This week I discuss Ann-Margret as emblematic of the changes in type that were taking place in the early 1960s. Welcome to episode 26 of Inside The Lines: The Catskills! On this episode, I am joined by Russ L and we talk about a former ski center that was on its way to being the best ski center in the north east: Bearpen Mountain. Bearpen Mountain once held a ski center called the Princeton Ski Bowl, which was built and running in the late 1950's. Bearpen, due to its unique shape, attracted more snow than other Catskill peaks and most other mountains in the north east. It was perfect for skiing. In this episode, we talk about the start of the Princeton Ski Bowl and why it was chosen by the owner, Dr. Ben Lane. This is a great intro to the amazing story that Russ tells on his webpage and it was so long, that I had to cut it into two parts! Focus on The Assay to Improve Overall IVD System DevelopmentAssay performance should be the priority when developing complex cartridge and instrument systems. The Assay is King! As assays are becoming more complex, taking an “inside-out” approach to IVD System Development is optimal for project success.This approach starts with first defining what the assay needs to do, then figuring out how to automate that process on the cartridge, and only then focusing on what a commercially viable instrument will look like. This leads to reduced costs, shortened timelines, and superior diagnostic performance. Put another way, the assay team should be the customer when architecting and developing IVD Systems—the chemistry should drive the project.How to implement the inside-out approachDefine the Assay FirstThe assay is where the magic happens and is where the intellectual property lies. That IP is what drives future acquisitions by global diagnostic players. For example, Roche recently acquired GenMark along with the ePlex system that Key Tech helped develop, to broaden its molecular testing portfolio.At the end of the day, the instrument serves the cartridge and the cartridge serves the assay. Before starting any product design work, you need to understand whether the chemistry is locked down. Is the benchtop assay working? If not, it may make sense to wait until it is, to avoid costly rework later.Our CEO and Co-Founder, Jenny Regan, stresses the importance of solidifying assay performance before instrument design:“Anyone can design a beautiful instrument that doesn't work. It's easy to do because it is a tangible milestone for investors and developers alike. The pressure to do that and make something that looks great before doing the nitty-gritty, difficult work of making a working assay is all too easy to fall into.”Collaborate on Menu Planning EarlyIt is critical to not only understand the lead assay, but also what other assays are being planned so that both cartridge and instrument can be planned accordingly. Early collaboration with the assay team will ensure that fewer changes and post-market updates need to be made. This approach also affects speed of regulatory approval, since most changes that need to be made to the cartridge or instrument, to support a new assay, will require applying for re-approval.For example, consider reagent quantity. If your lead assay requires eight reagents, but a few others need ten onboard, you need to ensure that the planned cartridge design takes that extra requirement into account and leaves space for the additional two.Sample type is another consideration. If the lead assay is going to work with a saliva sample, but a future panel will use a stool or blood sample, you need to ensure the cartridge is designed to be able to handle the future, additional sample types.De-Risk and Validate Assay Automation EarlyBecause a working assay is the end goal, it's critical to get working automated cartridge/instrument prototypes to the assay team early so they can validate that the chemistry is still working, and to design a workflow for their assay. It needs to be easy for the assay team to collect data and interact with these early prototypes.Often, this means developing test fixtures for each critical step in the assay process. When translating an assay from the bench to the cartridge, there could be steps that only work part of the time on the bench. These must be confirmed to work 100% of the time on the cartridge. Or there could be steps that always work on the bench, but are extremely complex to automate, requiring validation to see if that specific step is even practical in an automated instrument.At Key Tech, we have a proprietary software platform for hardware interaction, called KeySharp, that allows the assay team to control a prototype instrument while seeing a graphical engineering view of what the cartridge and instrument are doing. With this tool, they have a way to interact with the prototype, adjusting and testing variables, and designing a workflow for their assay.Constant Collaboration for Real-Time FeedbackThe “assay is king” approach requires an ongoing conversation between the engineering team and the assay team, and a consistent feedback loop.The traditional outside-in approach often creates unnecessary constraints on the assay team by forcing them to operate within an engineered product that was developed without them in mind. If the assay team has tweaked the bench assay to generate a better or quicker result, the design may not be able to support it.Pivots and updates in the bench assay are commonplace, so constant communication is the only way to anticipate and adapt the development effort in a timely way.Parallel Development of Cartridge and InstrumentOftentimes, three teams are working on a project – assay team, cartridge team, and instrument team – which makes efficient development exponentially more difficult. Especially if coordinating across multiple organizations.On the other hand, parallel cartridge and instrument development by a single interdisciplinary team eliminates conflicting priorities and incentives between vendors. This integrated approach helps your IVD system get to market faster, as both instrument and cartridge are developed under one roof toward a common goal. Ultimately serving the assay, designing the cartridge and instrument in parallel reduces your overall development process time and cost.Not all design and development firms offer both IVD cartridge and instrument design expertise to support a successful assay. Ben Lane, our Director of Engineering, shares,“Our claim of integration is truly the case. We have cartridge designers and instrument designers who are literally sitting next to each other and talking to each other all day long.”In SummaryAssays are becoming more and more complex, and ensuring that platform development actually serves the assay is critical. Placing your assay and engineering teams in constant collaboration and tackling de-risking, menu planning, and validation early in the process will lead to project success.

