Abstract: During perceptually guided decisions, correlates of choice are found as upstream as in the primary sensory areas. However, how well these choice signals align with early sensory representations, a prerequisite for their interpretation as feedforward substrates of perception, remains an open question. We designed a two alternative forced choice task (2AFC) in which mice compared stimulation frequencies applied to two adjacent vibrissae. The optogenetic silencing of individual columns in the primary somatosensory cortex (wS1) resulted in predicted shifts of psychometric functions, demonstrating that perception depends on focal, early sensory representations. Functional imaging of layer II/III single neurons revealed sensory, choice and engagement coding. From trial to trial, these three varied substantially, but independently from one another. Thus, coding of sensory and non-sensory variables co-exist in orthogonal subspace of the population activity, suggesting that perceptual variability does not originate from wS1 but rather from state or choice fluctuations in downstream areas. Abstract: The axon initial segment (AIS) constitutes not only the site of action potential initiation, but also a hub for activity-dependent modulation of output generation. Recent studies shedding light on AIS function used predominantly post hoc approaches since no robust murine in vivo live reporters exist. Here, we introduce a reporter line in which the AIS is intrinsically labeled by an ankyrin-G-GFP fusion protein activated by Cre recombinase, tagging the native Ank3 gene. Using confocal, superresolution, and two-photon microscopy as well as whole-cell patch-clamp recordings in vitro, ex vivo, and in vivo, we confirm that the subcellular scaffold of the AIS and electrophysiological parameters of labelled cells remain unchanged. We further uncover rapid AIS remodeling following increased network activity in this model system, as well as highly reproducible in vivo labeling of AIS over weeks. This novel reporter line allows longitudinal studies of AIS modulation and plasticity in vivo in real time and thus provides a unique approach to study subcellular plasticity in a broad range of applications. A evoluat pentru Rapid, unde a marcat alte 11 goluri, totalul de 28 aducându-i titlul de golgheter al Diviziei A. În același sezon a câștigat titlul de campion al României. Evoluțiile bune i-au adus o ofertă din Germania de la Stuttgart, echipă la care a evoluat timp de cinci sezoane. A plecat liber la Bursaspor, în Turcia, unde a petrecut doar șase luni și a semnat o înțelegere cu gruparea Wolverhampton Wanderers. A mai trecut pe la Dinamo, Poli Timișoara, iar finalul carierei de fotbalist l-a prins la Sănătatea Cluj.

În jurul lui roiesc alte personaje puternic conturate, un malefic bătrîn care umblă însoțit de un măgar înhămat la un cadru metalic, Șocaricii, oamenii fără casă, barăcanii, fiii muncitorilor moldoveni care au ridicat blocurile din cartier dar au rămas să locuiască într-un fel de barăci, prietenii lui Ainur, Doagă, Ciprian Ecosal și Bajdavela. Este prezent în antologia „European First Novel” (Budapesta 2017). În 2017 i-a apărut cel de-al doilea roman, „Miere” (Polirom), iar 2018 cel de-al treilea, „Porcii” (Polirom), roman nominalizat pe lista scurtă a Premiului Uniunii Europene pentru Literatură 2019. “Storytelling? I’m already reading books to my child – isn’t that enough?” Your child DOES get a lot out of reading books (which is why we’ve done a several episodes on that already, including Did you already miss the boat on teaching your toddler how to read? But it turns out that storytelling benefits our relationship with our child in ways that reading books really can’t, because you’re looking at the book rather than at your child. If you ask your child what kind of story they’d like you to tell, you also get incredible insight into both their interests and concerns – I can attest to this, as I’ve been singing story-songs about poop and various kinds of baby animals who can’t find their mamas on and off for several weeks now (we had an incident a few months back where she couldn’t find me in a store). We’ve done a couple of episodes on reading by now; episode 3 (which seems so long ago!) asked whether you might have missed the boat on teaching your toddler to read (https://yourparentingmojo.com/003-your-toddler-isnt-reading-yet-neither-is-mine/) .  Of course, we know that you’ve only missed the boat on that if you think that sitting your child in front of a video so they can recite the words they see without really understanding them counts as “reading.” Much more recently in episode 48 we talked with Dr. Laura Froyen about the benefits of shared reading (https://yourparentingmojo.com/reading/) with your child and how to do that according to best practices from the research literature. Those of you who subscribe to my newsletter will recall that I’ve been working on an episode on storytelling for months now.  Part of the reason it’s taking so long is that books on storytelling technique say to use original stories wherever possible because the language in them is so much richer, but if you’ve ever read something like an original fairytale you know they can be pretty gory, and even the most harmless ones actually contain some pretty adult themes if you read between the lines. So I wanted to know: what do children really learn from stories?  How do they figure out that we want them to learn morals from stories but not that animal characters walk on two legs and wear clothes?  How do they generalize that knowledge to the real world?  And are there specific types of books that promote learning?
