Mary I. O'Connor, MD is founder and Chief Medical Officer of Vori Health, a virtual musculoskeletal company focus on transforming the delivery of patient-centered and value drive care. She is a nationally recognized leader in health equity and Chair of the Board of Directors of Movement is Life, a nonprofit multi-stakeholder coalition committed to addressing musculoskeletal health disparities. Her new book, “Taking Care of You: The Empowered Woman's Guide to Better Health,” supports women advocating for equitable healthcare. A past Olympian (US Women's Rowing), she is passionate in her promotion of the power of sports for girls and women. Dr. O'Connor is Professor Emerita of Orthopedics at Mayo Clinic and past Professor of Orthopaedics and Rehabilitation at Yale School of Medicine. She received her MD from Drexel University and completed her residency in orthopedics and fellowship in orthopedic oncology at the Mayo Clinic in Rochester, MN. She practiced at Mayo Clinic in Florida until 2015 during which time she served in many leadership roles: Enterprise-wide Medical Director of the Office of Integrity and Compliance; Chair, Orthopedic Surgery Department (Florida); Medical Director for Development (Florida); and member of the Executive Operations Team (Florida). In 2015, she became the inaugural Director of the Center for Musculoskeletal Care at Yale School of Medicine and Yale New Haven Health. In February 2021, Dr. O'Connor became a founder and Chief Medical Officer at Vori Health to advance her passion for transforming musculoskeletal care. Dr. O'Connor has published extensively on clinical research and innovation in care pathways, hip fracture care, limb salvage for tumor, sex and gender differences in arthritis, and regenerative medicine. At Yale, she led the creation of a multidisciplinary hip fracture program which transformed clinical outcomes and promoted innovation in the care of this vulnerable population. She authors a quarterly column in Clinical Orthopaedics and Related Research entitled, “Equity360: Gender, Race and Ethnicity,” to advance health equity in the orthopedic profession. She has broken numerous gender barriers as the first female member of the Musculoskeletal Tumor Society (MSTS), the International Society of Limb Salvage (ISOLS), the American Association of Hip and Knee Surgeons (AAHKS) and The Knee Society. She is the Past President of the Association of Bone and Joint Surgeon, AAHKS, ISOLS, MSTS and the Ruth Jackson Orthopaedic Society. She is past Chair of the American Academy of Orthopaedic Surgery (AAOS) Diversity Advisory Board and the AAOS Women's Health Issues Advisory Board, founding member of the AAHKS Women in Arthroplasty Group, past board member of the Perry Foundation, and member of the AAHKS Diversity Advisory Board. She is past member of the Advisory Committee on Research on Women's Health at NIH. Dr. O'Connor has received numerous awards and honors during her training and career, including the 2023 AAOS Diversity Award, 2023 AAHKS Diversity Award, Distinguished Clinician Award at Mayo Clinic, and the Corinne Farrell Award from the International Skeletal Society. Elise is active in sports and brings awareness to childhood cancer, osteosarcoma, and amputees with her active lifestyle. Valerae O. Lewis, M.D. Professor, Orthopaedic Oncology, Division of Surgery Chair, Department of Orthopaedic Oncology, Division of Surgery Dr. John Murray Professor in Orthopaedic Oncology The University of Texas M. D. Anderson Cancer Center Dr. Lewis, the John Murray Professor of Orthopaedic Oncology, attended Yale University and matriculated at Harvard Medical School, graduating with honors. She completed her Orthopaedic training at the Harvard Combined Orthopaedic Residency Program in Boston, MA and her fellowship in Musculoskeletal Oncology at the University of Chicago. The first African American woman to be awarded the MD Anderson Faculty Achievement Award in Patient Care she is now the Chair of the Department of Orthopaedic Oncology at MD Anderson. Dr. Lewis is very active nationally and internationally within Orthopaedics and Musculoskeletal Oncology. She is a strong advocate of education, and as such, served as the Chair of the Musculoskeletal Tumor Society (MSTS) Education Committee, Chair of the American Academy of Orthopaedic Surgery (AAOS) Instructional Course Lecture Tumor Subcommittee, Chair of the Musculoskeletal Oncology Content Committee for the AAOS Council on Education and Chair of the International