationships. Therapy can help with communication but may not directly address libido issues. Medical interventions, like hormone therapy, can significantly impact well-being. Testosterone therapy revitalized Marcella's energy and libido, transforming her relationship. Awakening to a new self can create challenges in long-term relationships. Couples can navigate hormonal changes together for better understanding and support. Increased awareness of hormonal health can improve communication and intimacy. Hormones play a crucial role in mental health and overall well-being. Understanding the effects of hormones can lead to proactive health measures. Women often feel shame about their sexual desires. Listening to uncomfortable content can help challenge sexual shame. Orgasm is not just a sexual act; it's about personal joy and passion. Women deserve to feel turned on for their own pleasure, not just for others. Men's enjoyment in relationships often comes from seeing their partner's pleasure. Communication about sexual needs is crucial for both partners. Exploring sexuality can enhance intimacy and connection in relationships. Emotional honesty leads to a more expansive and fulfilling relationship. Hormone therapy can reignite passion and feelings in midlife. LETHAL DOSES - The Story Behind The Godfather Of Fentanyl

On a cold afternoon in February 1991, a frightening new drug hit the streets of New York City, a synthetic narcotic marketed in packets labeled “Tango & Cash.” As police scrambled to warn heroin users of the danger, the overdose victims began piling up in hospital emergency rooms and county morgues across three states. As a Drug Enforcement Administration agent said at the time, “We don't know yet who's putting this stuff out there, but whoever he is, he's an ice-cold son of a bitch.” Fentanyl had come to America. In 2024, fentanyl is killing nearly 200 Americans every day, and not just heroin users, a seemingly unstoppable narcotic curse like none ever seen before. But few know that this plague began in the brilliant mind of the high-school dropout and chemistry prodigy that the DEA called “the best and most dangerous clandestine chemist” it has ever encountered. The clandestine chemist was George Erik Marquardt. Starting at just twelve years old, Marquardt used his extraordinary talents to make every illegal drug in the book, from bootleg booze to heroin. He brewed LSD for Timothy Leary and the Grateful Dead, methamphetamine for outlaw motorcycle gangs, nerve gas for Idaho Nazis, and even life-saving AZT for AIDS patients. But when that ice-cold son of a bitch turned to fentanyl, thousands of Americans would die. In LETHAL DOSES: The Story Behind ‘The Godfather of Fentanyl,' award-winning author and former undercover agent for the Oklahoma Bureau of Narcotics John Madinger, tells the remarkable story of DEA's three-year pursuit, the genesis of our fentanyl problem today, and the uniquely dangerous evil genius he spent hundreds of hours interviewing. Now you can read the incredible book on which the hit docuseries, THE GODFATHER OF FENTANYL, is based!Become a supporter of this podcast: https://www.spreaker.com/podcast/the-opperman-report--1198501/support.
