BUFFALO, NY- March 20, 2024 – A new #research paper was #published in Oncotarget's Volume 15 on March 14, 2024, entitled, “Analytical validation of NeXT Personal®, an ultra-sensitive personalized circulating tumor DNA assay.” In this new study, researchers Josette Northcott, Gabor Bartha, Jason Harris, Conan Li, Fabio C.P. Navarro, Rachel Marty Pyke, Manqing Hong, Qi Zhang, Shuyuan Ma, Tina X. Chen, Janet Lai, Nitin Udar, Juan-Sebastian Saldivar, Erin Ayash, Joshua Anderson, Jiang Li, Tiange Cui, Tu Le, Ruthie Chow, Randy Jerel Velasco, Chris Mallo, Rose Santiago, Robert C. Bruce, Laurie J. Goodman, Yi Chen, Dan Norton, Richard O. Chen, and John M. Lyle from Personalis, Inc. describe the analytical validation of NeXT Personal®, an ultra-sensitive, tumor-informed circulating tumor DNA (ctDNA) assay for detecting residual disease, monitoring therapy response, and detecting recurrence in patients diagnosed with solid tumor cancers. “NeXT Personal uses whole genome sequencing of tumor and matched normal samples combined with advanced analytics to accurately identify up to ~1,800 somatic variants specific to the patient's tumor.” A personalized panel is created, targeting these variants and then used to sequence cell-free DNA extracted from patient plasma samples for ultra-sensitive detection of ctDNA. The NeXT Personal analytical validation is based on panels designed from tumor and matched normal samples from two cell lines, and from 123 patients across nine cancer types. Analytical measurements demonstrated a detection threshold of 1.67 parts per million (PPM) with a limit of detection at 95% (LOD95) of 3.45 PPM. NeXT Personal showed linearity over a range of 0.8 to 300,000 PPM (Pearson correlation coefficient = 0.9998). Precision varied from a coefficient of variation of 12.8% to 3.6% over a range of 25 to 25,000 PPM. The assay targets 99.9% specificity, with this validation study measuring 100% specificity and in silico methods giving a confidence interval of 99.92 to 100%. “In summary, this study demonstrates NeXT Personal as an ultra-sensitive, highly quantitative and robust ctDNA assay that can be used to detect residual disease, monitor treatment response, and detect recurrence in patients.” DOI - https://doi.org/10.18632/oncotarget.28565 Correspondence to - John M. Our educational systems, from pre school through the graduate level, are in a constant process of evolution and, in many ways improvement, as pedagogy becomes more sophisticated and curricula become more specialized in response to an ever more complex world. And yet, in some ways we're not at all keeping up and our schools and universities are falling short of how they need to be preparing students to succeed in the 21st century. But those gaps can also create opportunities for entrepreneurs and others to step in and help address unmet needs. Joshua Anderson is founder of Accelerant Education, a Baton Rouge-based company that develops software specifically designed to help high school students learn computer science and computer programming. Joshua started Accelerant when he realized that only about half of the high schools in Baton Rouge have computer science programs. Joshua understands the value of a computer science education: he is a self-taught software engineer and a graduate of LSU's Flores MBA program. He also directs StartupGrind Baton Rouge, a community of entrepreneurs and intrapreneurs. Marina Biragova is Executive Director of the Technology and Entrepreneurship Clinic at Southern University Law Center here in Baton Rouge. The clinic assists under-represented entrepreneurs across the state with their business formation and structuring, intellectual property, and compliance issues. Marina is also an Intellectual Property attorney and a small business advocate, with a specialty in technology and business law. She is a graduate from Pyatigorsk Institute of Economics and Governance School of Law, which is in southwest Russia, as well as the LSU Law Center. What new opportunities exist for those already in or considering entering the world of home energy ratings? Are there career specializations available? In today's podcast, Josh Anderson (TopBuild) and Scott Doyle (RESNET) help us understand the purpose, scope and functions of the RESNET HERS modeler designation. In 2020, the RESNET Board of Directors voted for the creation of a new RESNET HERS Rater modeling designation with a goal of increasing the consistency in the input in HERS Ratings through the training and certification of persons who conduct the modeling and analysis using accredited HERS Software Tools. Upon certification, HERS Modeler candidates are issued a unique RESNET Modeler ID which can be used for single or multiple HERS Software Tools. In many ways this certification is like the current Rating Field Inspector certification for field inspection and testing including candidates being posted on the RESNET Registry. Josh discusses how this is a natural evolution of the industry, especially as businesses scale. He also notes this helps to create an alternate career path within the RESNET ecosystem as it embodies a different skillset and an alternate work environment (office vs. field) for a more diverse workforce. Scott gives us an overview of the formal training required and notes that continuing education will be required to maintain the certification. The continuing education will be offered by the accredited HERS Rating Software Program developers. Scott notes that certified RESNET HERS Raters, having taken inputs and plan reviews as part of RESNET HERS Rater Training, will automatically receive the new certification without any additional fees, yet would still have to meet the continuing education requirements.