Society of Limb Salvage (ISOLS) Education Committee. A former board member of the American Orthopaedic Association, the Western Orthopaedic Association, MSTS and the ISOLS, she was recently elected to the Board of Directors of the AAOS Dr. Lewis' area of expertise is in the treatment of both children and adults with bone and soft tissue sarcomas. Her clinical research focuses on the outcome of patients after hemipelvectomy surgery and her lab investigates the gene functions tied to osteosarcoma and the development of treatments that might interrupt this process. He moved to the United States about 15 years ago and is currently based in Overland Park Kansas.TreviPay has had some significant changes in the past year and Brandon and I discuss those – which include the rebranding of the company from MSTS to TreviPay and the acquisition of the company by Corsair Capital. TreviPay is a business-to-business payments company and they focus on the seller and help them to grow their business through the extension of payment terms to their clients. Brandon is an avid sports fan and specifically a Kansas City Chiefs football fan – which of course has come with its high and lows over the last two years. On the professional side, leadership development is one of his primary passions.

The announcement of Justin Sun buying Steemit, Inc. has been met with so many mixed feelings and confusion. It's a big deal for anyone who's paying attention to Steem. There's good and bad to this whole situation. So far, nothing bad has happened. We've seen Steemit, Inc. leave the hands of a founder who had grown uninterested in the project (Ned) and lacked vision, execution and resources to bring Steem(it) any further. In my view, all the same risks exist. Ned could just as easily use Steemit and his STEEM stake to overtake the blockchain in the same way that people are worried that Justin might try to do. The thing that kept Ned from doing that is the same thing that will keep Justin from doing it and it also happens to be the same reason why 51% attacking the BTC network doesn't make any sense — the game is created in such a way that the financial incentives lie in propping up the blockchain and growing the network, not tearing it down. If Justin stomps Steem into the ground, then he loses a major investment. It doesn't make financial sense. If Justin, however, allows the Steemit team to continue their development on Communities and SMTs (not to be confused with MSTs), and then he puts his full marketing dollars and weight behind those launches and announces them to the world… think of what that could do for Steem. Think of all the major attention that our blockchain will get. The floodgates could open once again. There are pros and cons to every situation. There is a bright side, but *you* have to be willing to look at it. Keep your awareness about the downside and understand the **risks** of having Justin in a position of power, but also keep your awareness about the upside and understand the **benefits** of having Justin in a position of power. In plants, long distance transport of sugars from photosynthetic source leaves to sink organs comprises different crucial steps depending on the species and organ types. Sucrose, the main carbohydrate for long distance transport is synthesized in the mesophyll and then loaded into the phloem. After long distance transport through the phloem vessels, sucrose is finally unloaded towards sink organs. Alternatively, sugar can also be transferred to non-plant sinks and plant colonization by heterotrophic organisms increases the sink strength and creates an additional sugar demand for the host plant. These sugar fluxes are coordinated by transport systems. Main sugar transporters in plants comprise sucrose (SUTs) and monosaccharide (MSTs) transporters which constitute key components for carbon partitioning at the whole plant level and in interactions with fungi. Although complete SUTs and MSTs gene families have been identified from the reference Dicot Arabidopsis thaliana and Monocot rice (Oriza sativa), sugar transporter families of the leguminous plant Medicago truncatula, which represents a widely used model for studying plant-fungal interactions in arbuscular mycorrhiza (AM), have not yet been investigated. With the recent completion of the M. truncatula genome sequencing as well as the release of transcriptomic databases, monosaccharide and sucrose transporter families of M. truncatula were identified and now comprise 62 MtMSTs