Abstract: The 7 nicotinic acetylcholine receptor (nAChR), a potential drug target for treating cognitive disorders, mediates communication between neuronal and non-neuronal cells. Although many competitive antagonists, agonists, and partial-agonists have been found and synthesized, they have not led to effective therapeutic treatments. In this context, small molecules acting as positive allosteric modulators binding outside the orthosteric, acetylcholine, site have attracted considerable interest. Two single-domain antibody fragments, C4 and E3, generated through alpaca immunization with cells expressing the extracellular domain of the human 7-nAChR, are herein described. They bind to the 7-nAChR but not to the other major nAChR subtypes, 4{beta}2 and 3{beta}4. E3 acts as a slowly associating type I positive allosteric modulator, strongly potentiating the acetylcholine-elicited currents, while not significantly altering the desensitization of the receptor. An E3-E3 bivalent construct shows similar potentiating properties but displays very slow dissociation kinetics conferring quasi-irreversible properties. Whereas, C4 does not alter the receptor function, but fully inhibits the E3-evoked potentiation, showing it is a silent allosteric modulator competing with E3 binding. Both nanobodies do not compete with -bungarotoxin, localizing at an allosteric extracellular binding site away from the orthosteric site. The functional differences of each nanobody, as well as the alteration of functional properties through nanobody modifications indicate the importance of this extracellular site. The nanobodies will be useful for pharmacological and structural investigations; moreover, they, along with the extracellular site, have a direct potential for clinical applications. Abstract: The human 7 nicotinic receptor is a pentameric channel mediating cellular and neuronal communication. It has attracted considerable interest to design ligands for the treatment of neurological and psychiatric disorders. To develop a novel class of 7 ligands, we recently generated two nanobodies named E3 and C4 acting as positive and silent allosteric modulators respectively. Here, we solved the cryo-EM structures of the nanobody-receptor complexes. E3 and C4 bind to a common epitope involving two subunits at the apex of the receptor. They form by themselves a symmetric pentameric assembly that extends the extracellular domain. Unlike C4, the binding of E3 drives an active or desensitized conformation in the absence of orthosteric agonist, and mutational analysis shows a key contribution of a N-linked sugar moiety in mediating E3 potentiation. The nanobody E3, by remotely controlling the global allosteric conformation of the receptor, implements an original mechanism of regulation which opens new avenues for drug design. The C-Suite – is that something you strive for or where you are invited in? And more importantly, as a communicator, how do you make yourself listened to once you get there? The management paradigm, of which Grunig and his excellence theory in public relations being its strongest representative, argue that for PR to truly matter, the senior PR council should not only be part of but actively be involved in decision-making. This shifts PR from a purely technical role to a strategic and managerial role. One way to get there, Grunig (and all his supporters argue) is through education, through professionalization. Now this debate started in the 1980 but is as current as ever and that is how do you get into PR and how do you succeed in PR? Are PR practitioners born communicators or they become so after pursuing either a degree, or some other sort of organized form of studying PR or comms. In a sense, studying PR would put someone right at the heart of what the profession requires but also would have them exposed to all the questions about how PR came to be and what the future looks like: what is PR’s role in society, ethical conundrums, loyalties. On the other hand, starting straight into PR and learning on the go as a trainee or apprentice one would be focused on solving problems and therefore be more atuned to ..maybe… how the world ticks. My guest in this episode has done Edna now heads MarComms at Panalpina in Switzerland. She is an American who studied both in the US and Europe. She is a traveler, an explorer, a mother, a writer, a strategist. And she is also the managing Vice-President of EACD, the European Association of Communication Directors. Founded in Brussels in 2006, the EACD is a network for communication professionals from all fields promoting diversity, overcoming cultural barriers and fostering professionalism in communications. And today, I will ask her how she got to where she’s at.

Rachel has completed not one, but two things - hooray! First up is her Windmill Bay Stole, 8 stinking feet of fingering weight alpaca lovelieness. There is much relief that it is finished. She's also finished a handspun shawl for the Porpoise Fur booth at I Knit Fandango - Nymphalidea from Knitty.com designed by Melinda VerMeer, in Clotted Cream and YFP (aka fluorescent yellow) Shetland. She is still working on Painted, and has started another handspun shawl - Penrose Tile by Carol Feller.Allison finished her Green Memories colourwork hat for the Yarn in the City Not-a-KAL-KAL, and has done some spinning, as she is about to start adding to her fibre stash in a monthly fashion with the Hello Yarn Fiber Club.Charity KnittingWe start off with the amazing charity knitting done by 2014 Great London Yarn Crawlers, who donated over 150 items to our charity partner Refuge. We will be collecting again for Refuge this year, so please bring any and all charity items to the kick off party at the Yarn in the City Pop Up Marketplace on 5th September.We also recently learned about AYME (the Association of Young People with ME), an organization that supports the estimated 25,000 children and young people in the UK who suffer from myalgic enchephalomyelitis, also known as Chronic Fatigue Syndrome. AYME is launching an awareness campaign called "Knit a Tulip for ME", with a goal of collecting one knitted tulip for every ME-affected young person in the UK in 2015. So far, they've collected 5,000 woolen tulips, and they are holding an official launch on 14th May at the Double Tree by Hilton London - Chelsea with a display of some of the tulips already contributed. The campaign also has a Justgiving page, if you'd like to make a donation. You can also email Jackie (scroll down for the email address) for the pattern and a sponsorship form. There are a number of other charity knitting opportunities out there, including Knit for Peace, which supports people in India, Pakistan and Rwanda.
