M., Möbius, W., Früholz, I., Ackermann, F., Gültas, M., Garner, C. C., Hamra, F. K., Neef, J., Strenzke, N., Moser, T. Abstract: Cochlear inner hair cells (IHCs) form specialized ribbon synapses with spiral ganglion neurons that tirelessly transmit sound information at high rates over long time periods with extreme temporal precision. This functional specialization is essential for precise sound encoding and is attributed to a distinct molecular machinery with unique players or splice variants compared to conventional neuronal synapses. Among these is the active zone (AZ) scaffold protein piccolo/aczonin, which is represented by its short splice variant piccolino at cochlear and retinal ribbon synapses. While the function of piccolo at synapses of the central nervous system has been intensively investigated, the role of piccolino at IHC synapses remains unclear. In this study, we characterized the structure and function of IHC-synapses in piccolo gene-trap mutant rats (Pclogt/gt). We found a mild hearing deficit with elevated thresholds and reduced amplitudes of auditory brainstem responses. Ca2+ channel distribution and ribbon morphology were altered in apical IHCs, while their presynaptic function seemed unchanged. We conclude that piccolino contributes to the AZ organization in IHCs and is essential for normal synaptic transmission. Copy rights belong to original authors.
Prettige maandag! We are still away from the office for the holidays, but before we left, I recorded this episode on how to do a calendar audit for the year. This is a life-changing strategy I employ every year to really dig into what propelled me forward and, even more importantly, what I didn't even realize was holding me back. I know there are a lot of things out there we couldn't control this year, but that's all the more reason to focus on the things we can. Let's learn from 2020 and then make 2021 OUR. FREAKIN. YEAR. I've got a real treat for you folks this week - you get to hear me coach myself live on the podcast! Long story short, I saw all of your comments, posts, and questions this week all about hard things: what on earth are we supposed to do when it all feels this heavy? Between the seemingly never-ending global pandemic, the contentious U.S. election, and all of the tiny little things that add up to a very heavy weight on your shoulders, it can feel incredibly overwhelming. Today we're going to go over the right questions to ask yourself in order to create the best possible strategy for YOU.Things are hard, but never forget: I'm rooting for you.--Have you checked out the Handpicked by Rach collection? This week I wanted to share the audiobook version of the sixth chapter from my new book, Didn't See That Coming: Putting Life Back Together When Your World Falls Apart. In this chapter I talk about courage. Sometimes, the things we're afraid of are really a reminder of something that went wrong in the past. You have to make the decision that pushing past your fears is a necessity, not just something that you might do eventually. Enjoy this gift of chapter six and remember: I'm rooting for you!
