On October 18, 2024, the FDA approved zolbetuximab plus fluoropyrimidine- and platinum-containing chemotherapy for the frontline treatment of adult patients with locally advanced unresectable or metastatic, HER2-negative, CLDN18.2-positive gastric or GEJ adenocarcinoma. This regulatory decision was backed by findings from the phase 3 SPOTLIGHT (NCT03504397) and GLOW (NCT03653507) trials. In SPOTLIGHT, In SPOTLIGHT, patients who received zolbetuximab plus mFOLFOX6 (5-fluorouracil, leucovorin, and oxaliplatin) achieved a median progression-free survival (PFS) of 10.6 months (95% CI, 8.9-12.5) vs 8.7 months (95% CI, 8.2-10.3) in those who received placebo plus chemotherapy (HR, 0.750; 95% CI, 0.601-0.936; 1-sided P = .0053). In GLOW, zolbetuximab plus CAPOX (capecitabine and oxaliplatin) generated a median PFS of 8.2 months (95% CI, 7.5-8.8) vs 6.8 months (95% CI, 6.1-8.1) with placebo plus CAPOX (HR, 0.771; 95% CI, 0.615-0.965; 1-sided P = .0118). In our exclusive interview, Dr Ajani discussed key considerations for the clinical use of zolbetuximab, including its anticipated adverse effect profile and the introduction of CLDN18.2 as a biomarker for assessment prior to treatment initiation.

Dus waarom geen kerstlied gezongen door een grote groep vocalisten uit de Nederlandse theaterwereld? Het werd een hele grote groep en was het een hele grote klus dat een uitstekende organisatie vereiste. Het resultaat is fantastisch! Luister maar en het lied kruipt direct onder je huid. When we speak about India and its relationship with the world, we often focus on what we have received from the world. But what about what we have given to it? This doesn’t get covered as much. Even when it does, we go to the extremes. Either we were a vishwaguru or our only contribution is ‘zero’. Is there a more nuanced view? That’s what we explore in this episode. The effects of COVID-19 reach far beyond the realm of human health. In Spotlight this week Pastor Calvin Lindstrom and I consider Americans' right of religious liberty vs. submission to governmental authorities, especially in light of restrictions on “non-essential,” in-person worship services and church gatherings. Should Christians be governed by Hebrews 10:25 or Romans 13 and what is a proper, biblical response to government overreach? After the break the conversation turns to pending legislation, specifically SB 2762 which would require comprehensive sex ed starting in kindergarten (with NO parental opt-out) and SB 3788 which, among other things, deletes teaching honor and respect for monogamous, heterosexual marriage. Pastor Lindstrom presents strong reasons to pursue private Christian education or home education once the school shutdown is lifted and provides informative resources for families (here and here). The podcast concludes with encouragement from Pastor Lindstrom to stay engaged in the political process and to VOTE! In Spotlight werden wir in unregelmäßigen Abständen Leute aus der (vorerst) deutschsprachigen Comic-Szene Interviewen und diese somit euch etwas näher bringen. Das kann von PR-Menschen eines größeren Verlags über Zeichner oder Autoren gehen und der Anfang macht Jano Rohleder. Jano ist der Chef des Ein-Mann-Comic-Verlages Dani Books und spricht mit unserem Chris über seinen Alltag, den Tücken des Verlagslebens und wie man als deutscher Verlag überhaupt an Lizenzen kommt. In Spotlight on Asia this week, RFI's Rosslyn Hyams meets Jean-Joseph Boillot who spends his time as an academic researcher travelling between France and India. Boillot's latest book -  soon to be published in English in India - is called L'Inde Ancienne au chevet des Politiques (literally Ancient India on the Politicians' Bedside Table) takes parts of an ancient Indian treatise on how to rule, and what leaders should do to attain prosperity for all. He says that Narendra Modi, apparently a fan of the Arthashashtra represents a new generation of Indian leaders who refer to their own culture's teachings and experiences rather than borrowing from foreign sources. Boillot suggests that European politicians today take a leaf out of the book of Kautilya, the right-hand man of the Indian Maurya Empire's Ashoka who reigned some 2300 years ago. Under the headings such as "on discipline", "on good behaviour of counsellors and ministers", "on safety of assets and persons" , "on the kingdom's vices and calamities", it sounds quite contemporary. Boillot makes it more relevant by updating the vocabulary in his translation, while reminding us that even Ashoka, remembered as a great leader, made mistakes and lost his empire.