and 6 MtSUTs. I focused on the study of the newly identified MtSUTs at a full family scale; phylogenetic analyses showed that the 6 members of the MtSUT family distributed in all three Dicotyledonous SUT clades; they were named upon phylogenetic grouping into particular clades: MtSUT1-1, MtSUT1-2, MtSUT1-3, MtSUT2, MtSUT4-1 and MtSUT4-2. Functional analyses by yeast complementation and expression profiles obtained by quantitative RT-PCR revealed that MtSUT1-1 and MtSUT4-1 are H+/sucrose symporters and represent key members of the MtSUT family. Conservation of transport capacity between orthologous leguminous proteins, expression profiles and subcellular localization compared to previously characterized plant SUTs indicate that MtSUT1-1 is the main protein involved in phloem loading in source leaves whilst MtSUT4-1 mediates vacuolar sucrose export for remobilization of intracellular reserve. The AM symbiosis between plants and fungi from the phylum Glomeromycota is characterized by trophic exchanges between the two partners. The fungus supplies the autotrophic host with nutrients and thereby promotes plant growth. In return, the host plant provides photosynthate (sugars) to the heterotrophic symbiont. Here, sugar fluxes from plant source leaves towards colonized sink roots in the association between the model leguminous plant M. truncatula and the arbuscular mycorrhizal fungus (AMF) Glomus intraradices were investigated. Sugar transporter candidates from both the plant and fungal partners presenting differential expression profiles using available transcriptomic tools were pinpointed. Gene expression profiles of MtSUTs and sugar quantification analyses upon high and low phosphorus nutrient supply and inoculation by the AMF suggest a mycorrhiza-driven stronger sink in AM roots with a finetuning regulation of MtSUT gene expression. Conserved regulation patterns were observed for orthologous SUTs in response to colonization by glomeromycotan fungi. In parallel, a non-targeted strategy consisting in the development of a M. truncatula - G. intraradices expression library suitable for yeast functional complementation and screening of symbiotic marker genes, similar to the approach that led to the identification of the first glomeromycotan hexose transporter (GpMST1), has been developed in this study. Taken together, with the identification, functional characterization and gene expression pattern of sugar transporter families, a more complete picture of sugar fluxes in the AM symbiosis has begun to emerge. This study opens new perspectives by identifying interesting candidate genes involved in sugar partitioning at both the plant and fungal levels and at the symbiotic interface in the widely used AM symbiosis model between M. truncatula and G. intraradices.

Background: Prognosis of patients with metastatic soft tissue sarcomas (MSTS) is poor even after response to doxorubicin-based chemotherapy. We report phase II data of highdose chemotherapy and peripheral blood stem cell (PBSC) rescue in patients with MSTS responding to AI-G chemotherapy. Patients and Methods: From 1997 to 2002, 55 patients with MSTS were prospectively treated with 4 cycles of AI-G (doxorubicin 75 mg/m(2), ifosfamide 6 g/m(2) with G-CSF support). Responders received 2 further cycles of AI-G with collection of PBSCs. High-dose chemotherapy consisted of ifosfamide 12 g/m(2), carboplatin 1.2 g/m(2) and etoposide 1.2 g/m(2) (HD-ICE) followed by reinfusion of PBSCs. Results: Twenty-one of 55 patients (38%) were assessed as responders (3 complete response, 18 partial response). All but 2 patients refusing treatment received high-dose chemotherapy with PBSC rescue leading to grade IV hematologic toxicity without severe infections in all patients. No toxic death occurred. After a median follow-up time of 30 months, the median progression-free time was 12 months and survival time was 22 months for the entire group. By intent-totreat analysis the probability of 5-year progression-free survival was significantly higher for patients allocated to HD-ICE compared to patients receiving second-line chemotherapy after failure of AI-G (14 vs. 3%; p = 0.003). The estimated 5-year overall survival between the 2 groups was different (27% vs. not reached) but did not reach significance (p = 0.08). Conclusion: HD-ICE is feasible and promising in patients with chemosensitive MSTS. A randomized phase III trial is warranted to further define the role of HD-ICE as consolidation treatment in these patients.