For the first year over half of all new cars sold are pure electric. The share of plug-in hybrids (PHEV) was 20.4 %. In other words, 3 of 4 car sold in 2020 has a charging socket." writes Jan for eMobilityNorway.com: "The four most sold car model was electric. Audi e-tron with 9 227 car (6.5 % market share). Tesla Model 3 with 7 770 cars (5.5 % market share). Volkswagen ID.3 with 7 754 cars (5.5 % market share). Nissan Leaf with 5 221 cars (3.7 % market share). For December the numbers were even better. 66.7 % market share for BEVs and 20.4 % for PHEVs. This gives a market share of 87.1 % for cars with a charging socket." Autoblog on why Norway is not a typical market: "Seeking to become the first nation to end the sale of petrol and diesel cars by 2025, oil-producing Norway exempts fully electric vehicles from taxes imposed on those relying on fossil fuels. The policy has turned the country's car market into a laboratory for automakers seeking a path to a future without internal combustion engines, vaulting new brands and models to the top of bestseller lists in recent years. While the sale of BEVs had broken the 50%-mark in individual months, 2020 was the first time that fully electric cars outsold the combined volume of models containing internal combustion engines for a year as a whole." BAIC BJEV-one of the first automakers in China to carry out large-scale investment and operation in the NEV battery swapping industry-had set up 225 stations in 19 cities in China and had 22,000 battery-swapping vehicles on the road as of November." according to China Daily: "Statistics show that the construction cost of a battery-swap station is around 3-10 million yuan ($460,000-$1.53 million). Calculated on the basis that each station will have a reserve of 20 batteries and each battery is valued 100,000 yuan, the cost of batteries for each station will reach 2 million yuan. And that's not counting land, labor and electricity costs. That drove mortgage rates down to lows that had never been seen before, under 3.3% for a 30-year fixed rate, and that led to a refinance boom that just exploded.He goes on to explain the consequences that this had on the market, along with the biggest pain points that it created. NISSAN AIMS TO SELL 1 MILLION EVS ANNUALLY BY 2022 According to Business Insider: “Nissan Motor Co on Friday said it planned to sell 1 million electric vehicles annually by 2022, including all-battery EVs and cars powered by its "e-Power" petrol hybrid technology. During that period, the Japanese automaker said it would develop eight new all-battery EVs, including four models for China, where it is planning to expand. Its luxury Infiniti brand would begin carrying new electric models from 2021, it added. Nissan and its automaking partners, France's Renault SA and Mitsubishi Motors Corp , together plan to launch 12 electric models as part of their strategy to achieve annual vehicle sales totaling 14 million units by 2022, compared with 10.6 million units in 2017.” AND THEY WANT TO LIGHT UP OLD BATTERIES And sticking with Nissan, they are still thinking about how to use old LEAF batteries. You may remember many of the early cars were on battery leases so if they’re back with Nissan now, what to do with them? There’s always home storage. But what about using them for light? Slashgear point out: They’ll be installing new street lighting in Namie, a Japanese town heavily affected by both the 2011 Tohoku earthquake and the Fukushima Daiichi nuclear disaster. By using solar panels and LEDs, the custom lamps will be completely independent from the traditional main power grid, and require no electric cables or outlets” “The design resembles a stylized shepherd’s crook, with the solar panel on the upper section and a sheet of LED lights underneath. In the base, battery packs repurposed from old electric cars like the Nissan Leaf store the electricity generated from the sun during the day, and then release it to power the lights at night. The lamps stand just over nine feet high.” NEW ALL ELECTRIC BENTLEY HAS MISSION E HELP Bentley are moving on with their plan for an all-electric luxury 4-door coupe. And they’ll be using some of the investment already poured into Porsche by the VW group, for their Mission E car. Evo say: “Bentley has already confirmed that it will offer electrified versions of its entire current model range by 2025. The move to develop a new all-electric flagship in addition to this comes as the company seeks to reposition itself as a luxury and technology brand PEER TO PEER EV CHARGING Coming via Fleet News: “Zap-Map has launched a peer-to-peer EV charging network, allowing charge point owners to share electricity with other registered users. Almost 50% of EV drivers told Zap-Map that they were willing to share their home charging with other users, resulting in around 60,000 chargers that could be added to the network. The initiative, called Zap-Home, also provides owners with the opportunity to earn money by sharing their charger. Registered users can set a cost per charge, payable via PayPal.Me. Zap-Map’s chief technology officer Dr Ben Lane said: “Of our 60,000 monthly userbase, we know that around half are willing to share their home point with other Zap-Map users. "Businesses are particularly proactive with 5% of workplace charging points already.
